Vector and Cell Line Engineering Technologies Toward Recombinant Protein Expression in Mammalian Cell Lines

The rapid growth of global biopharmaceutical market in the recent years has been a good indication of its significance in biotechnology industry. During a long period of time in recombinant protein production from 1980s, optimizations in both upstream and downstream processes were launched. In this regard, one of the most promising strategies is expression vector engineering technology based on incorporation of DNA opening elements found in the chromatin border regions of vectors as well as targeting gene integration. Along with these approaches, cell line engineering has revealed convenient outcomes in isolating high-producing clones. According to the fact that more than 50% of the approved therapeutic proteins is being manufactured in mammalian cell lines, in this review, we focus on several approaches and developments in vector and cell line engineering technologies in mammalian cell culture.     

Available and Waiting Times for Cognitive Radios

Cognitive radios (CRs) have been recently proposed for the problem of spectrum scarcity. The principle of CRs?? operation is based on the opportunistic access to the frequency spectrum mainly dedicated to primary users (PUs). The statistical time pattern of PUs?? channel usage and arrival can affect the usability of specific frequency bands for CRs. In this note, the effect of the arrival rate and channel holding time of PUs on the available times for CRs is analyzed. To this end, first, based on Poissonian arrivals, the available time for CRs is calculated. Then, assuming a gamma distribution for the inter-arrival times and a uniform distribution of channel holding time of PU in these intervals, the probability density function and moments of the available time for CRs are derived. Next, the effect of PUs statistical parameters on the average number of packets and the average symbol rate that a CR can transmit is analyzed. Also, taking that CR needs at least T seconds, the average waiting time is calculated.     

Old Reagent,New Results: Aromatization of Hantzsch 1,4-Dihydropyridines with Supported Bismuth Nitrate under Microwave Irradiation in Solventless System

Hantzsch 1,4-dihydropyriines were rapidly and efficiently oxidized using bismuth nitrate supported onto silica gel under microwave irradiation.     

Measuring persistence in a stationary time series using the complex network theory

A growing interest exists currently in the analysis of time series by the complex network theory. Here we present a simple and quick way for mapping time series to complex networks. Using a simple rule allows us to transform time series into a textual sequence then we divide it into words with fixed size. Distinct words are nodes of the network, and we have complete control on the network scale by adjusting the word size. Two nodes are linked if their associated words co-occur in sequence. We show that the network topological measures quantify the persistence and the long range correlations in fractional Brownian processes. For a particular word size we assume some relations between the topological measures and the Hurst exponent which characterised the persistence in fractional Brownian processes.     

On U(1) Gauge Theory Transfer-Matrix in Fourier Basis*

The properties of the transfer-matrix of U(1) lattice gauge theory in the Fourier basis are explored. Among other statements it is shown: (i) the transfer-matrix is block-diagonal, (ii) all consisting vectors of a block are known based on an arbitrary block vector, (iii) the ground-state belongs to the zero-mode's block. The emergence of maximum-points in matrix-elements as functions of the gauge coupling is clarified. Based on explicit expressions for the matrix-elements we present numerical results as tests of our statements.     

A Methodology for Temperature Correction When Using Two-Color Pyrometers - Compensation for Surface Topography and Material

In this investigation, the applicability of the two-color pyrometer technique for temperature measurements in dry hard turning of AISI 52100 steel was studied, where both machined surfaces as well as cutting tools were considered. The impacts of differing hard turned surface topography on the two-color pyrometer readings was studied by conducting temperature measurements on reference samples created using cutting tools with different degrees of tool flank wear. In order to conduct measurements in a controlled environment, a specially designed furnace was developed in which the samples were heated step-wise up to 1,000 °C in a protective atmosphere. At each testing temperature, the temperatures measured by the two-color pyrometer were compared with temperatures recorded by thermocouples. For all materials and surfaces as studied here, the two-color pyrometer generally recorded significantly lower temperatures than the thermocouples; for the hard turned surfaces, depending on the surface topography, the temperatures were as much as ~20 % lower and for the CBN cutting tools, ~13 % lower. To be able to use the two-color pyrometer technique for temperature measurements in hard turning of AISI 52100 steel, a linear approximation function was determined resulting in three unique equations, one for each of the studied materials and surfaces. By using the developed approximation function, the measured cutting temperatures can be adjusted to compensate for differing materials or surface topographies for comparable machining conditions. Even though the proposed equations are unique for the hard turning conditions as studied here, the proposed methodology can be applied to determine the temperature compensation required for other surface topographies, as well as other materials.     

A New Reaction of Isatin,Cyclic 1,3‐Diketone,and 2‐Cyanoacetamide: A Four‐Component Synthesis of Spirooxindoles

A novel reaction of isatins, 2‐cyanoacetamide, and cyclic 1,3‐diketones in ethanol was reported. The reaction gave the unexpected spirooxindole ethyl carboxylates in excellent yields and the spirooxindole carboxamide was not observed.
