The microRNA, miR-141, is a promising biomarker for prostate cancer. We implement here a two-step sensing platform for the sensitive detection of miR-141. The first step involves the use of semiconductor CdSe/ZnS quantum dots (QDs) modified by FRET quencher-functionalized nucleic acids, that include the recognition sequence for miR-141 and a telomerase primer sequence for the second step of the analytical platform. Subjecting the probe-modified QDs to miR-141, in the presence of duplex specific nuclease, DSN, leads to the formation of a miR-141/probe duplex and to its DSN-mediated cleavage, while regenerating the miR-141. The DSN-induced cleavage of the quencher units leads to the activation of the fluorescence of the QDs, thus allowing the optical detection of miR-141 with a sensitivity corresponding to 1.0 × 10–12 M. The nucleic acid residues associated with the QDs after cleavage of the probe nucleic acids by DSN act as primers for telomerase. The subsequent telomerase/dNTPs-stimulated elongation of the primer units forms G-quadruplex telomer chains. Incorporation of hemin in the resulting G-quadruplex telomer chains yields horseradish peroxidase-mimicking DNAzyme units, that catalyze the generation of chemiluminescence in the presence of luminol/H2O2. The resulting chemiluminescence intensities provide a readout signal for miR-141, DL = 2.8 × 10–13 M. The first step of the sensing platform is non-selective toward miR-141 and the resulting fluorescence may be considered only as an indicator for the existence of miR-141. The second step in the sensing protocol, involving telomerase, provides a selective chemiluminescence signal for the existence of miR-141. The two-step sensing platform is implemented for the analysis of miR-141 in serum samples from healthy individuals and prostate cancer carriers. Impressive discrimination between healthy individuals and prostate cancer carriers is demonstrated.  相似文献

Toward developing a micro HPLC cartridge, we have recently built a high-pressure electroosmotic pump (EOP). However, we do not recommend people to use this pump to deliver an organic solvent directly, because it often makes the pump rate unstable. We have experimented several approaches to address this issue, but none of them are satisfactory. Here, we develop an innovative approach to address this issue. We first create an abruption (a dead-volume) within a fluid conduit. We then utilize an EOP to withdraw, via a selection valve, a train of eluent solutions having decreasing eluting power into the fluid conduit. When these solutions are further aspirated through the dead-volume, these solutions are partially mixed, smoothening concentration transitions between two adjacent eluent solutions. As these solutions are pushed back, through the dead-volume again, a smooth gradient profile is formed. In this work, we characterize this scheme for gradient formation, and we incorporate this approach with a high-pressure EOP, a nanoliter injection valve, and a capillary column, yielding a micro HPLC system. We then couple this micro HPLC with an electrospray ionization – mass spectrometer for peptide and protein separations and identifications.  相似文献

In this paper,we present a simple method by combining surface wrinkling and template replication to create a series of hierarchical structures on polydimethylsiloxane(PDMS) elastomer.The primary stable lined patterns are formed by duplicating commercialized compact disk and digital versatile disk with PDMS.The secondary microscale patterns are from surface wrinkling,which is elicited by oxygen plasma(OP) treatment of the prestrained PDMS stamp followed with the prestrain release.By systematically varying the OP exposure duration,the prestrain,and the angle(θ) between the primary pattern orientation and the prestrain direction,we obtain highly ordered well-organized composite patterns from different patterning techniques and with different length scales and mechanical stabilities.  相似文献

In the present paper, a comparison of the performance between moving cutting data-rescaled range analysis(MCR/S) and moving cutting data-rescaled variance analysis(MC-V/S) is made. The results clearly indicate that the operating efficiency of the MC-R/S algorithm is higher than that of the MC-V/S algorithm. In our numerical test, the computer time consumed by MC-V/S is approximately 25 times that by MC-R/S for an identical window size in artificial data. Except for the difference in operating efficiency, there are no significant differences in performance between MC-R/S and MC-V/S for the abrupt dynamic change detection. MC-R/S and MC-V/S both display some degree of anti-noise ability. However, it is important to consider the influences of strong noise on the detection results of MC-R/S and MC-V/S in practical application processes.  相似文献

We report the properties of a new series of wide band gap photovoltaic polymers based on the N-alkyl 2-pyridone dithiophene (PDT) unit. These polymers are effective bulk heterojunction solar cell materials when blended with phenyl-C71-butyric acid methyl ester (PC71BM). They achieve power conversion efficiencies (up to 5.33%) high for polymers having such large bandgaps, ca. 2.0 eV (optical) and 2.5 eV (electrochemical). Grazing incidence wide-angle X-ray scattering (GIWAXS) reveals strong correlations between π–π stacking distance and regularity, polymer backbone planarity, optical absorption maximum energy, and photovoltaic efficiency.  相似文献

A new camphor-based 2-phenylimino-2-oxazolidine chiral auxiliary was prepared and it was shown to be a particularly effective chiral auxiliary for asymmetric alkylations affording high yields and diastereoselectivities. The alkylation products were readily cleaved by simple alkaline hydrolysis to give a-alkylated carboxylic acids in good yield and in almost enatiomerically pure form.  相似文献

This work investigates the oxidation of hydrogen near its second explosion limit in a turbulent flow reactor at pressures of 1 to 8 bar, temperatures of 950 K and an equivalence ratio of 0.035. The concentrations of H2, O2 and H2O are measured along the reactor and simulated using several kinetic models from the literature. These experiments demonstrate evident negative pressure dependence from roughly 1 to 4 bar, with further increases in pressure resuming its positive impact on reaction rates. The simulated and measured species concentrations along the reactor generally agree within a factor of 2.Further investigation is then conducted to measure the rate coefficient of reaction H + O2 (+ M) = HO2 (+M) (R2), which is one of the most sensitive reactions in hydrogen's oxidation chemistry at these conditions. This investigation is conducted by using nitric oxide (NO) as a dopant and measuring the resulting, quasi-steady-state concentrations of NO2. The rate coefficients are obtained at 950 – 1010 K. Combined with literature results, an Arrhenius expression is proposed, k2,0N2 = 4.50 × 1020 (T/K)?1.73 [cm6 mole?2 s?1], for the reaction rate at the low-pressure limit over 500 K – 2000 K with N2 as the bath gas. Simulations using the models from the literature with the proposed Arrhenius expression for this reaction then demonstrate improved agreement with the experiments.  相似文献
