Changes in MRI signal intensity during hypercapnic challenge under conscious and anesthetized conditions 总被引:5,自引:0,他引:5
Most functional magnetic resonance imaging (fMRI) studies in animals are conducted under anesthesia to minimize motion artifacts. However, methods and techniques have been developed recently for imaging fully conscious rats. Functional MRI studies on conscious animals report enhanced BOLD signal changes as compared to the anesthetized condition. In this study, rats were exposed to different concentrations of carbon dioxide (CO(2)) while conscious and anesthetized to test whether cerebrovascular reactivity may be contributing to these enhanced BOLD signal changes. Hypercapnia produced significantly greater increases in MRI signal intensity in fully conscious animals (6.7-13.3% changes) as when anesthetized with 1% isoflurane (3.2-4.9% changes). In addition, the response to hypercapnia was more immediate in the conscious condition (< 30s) with signal risetimes twice as fast as in the anesthetized state (60s). Both cortical and subcortical brain regions showed a robust, dose- dependent increase in MRI signal intensity with hypercapnic challenge while the animals were conscious but little or no change when anesthetized. Baseline variations in MRI signal were higher while animals were conscious but this was off set by greater signal intensity changes leading to a greater contrast-to-noise ratio, 13.1 in conscious animals, as compared to 8.0 in the anesthetized condition. In summary, cerebral vasculature appears to be more sensitive to hypercapnic challenge in the conscious condition resulting in enhanced T2* MRI signal intensity and the potential for better BOLD signal changes during functional imaging. 相似文献

The role of hemoglobin (Hb) in transmitting the vasodilatory property of NO throughout the vascular system is of much current interest. NO exchange between Hb and low-molecular-weight nitrosothiols such as S-nitrosoglutathione (GSNO) has been speculated and reported in vitro. Previously, we reported that NO delivery from GSNO to Cysbeta93 of human oxyHb is prevented in the presence of the Cu chelators, neocuproine, and DTPA.(1) In the present work, 5 mM solutions of commercial human Hb were found by ICP-MS to contain approximately 20 microM Cu and Zn, suggesting the presence of Cu,Zn-superoxide dismutase (CuZnSOD), which was confirmed by Western blotting. SOD activity measurements were consistent with the presence of approximately 20 microM CuZnSOD monomer in 5 mM Hb solutions, which is the physiological concentrations of these proteins in the red blood cell. Incubation of 3.75 mM oxyHb (15 mM heme; 7.5 mM Cysbeta93) with 3.75 or 7.5 mM GSNO gave rise to 50% or 100% S-nitrosation, respectively, of Cysbeta93 as monitored by FTIR nu(SH) absorption, whereas excess GSNO over Cysbeta93 converted oxyHb to metHb due to the reaction, oxyHb + NO<==>metHb + NO(3)(-). Removal of CuZnSOD by anion-exchange chromatography yielded an oxyHb sample that was unreactive toward GSNO, and replacement with bovine CuZnSOD restored reactivity. Addition of 1 microM GSNO (Cysbeta93/GSNO = 1) to solutions diluted 10(4)-fold from physiological concentrations of oxyHb and CuZnSOD resulted largely in metHb formation. Thus, this work reports the following key findings: CuZnSOD is an efficient catalyst of NO transfer between GSNO and Cysbeta93 of oxyHb; metHb is not detected in oxyHb/GSNO incubates containing close to the physiological concentration (5 mM) of Hb and CuZnSOD when the Cysbeta93/GSNO molar ratio is 0.5 to 1.0, but metHb is detected when the total Hb concentration is low micromolar. These results suggest that erythrocyte CuZnSOD may play a critical role in preserving the biological activity of NO by targeting it from GSNO to Cysbeta93 of oxyHb rather than to its oxyheme. 相似文献

A new design method for complex-valued two-channel finite impulse response (FIR) filter banks with both orthogonality and symmetry properties is developed. Based on a novel linear matrix inequality (LMI) characterization of trigonometric curves, the optimal design of perfect-reconstruction filter banks is cast into a semidefinite programming (SDP) problem. The dimension of the resulting SDP problem is further reduced by exploiting convex duality. To explore mobile cell phone utility use, 99 people completed a mobile phone usage questionnaire. Fourier transform infrared microscopy has been used to investigate in situ dehydroxylation of goethite to form hematite. The characterisation was based on the behaviour of hydroxyl units, which were observed in the hydroxyl stretching and hydroxyl deformation and water bending regions, and the Fe-O vibrations of the newly formed hematite during the thermal dehydroxylation process. Two hydroxyl stretching modes (v1 and v2), and three bending (V(bending-1, 2, 3)) and two deformation (V(deformation-1, 2)) modes were observed for goethite. The characteristic vibration at 916 cm(-1) was observed together with the residuals of the v1 and v2 bands in hematite spectrum. The structural transformation between goethite and hematite through thermal dehydroxylation was interpreted in order to provide criteria that can be used for the characterisation of thermally activated bauxite and their conversion to activated alumina phases. 相似文献

Cooperative spatial multiplexing (CSM) system has played an important role in wireless networks by offering a substantial improvement in multiplexing gain compared with its cooperative diversity counterpart. However, there is a limited number of research works that consider the performance of CSM systems. As such, in this paper, we have derived exact performance of CSM with amplify‐and‐forward and decode‐and‐forward relays in terms of outage capacity and ergodic capacity. We have shown that CSM systems yield a unity diversity order regardless of the number of antennas at the destination and the number of relays in the networks, which is the direct result of diversity and multiplexing gain trade‐off. This study investigated jurors' reactions to judges' nonverbal communication in mock trials. Participants in 80 mock juries viewed taped segments of direct and cross examination that varied the judges' level of nonverbal involvement. Comments about the judge were solicited in predeliberation questionnaires and unsolicited comments obtained from dialogue that occurred during mock jury deliberations. The investigation found that jurors are aware of nonverbal cues from judges, particularly negative behaviors. Although the high- and low-involved judges solicited roughly the same number of comments, more negative comments were made about the judge displaying low than high nonverbal involvement. The results also revealed that jurors made more negative comments than positive ones. Overall, jurors are most aware of a judge's lack of involvement and apparent bias. Discussion focuses on ways to minimize the influence of judge nonverbal communication on jurors. 相似文献

Amyloid precursor protein (APP) is the precursor protein to amyloid β (Aβ), the main constituent of senile plaques in Alzheimer's disease (AD). Endogenous Aβ peptides reflect the APP processing, and greater knowledge of different APP degradation pathways is important to understand the mechanism underlying AD pathology. When one analyzes longer Aβ peptides by low‐energy collision‐induced dissociation tandem mass spectrometry (MS/MS), mainly long b‐fragments are observed, limiting the possibility to determine variations such as amino acid variants or post‐translational modifications (PTMs) within the N‐terminal half of the peptide. However, by using electron capture dissociation (ECD), we obtained a more comprehensive sequence coverage for several APP/Aβ peptide species, thus enabling a deeper characterization of possible variants and PTMs. Abnormal APP/Aβ processing has also been described in the lysosomal storage disease Niemann–Pick type C and the major large animal used for studying this disease is cat. By ECD MS/MS, a substitution of Asp7 → Glu in cat Aβ was identified. Further, sialylated core 1 like O‐glycans at Tyr10, recently discovered in human Aβ (a previously unknown glycosylation type), were identified also in cat cerebrospinal fluid (CSF). It is therefore likely that this unusual type of glycosylation is common for (at least) species belonging to the magnorder Boreoeutheria.
