A novel water-compatible molecularly imprinted solid-phase extraction (MISPE) combined with zwitterionic hydrophilic interaction liquid chromatography (ZIC-HILIC) method for selective extraction and determination of sitagliptin in rat serum and urine was developed and validated. The effects of progenic solvents, pH, cross linker and amount of monomer were studied to optimize the efficiency and selectivity. The adsorption kinetics and isotherms were measured. The molecularly imprinted polymer (MIP) showed good specific adsorption capacity with an optimum of 180 mg/g at pH 7.5 and selective extraction of sitagliptin from rat plasma and urine. The recovery of sitagliptin from rat urine and plasma was >98%. The limits of detection (LOD) and quantification (LOQ) were 0.03 and 0.10 μg/L respectively. The proposed method overcomes the matrix effects of phospholipids generally encountered while preparation of plasma samples by precipitation of proteins.  相似文献

pH‐zone‐refining centrifugal partition chromatography was successively applied in the large‐scale separation of close Rf antipsychotic indole alkaloids directly from CHCl3 fraction of Rauwolfia tetraphylla leaves. Two experiments with increasing mass from 500 mg to 3 g of crude alkaloid extracts ( 1 C) of R. tetraphylla were carried out in normal‐displacement mode using a two‐phase solvent system composed of methyl tert‐butyl ether/ACN/water (4:1:5, v/v/v) where HCl (12 mM) was added to the lower aqueous stationary phase as a retainer and triethylamine (5 mM) to the organic mobile phase as an eluter. The two centrifugal partition chromatography separations afforded a total of 162.6 mg of 10‐methoxytetrahydroalstonine ( 1 ) and 296.5 mg of isoreserpiline ( 2 ) in 97% and 95.5% purity, respectively, along with a 400.9 mg mixture of α‐yohimbine and reserpiline ( 3 and 4 ). Further, this mixture was resolved over medium pressure LC using TLC grade silica gel H (average particle size 10 μm), which afforded 160.4 mg of α‐yohimbine ( 3) and 150.2 mg of reserpiline ( 4) in >95% purities. The purity of the isolated antipsychotic alkaloids was analyzed by high‐performance LC and their structures were characterized on the basis of their 1D, 2D NMR and electrospray ionization‐mass spectroscopic data.  相似文献

Yttrium oxide (Y2O3) doped with Dy3+ & Eu3+ nanoparticle has been synthesized by solution combustion method. The formation of the compounds has been checked by X-ray diffraction method. The crystallite/particle size has been measured using Scherrer formula as well as by transmission electron microscopy which show that the size of the particles are in the nanorange. The frequency and temperature dependent variation of impedance Z*, dielectric constant (ε′), dielectric loss (ε″) and AC conductivity (σ) of Y2O3: Dy3+ & Eu3+ nanoparticles were also measured. The real and imaginary part of complex impedance makes semicircle in the complex plane. The center of semicircle arc is found to be shifted toward higher value of real part of impedance with increasing temperature. This indicates that the conductivity of the material increases with the increase in temperature. Cole–Cole plots demonstrate that the dielectric relaxation process occurs in the material. The AC conductivity (σ AC) increases with the increase in temperature within the frequency range of 103–107 Hz confirming the hopping of the electrons in the conduction process. The value of impedance decreases sharply with increasing frequency and attains minimum value after 105 Hz at all temperatures.  相似文献

3-Formylsalicylic acid (Hfsal), covalently bound to chloromethylated polystyrene (PS) and cross-linked with 5% divinylbenzene reacts with d,l-alanine and l-isoleucine to give the Schiff-base tridentate ligands PS-H(2)fsal-d,l-Ala and PS-H(2)fsal-l-Ile, respectively. These anchored ligands upon reaction with VOSO(4) and Cu(CH(3)COO)(2).H(2)O form the complexes PS-[VO(fsal-d,l-Ala)(H(2)O)], PS-[Cu(fsal-d,l-Ala)(H(2)O)], PS-[VO(fsal-l-Ile)(H(2)O)] and PS-[Cu(fsal-l-Ile)(H(2)O)]. The structures of these immobilized complexes have been established on the basis of scanning electron micrographs, spectroscopic (infrared, electronic and EPR), thermogravimetric and elemental analysis studies. The oxidation of p-chlorotoluene and cyclohexene has been investigated using these complexes as the catalysts in the presence of H(2)O(2) as the oxidant. Reaction conditions have been optimised by considering the concentration of the oxidant, the amount of catalyst used and the temperature of the reaction mixture. Under the optimised conditions, p-chlorotoluene gave a maximum of 14% conversion using PS-[VO(fsal-d,l-Ala)(H(2)O)] as the catalyst, with the main products having a selectivity order of: p-chlorobenzaldehyde > p-chlorobenzylalcohol > p-chlorobenzoic acid > 2-methyl-5-chlorophenol > 3-methyl-6-chlorophenol. The oxidation of cyclohexene with PS-[VO(fsal-d,l-Ala)(H(2)O)] proceeds with 79% conversion, which is followed by PS-[VO(fsal-l-Ile)(H(2)O)] with 77% conversion, and the oxidation of cyclohexene by Cu-based catalysts occurs with considerably lower conversions (29-32%). The selectivity of the products follows the order: 2-cyclohexene-1-ol > cyclohexene oxide > cyclohexane-1,2-diol > 2-cyclohexene-1-one. Recycling studies indicate that these catalysts can be reused at least three times without any significant loss in their catalytic potential. However, EPR studies indicate that while the polymer supported V(iv)O-complexes do not change after being used, the EPR spectra of the Cu-complexes show significant changes. The corresponding non-polymer bound complexes [VO(fsal-d,l-Ala)(H(2)O)], [Cu(fsal-d,l-Ala)(H(2)O)], [VO(fsal-l-Ile)(H(2)O)] and [Cu(fsal-l-Ile)(H(2)O)] have also been prepared in order to compare their spectral properties and catalytic activities. The non-polymer bound complexes exhibit lower conversion, along with lower turn-over frequency as compared to their polymer-bound analogues. Several EPR, (51)V NMR and UV-vis studies have been undertaken to detect the intermediate species, and outlines for the mechanisms of the catalytic reactions are proposed.  相似文献

The Schiff base (H2fsal-ohyba) derived from 3-formylsalicylic acid and o-hydroxybenzylamine has been covalently bonded to chloromethylated polystyrene cross-linked with 5% divinylbenzene (abbreviated as PS-H(2)fsal-ohyba, I). Treatment of [VO(acac)2] with PS-H2fsal-ohyba in dimethylformamide (DMF) gave the oxovanadium(iv) complex PS-[VO(fsal-ohyba).DMF] (1). Complex 1 can be oxidized into the dioxovanadium(v) species, PS-K[VO2(fsal-ohyba)] (2) on aerial oxidation in the presence of KOH or into the oxoperoxo species, PS-K[VO(O2)(fsal-ohyba)] (3) in the presence of H2O2 and KOH in DMF suspension. Similarly, PS-[MoO(2)(fsal-ohyba).DMF] (4) has been isolated by the reaction of [MoO2(acac)2] with PS-H2fsal-ohyba. All these complexes have been characterised by various techniques. These complexes catalyse the oxidation of styrene, ethylbenzene and phenol efficiently. Styrene gives five reaction products namely styrene oxide, benzaldehyde, 1-phenylethane-1,2-diol, benzoic acid and phenylacetaldehyde, while ethylbenzene gives benzaldehyde, phenyl acetic acid, styrene and 1-phenylethane-1,2-diol. The oxidation products of phenol are catechol and p-hydroquinone. These catalysts are also able to catalyse the oxidative bromination of salicylaldehyde to 5-bromosalicylaldehyde with ca. 80% selectively in the presence of aqueous 30% H2O2/KBr, a reaction similar to that exhibited by vanadate-dependent haloperoxidases. Their corresponding neat complexes have also been prepared and their catalytic activities have been compared.  相似文献

Binuclear, mu-bis(oxo)bis{oxovanadium(V)} complexes [(VOL)2(mu-O)2](2 and 7)(where HL are the hydrazones Hacpy-nah I or Hacpy-fah II; acpy = 2-acetylpyridine, nah = nicotinic acid hydrazide and fah = 2-furoic acid hydrazide) were prepared by the reaction of [VO(acac)2] and the ligands in methanol followed by aerial oxidation. The paramagnetic intermediate complexes [VO(acac)(acpy-nah)](1) and [VO(acac)(acpy-fah)](6) have also been isolated. Treatment of [VO(acac)(acpy-nah)] and [VO(acac)(acpy-fah)] with aqueous H2O2 yields the oxoperoxovanadium(V) complexes [VO(O2)(acpy-nah)](3) and [VO(O2)(acpy-fah)](8). In the presence of catechol (H2cat) or benzohydroxamic acid (H2bha), 1 and 6 give the mixed chelate complexes [VO(cat)L](HL =I: 4, HL =II: 9) or [VO(bha)L](HL =I: 5, HL =II: 10). Complexes 4, 5, 9 and 10 slowly convert to the corresponding oxo-mu-oxo species 2 and 7 in DMF solution. Ascorbic acid enhances this conversion under aerobic conditions, possibly through reduction of these complexes with concomitant removal of coordinated catecholate or benzohydroxamate. Acidification of 7 with HCl dissolved in methanol afforded a hydroxo(oxo) complex. The crystal and molecular structure of 2.1.5H2O has been determined, and the structure of 7 re-determined, by single crystal X-ray diffraction. Both of these binuclear complexes contain the uncommon asymmetrical {VO(mu-O)}2 diamond core. The in vitro tests of the antiamoebic activity of ligands I and II and their binuclear complexes 2 and 7 against the protozoan parasite Entamoeba histolytica show that the ligands have no amoebicidal activity while their vanadium complexes 2 and 7 display more effective amoebicidal activity than the most commonly used drug metronidazole (IC50 values are 1.68 and 0.45 microM, respectively vs 1.81 microM for metronidazole). Complexes 2 and 7 catalyse the oxidation of styrene and ethyl benzene effectively. Oxidation of styrene, using H2O2 as an oxidant, gives styrene epoxide, 2-phenylacetaldehyde, benzaldehyde, benzoic acid and 1-phenyl-ethane-1,2-diol, while ethyl benzene yields benzyl alcohol, benzaldehyde and 1-phenyl-ethane-1,2-diol.  相似文献

Reaction between [VO(acac)(2)] and the ONN donor Schiff base Hsal-ambmz (I) (Hsal-ambmz = Schiff base obtained by the condensation of salicylaldehyde and 2-aminomethylbenzimidazole) resulted in the formation of the complexes [V(IV)O(acac)(sal-ambmz)] (1), [V(V)O(2)(acac-ambmz)] (2) (Hacac-ambmz = Schiff base derived from acetylacetone and 2-aminomethylbenzimidazole), and the known complex [V(IV)O(sal-phen)] (3) (H(2)sal-phen = Schiff base derived from salicylaldehyde and o-phenylenediamine). Similarly, [V(IV)O(acac)(sal-aebmz)] (7) has been isolated from the reaction with Hsal-aebmz (II) (Hsal-aebmz derived from salicylaldehyde and 2-aminoethylbenzimidazole). Aerial oxidation of the methanolic solutions/suspensions of 1 and 7 yielded the dioxovanadium(V) complexes [V(V)O(2)(sal-ambmz)] (4) and [V(V)O(2)(sal-aebmz)] (8), respectively. Reaction of VOSO(4) with II gave [{V(IV)O(sal-aebmz)}(2)SO(4)] (9) and [V(IV)O(sal-aebmz)(2)] (10), along with 3 and 8. Under similar reaction conditions, I gave only [{V(IV)O(sal-ambmz)}(2)SO(4)] (5) and 3 as major products. Treatment of 1 and 7 with benzohydroxamic acid (Hbha) yielded the mixed-chelate complexes [V(V)O(bha)(sal-ambmz)] (6) and [V(V)O(bha)(sal-aebmz)] (11). The crystal and molecular structures of 2, 3.1/2DMF, 7.1/4H(2)O, 8, 9.2H(2)O, 10, and 11 have been determined, confirming the ONN binding mode of the ligands. In complex 10, one of the ligands is coordinated through the azomethine nitrogen and phenolate oxygen only, leaving the benzimidazole group free. In the dinuclear complex 9, bridging functions are the phenolate oxygens from both of the ligands and two oxygens of the sulfato group. The unstable oxoperoxovanadium(V) complex [V(V)O(O(2))(sal-aebmz)] (12) has been prepared by treatment of 7 with aqueous H(2)O(2). Acidification of methanolic solutions of 7 and 10 lead to (reversible) protonation of the bemzimidazole, while 8 was converted to an oxo-hydroxo species. Complexes 2, 4, and 8 catalyze the oxidation of methyl phenyl sulfide to methyl phenyl sulfoxide and methyl phenyl sulfone, a reaction mimicking the sulfideperoxidase activity of vanadate-dependent haloperoxidases. These complexes are also catalytically active in the oxidation of styrene to styrene oxide, benzaldehyde, benzoic acid, and 1-phenylethane-1,2-diol.  相似文献

To support the pharmacokinetic and bioavailability study of a once-daily fexofenadine/pseudoephedrine combination, a high-performance liquid chromatography/positive ion electrospray tandem mass spectrometry (HPLC/ESI-MS/MS) method for the simultaneous quantification of fexofenadine and pseudoephedrine was developed and validated with 500 microL human plasma using mosapride as an internal standard (IS). Following solid-phase extraction, the analytes were separated using an isocratic mobile phase on a reversed-phase column and analyzed by MS/MS in the multiple reaction monitoring mode using the respective [M+H]+ ions, m/z 502/466 for fexofenadine, m/z 166/148 for pseuoephedrine and m/z 422/198 for the IS. The method exhibited linear dynamic ranges of 1-500 ng/mL and 2-1000 ng/mL for fexofenadine and pseudoephedrine, respectively, in human plasma. The lower limits of quantification were 1 and 2 ng/mL with a relative standard deviation of less than 10% for fexofenadine and pseudoephedrine, respectively. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. The total chromatographic run time was 2 min and more than 400 human plasma samples could be analyzed in one day by running the system overnight. The method is precise and sensitive enough for its intended purpose.  相似文献

Hybrid organic–inorganic light emitting devices combine the color purity and durability of inorganic light emitting diodes (LEDs) with high efficiency, flexibility and low processing cost of organic LEDs (OLEDs). A significant challenge is to incorporate inorganic nanocrystals inside the OLED structure. In the present work, thin films of CdS were successfully incorporated inside standard OLED structure using vacuum thermal evaporation technique. For the characterization of these films, they were deposited on plain glass plates at room temperature and studied using structural (XRD and TEM), morphological (SEM and AFM) and optical (UV and PL) techniques. The films were found to be composed of nanocrystals of CdS in which the size of the crystals increased with the increase in film thickness. The hybrid organic–inorganic LEDs showed improved luminance and efficiency as compared to the organic LED without CdS layers.  相似文献
