The ability of vesicle-coated silica to aid the extraction of organic compounds from water prior to liquid chromatographic analysis is presented for the first time. The method is based on the formation of silica supported cationic multi-lamellar vesicles of gemini surfactants inherently ensuring the presence of hydrophilic and hydrophobic sites for the partitioning of analytes bearing different properties. Method development is illustrated by studying the adsolubilization of UV absorbing chemicals from swimming pool water. Due to the requirement for external energy input (intense shearing) a method based on solid-phase dispersion (SPD) was applied producing better results than off-line solid-phase extraction (SPE). Meticulous investigation of the experimental parameters was conducted in order to elucidate the mechanisms behind the proposed extraction pattern. Analyte recoveries were quantitative under the optimum experimental conditions offering recoveries higher than 96% with RSD values below 5%.  相似文献

Numerous molecular biological experiments performed throughout the world require the detection or quantification of a protein of interest. Western blotting is one of the most popular techniques used for this purpose and offers quantitative information with the aid of specialized software. However, its dependence on the picture that is captured, and the background and the absence of a common protocol prevent the technique from being completely quantitative. To overcome these obstacles, we present a simple and reliable assay that is similar to the regular technique, with the exception of the last stage of band visualization and quantification. We propose that small pieces of the blot that include the protein of interest can be cut and dipped in a small volume of 3,3',5,5'-tetramethylbenzidine solution, giving a colorimetric signal with linear dependence on the quantity of the protein. The reaction is stopped with H2SO4, and the signal is measured in a plate reader. This modification shows high linearity without additional costs and can be applied for both purified proteins and proteins found in a lysate. The results obtained with our proposed technique were compared with those obtained by the conventional method and proved to be more reliable.  相似文献

At the beginning of the twentieth century, historians associated the alchemy of the third-century alchemist Zosimus of Panopolis with Platonism and Aristotelianism, explicating his theory of alchemical transmutation under the intellectual umbrella of these philosophical traditions. More recently, scholars of alchemy such as Christina Viano and William Newman have suggested a connection between Zosimean alchemy and Stoicism. Through a close reading of texts in Zosimus’s corpus, this paper posits a Stoic interpretation of several aspects of Zosimean alchemy, focusing on the concepts of pneuma and tension. For Zosimus, I argue, pneuma played a vital role in colouring metals, while tension conferred stability and cohesion upon metallic compounds. This interpretation suggests that Zosimus applied Stoic concepts to describe the alchemical process of tincturing metals.  相似文献

A number of pyrrolo[2,3-c]pyridines, pyrrolo[3,2-d]pyrimidines and pyrazolo[4,3-d]pyrimidines were designed and synthesized as antiproliferative agents. The target compounds possessed selected substituents in analogous positions on the central scaffold that allowed the extraction of interesting SARs. The cytotoxic activity of the new derivatives was evaluated against prostatic (PC-3) and colon (HCT116) cell lines, and the most potent analogues showed IC50 values in the nM to low µM range, while they were found to be non-toxic against normal human fibroblasts (WI-38). Flow cytometric analysis of DNA content revealed that the most promising derivative 14b caused a statistically significant accumulation of PC-3 cells at G2/M phase and induced apoptosis in PC-3 cells.  相似文献

Most often, in bioseparations involving charged macromolecules, the chromatographic systems have low Reynolds and high Peclet numbers. For such systems, an expression is developed and presented in this work for evaluating the throughput in polymeric monoliths where ion-exchange adsorption occurs, as a function of (i) the pressure drop along the length of the monolith, (ii) the functional form and width of the throughpore-size distribution of the monolith, and (iii) the magnitude of the zeta potential on the surface of the throughpores of the monolith. Gaussian and log-normal throughpore-size distributions whose mean throughpore-size and standard deviation values are based on experimentally measured throughpore-size distribution data by mercury porosimetry employed on polymeric monoliths are used in this work, and their effect on the throughput relative to that obtained from a polymeric monolith having a uniform throughpore-size distribution is studied for different values of the ratio of the standard deviation to the mean throughpore-size. The results indicate that relatively modest increases in the throughput, when compared with the throughput that could be achieved in a polymeric monolith having a uniform throughpore-size distribution, could be obtained in polymeric monoliths having disperse throughpore-size distributions, and the magnitude of the increase becomes larger when the disperse distribution is skewed to larger throughpore sizes. Furthermore, the results of this work indicate that, under certain conditions, relatively modest increases in the throughput of a charged analyte could also be achieved by altering the value of the zeta potential on the surface of the throughpores of the monolith. Due to the difficulties inherent in controlling the functional form and width of the throughpore-size distribution during the synthesis of polymeric monoliths, it would appear to be more practical to increase the value of the throughput of a charged analyte by altering the value of the zeta potential through prudent selection of the ion-exchange surface functional groups and fine-tuned with the pH of the mobile phase. Thus, for ion-exchange chromatography systems, the zeta potential could be considered an important parameter for column designers and operators to manipulate, since its alteration could increase the through-put of a charged analyte in polymeric monoliths or in columns packed with charged particles.  相似文献

The β3 subunit of nicotinic acetylcholine receptors (nAChRs) participates in heteropentameric assemblies with some α and other β neuronal subunits forming a plethora of various subtypes, differing in their electrophysiological and pharmacological properties. While β3 has for several years been considered an accessory subunit without direct participation in the formation of functional binding sites, recent electrophysiology data have disputed this notion and indicated the presence of a functional (+) side on the extracellular domain (ECD) of β3. In this study, we present the 2.4 Å resolution crystal structure of the monomeric β3 ECD, which revealed rather distinctive loop C features as compared to those of α nAChR subunits, leading to intramolecular stereochemical hindrance of the binding site cavity. Vigorous molecular dynamics simulations in the context of full length pentameric β3-containing nAChRs, while not excluding the possibility of a β3 (+) binding site, demonstrate that this site cannot efficiently accommodate the agonist nicotine. From the structural perspective, our results endorse the accessory rather than functional role of the β3 nAChR subunit, in accordance with earlier functional studies on β3-containing nAChRs.  相似文献

The present work aims to establish a generic reforming reaction scheme to evaluate the performance of catalytic reforming systems with the aid of a one‐dimensional heterogeneous dynamic model. The novelty of the numerical model stems from the direct inclusion of interphase (fluid‐to‐particle surface), intraparticle (within particle), and intrareactor heat and mass transport resistances under transient conditions. The developed model accounts for the multicomponent gas mixture physicochemical properties and correlations for calculating mass and heat transfer coefficients. Effective macroscopic properties within the particle are calculated by incorporating diffusivities and conductivities of the porous network characteristics accounting for Knudsen and molecular transport as well as tortuosity and porosity of the overall porous path. The industrial case of a steam‐methane reforming multitubular reactor was studied as the most representative case of the generic reaction scheme, with all mass/energy resistances present under severe pressure and temperature conditions. It was shown that there are notable diffusional limitations within the particle, whereas there are also temperature and partial pressure gradients due to the heat and mass transport resistances in the particle film layer. It is further demonstrated that the proposed model can be utilized as a versatile design tool for catalytic reactor development and optimization.  相似文献
