A refined version of the pre-column oxidation liquid chromatography with fluorescence detection (ox-LC-FLD) official method AOAC 2005.06 was developed in the UK and validated for the determination of paralytic shellfish poisoning toxins in UK shellfish. Analysis was undertaken here for the comparison of PSP toxicities determined using the LC method for a range of UK bivalve shellfish species against the official European reference method, the PSP mouse bioassay (MBA, AOAC 959.08). Comparative results indicated a good correlation in results for some species (mussels, cockles and clams) but a poor correlation for two species of oysters (Pacific oysters and native oysters), where the LC results in terms of total saxitoxin equivalents were found to be on average more than double the values determined by MBA. With the potential for either LC over-estimation or MBA under-estimation, additional oyster and mussel samples were analysed using MBA and ox-LC-FLD together with further analytical and functional methodologies: a post-column oxidation LC method (LC-ox-FLD), an electrophysiological assay and hydrophilic interaction liquid chromatography with tandem mass spectrometric detection. Results highlighted a good correlation among non-bioassay results, indicating a likely cause of difference was the under-estimation in the MBA, rather than an over-estimation in the LC results.  相似文献

The March 2011 Mega-Tsunami in eastern Japan damaged at different degrees the black pine (Pinus thunbergii) forests along the coast. In order to evaluate the recovery of black pine four years later, tree-ring samples from 9 trees for the period 2002–2014 were analyzed for ring growth and stable isotopes (δ13C, δ15N and δ18O). The results showed that annual tree-ring width decreased approximately 70?% from the year 2011 to 2014 compared to the period previous to the tsunami (2002–2010). The multiple isotopic analyses showed that the reduction in growth was caused by soil salinity that prompted stomatal closure and an abrupt increase of tree-ring δ13C. Sea water deposition in the soil did not affect tree-ring δ18O values. Two years after the tsunami, decreasing tree-ring δ13C values caused by apparently photosynthetic recovery did not translate into radial tree-growth, indicating a possible shift in carbon allocation to foliage and mainly roots as a defense mechanism to sodium toxicity. The dual δ13C-δ18O model explains neither the limited growth nor the subsequent recovery in δ13C. Similarly tree-ring δ15N indicated that there was no difference in nitrogen availability before and after the tsunami, suggesting that nutrients were not a limitation but rather soil salinity.  相似文献

Molar densities have been measured by using an expansion technique at pressures up to 104 MPa and temperatures from 198 to 298 K for the mixture {x CS2 + (1-x) Si(CH3)4}(1) over the whole composition range. Several thennodynamic properties (isothermal compressibility, thermal expansivity, and molar internal energy, enthalpy and entropy increments relative to liquid at saturation) have been directly obtained from the experimental densities. The isothermal compressibility and the thermal expansivity are both smooth functions of pressure, temperature and composition, although the isotherms of the thermal expansivity exhibit a characteristic crossover at high pressures. The intersection of isotherms of thermal expansivity seems to occur at a single point of the (pp) diagram, showing a nearly linear dependence with the composition.  相似文献

We use a dynamic light scattering technique to measure both polarized (VV) and depolarized (VH) spectra of liquid diphenylmethane (DPM) between 288 and 362 K, covering both normal and supercooled liquid ranges. Our results allow extracting information on structural relaxation processes, rotational motions, rotation-translation couplings, and molecular reorientation phenomena in liquid DPM. The VV spectra are modeled according to the microscopic theory of Wang, which assumes that a structural relaxation process dominates the spectrum. We find that the relaxation time of the structural relaxation in DPM follows an Arrhenius behavior. The Rayleigh dip was observed in the VH spectra, which are described using the Andersen-Pecora theory. Our results are discussed in terms of the rotation-translation coupling parameter, which we find independent of temperature over the experimental range. The collective reorientation time also follows an Arrhenius behavior with temperature. Finally, we calculate the hydrodynamic volumes for the reorientation process from geometric molecular models in two hydrodynamic limits: slip and stick boundary conditions. Our results suggest that the DMP molecule reorientates in quasi-slipping conditions in the bulk liquid.  相似文献

The unbound nucleus 18Na, the intermediate nucleus in the two-proton radioactivity of 19Mg, was studied by the measurement of the resonant elastic scattering reaction H(17Ne,p)17Ne performed at 4 A.MeV using a radioactive beam from the SPIRAL Facility. Spectroscopic properties of the low-lying states were obtained in an R-matrix analysis of the excitation function.  相似文献

Temporal variations in N concentration and δ(15)N value of annual tree rings (1 year of time resolution) of two Japanese Black Pine (Pinus thunbergii) and three Japanese Red Pine (Pinus densiflora) trees under current breeding activity of the Great Cormorant (Pharacrocorax carbo) and the Black-tailed Gull (Larus crassirostris), respectively, in central and northeastern Japan were studied. Both species from control sites where no avian input occurs show negative values (δ(15)N = around -4 ‰ to -2 ‰) which are common among higher plants growing under high rainfall regimes. The δ(15)N values of P. densiflora show uniformly positive values several years before and after the breeding event, indicating N translocation that moved the absorbed N of a given growth year to tree rings of the previous year while a clear historical value of soil N dynamics was kept intact in the annual rings of P. thunbergii. Long-term N trends inferred from tree rings must take into account tree species with limited translocation rates that can retain actual N annual acquisition.  相似文献

This paper focuses on the Vehicle Routing Problem with Stochastic Demands (VRPSD) and discusses how Parallel and Distributed Computing Systems can be employed to efficiently solve the VRPSD. Our approach deals with uncertainty in the customer demands by considering safety stocks, i.e. when designing the routes, part of the vehicle capacity is reserved to deal with potential emergency situations caused by unexpected demands. Thus, for a given VRPSD instance, our algorithm considers different levels of safety stocks. For each of these levels, a different scenario is defined. Then, the algorithm solves each scenario by integrating Monte Carlo simulation inside a heuristic-randomization process. This way, expected variable costs due to route failures can be naturally estimated even when customers’ demands follow a non-normal probability distribution. Use of parallelization strategies is then considered to run multiple instances of the algorithm in a concurrent way. The resulting concurrent solutions are then compared and the one with the minimum total costs is selected. Two numerical experiments allow analyzing the algorithm’s performance under different parallelization schemas.  相似文献

Despite the importance of protein–surface interactions in both biology and biotechnology, our understanding of their origins is limited due to a paucity of experimental studies of the thermodynamics behind such interactions. In response, we have characterized the extent to which interaction with a chemically well‐defined macroscopic surface alters the stability of protein L. To do so, we site‐specifically attached a redox‐reporter‐modified protein variant to a hydroxy‐terminated monolayer on a gold surface and then used electrochemistry to monitor its guanidine denaturation and determine its folding free energy. Comparison with the free energy seen in solution indicates that interaction with this surface stabilizes the protein by 6 kJ mol?1, a value that is in good agreement with theoretical estimates of the entropic consequences of surface‐induced excluded volume effects, thus suggesting that chemically specific interactions with this surface (e.g., electrostatic interactions) are limited in magnitude.  相似文献

This paper addresses a multi-period production/inventory problem with two suppliers, where demand sizes and supplier lead time are stochastic and correlated. A discrete time, single item inventory system is considered, where inventory levels are reviewed periodically and managed using a base-stock policy. At the end of each period, a replenishment order is placed, which enters a queue at the buffer stage and is consequently forwarded to the first available supplier. We present a mathematical model of this inventory system and determine optimal safety stock levels for it, in closed form, using matrix analytic techniques and the properties of phase type distributions. To account for the effect of order crossovers, which occur whenever replenishment orders do not arrive in the sequence in which they were placed, the inventory shortfall distribution is analyzed. Finally, a set of numerical experiments with a system with two suppliers is presented, where the proposed model is compared to other existing models.  相似文献
