采用紫外-可见吸收、荧光及圆二色(CD)等光谱法为主要分析手段, 对光诱导高铁肌红蛋白(metmyoglobin, metMb)还原及其引起的蛋白结构变化进行了研究. 采用氙灯430 nm的单色光和紫外灯对高铁肌红蛋白溶液进行光照实验, 实验结果显示, 在metMb的Q吸收带545 nm和580 nm附近吸收峰强度显著增强. 通过验证性实验, 证明光照后metMb发生了还原反应. 验证性实验包括: metMb中加入还原剂连二亚硫酸钠实验|光照后样品加入氧化剂铁氰化钾的可逆性实验|光照的同时通入CO实验. 进一步讨论表明, 采用氙灯430 nm的单色光诱导metMb还原的机理为光诱导的分子内电子转移. 此外, 对光还原反应后的样品进行同步荧光和CD光谱分析, 发现蛋白质色氨酸残基微环境极性增加, α-螺旋的含量由 63%下降到57%, 而β-折叠的含量增加了7%. 相似文献

A new alkaloid, 3-dodecyl pyridine containing a terminal cyano group (1), was isolated from the methanol extract of an Indonesia marine sponge Haliclona sp. Its chemical structure was determined by a combination of spectroscopic methods, including 1D and 2D NMR. Multifunctional polymer unimolecular micelles, which are used as templates to fabricate stable gold nanoparticles (GNPs) in one‐step without external reductant, have been designed and prepared. Amphiphilic 21‐arm star‐like block copolymers β‐cyclodextrin‐{poly(lactide)‐poly(2‐(dimethylamino) ethyl methacrylate)‐poly[oligo(2‐ethyl‐2‐oxazoline)methacrylate]}21 [β‐CD‐(PLA‐PDMAEMA‐PEtOxMA)21] and the precursors are synthesized by the combination of ring‐opening polymerization (ROP) and activators regenerated by electron transfer atom transfer radical polymerization (ARGET ATRP). The tertiary amine groups of PDMAEMA block reduce the counterion to zerovalent gold in situ, and these gold atoms combine mutually to form final GNPs. GNPs with relatively small size and narrow size distribution can be obtained in longer DMAEMA block copolymer, larger molar ratio of DMAEMA to HAuCl4 and smaller absolute concentrations of both polymer and HAuCl4. These results showed that the unimolecular micelles can be used as templates for preparing and stabilizing GNPs in situ without any external reducing agents and organic solvents, suggesting that the nanocomposite systems are latent nanocarriers for further biomedical application. © 2015 Wiley Periodicals, Inc. 以人工合成的13X型分子筛为吸附剂,研究了水中Pb2 、Cd2 和Cu2 在分子筛上的吸附行为,讨论了Pb2 、Cd2 和Cu2 共存条件下的竞争吸附,并考察了水中存在的Na 、Mg2 、Ca2 等离子对分子筛吸附去除重金属的影响.实验结果表明,13X分子筛对Pb2 、Cd2 和Cu2 3种离子的吸附符合Langmuir模型,最大吸附量分别为2.47mmol/L、2.0mmol/L和1.78mmol/L.在竞争吸附条件下,13X分子筛对3种重金属离子的选择性依次为pb2 >Cd2 >Cu2 .水中存在的Na 、Mg2 、Ca2 等对分子筛吸附重金属效率有一定的影响,其中Ca2 对Cu2 在分子筛上的去除影响最为显著. 相似文献

The rotationally resolved ultraviolet absorption cross sections for the 2(0)(0)4(1)(0) vibrational band of the A(1)A(2)-X(1)A(1) electronic transition of formaldehyde (HCHO) at an apodized resolution of 0.027 cm(-1) (approximately 0.0003 nm at 352 nm) over the spectral range 28100-28500 cm(-1) (351-356 nm) at 298 and 220 K, using Fourier transform spectroscopy, are first reported here. Accurate rotationally resolved cross sections are important for the development of in situ HCHO laser-induced fluorescence (LIF) instruments and for atmospheric monitoring. Pressure dependence of the cross sections between 75 and 400 Torr at 298 K was explored, and an average pressure broadening coefficient in dry air of 1.8 x 10(-4) cm(-1) Torr(-1) for several isolated lines is reported. Gaseous HCHO was quantitatively introduced into a flow cell by evaporating micron-sized droplets of HCHO solution, using a novel microinjector technique. The condensed-phase concentrations of HCHO were determined by iodometric titrations to an accuracy of <1%. Accuracy of the measured absorption cross sections is estimated to be better than +/-5%. Integrated and differential cross sections over the entire band at low resolution (approximately 1 cm(-1)) obtained with our calibration technique are in excellent agreement with previous measurements. A maximum differential cross section of 5.7 x 10(-19) cm(2) molecule(-1) was observed at high resolution-almost an order of magnitude greater than any previously reported data at low resolution. 相似文献
