In the present study, we established a practical and cost-effective high throughput screening assay, which relies on the measurement of the motility of Caenorhabditis elegans by infrared light-interference. Using this assay, we screened 14,400 small molecules from the “HitFinder” library (Maybridge), achieving a hit rate of 0.3%. We identified small molecules that reproducibly inhibited the motility of C. elegans (young adults) and assessed dose relationships for a subset of compounds. Future work will critically evaluate the potential of some of these hits as candidates for subsequent optimisation or repurposing as nematocides or nematostats. This high throughput screening assay has the advantage over many previous assays in that it is cost- and time-effective to carry out and achieves a markedly higher throughput (~10,000 compounds per week); therefore, it is suited to the screening of libraries of tens to hundreds of thousands of compounds for subsequent evaluation and development. The present phenotypic whole-worm assay should be readily adaptable to a range of socioeconomically important parasitic nematodes of humans and animals, depending on their dimensions and motility characteristics in vitro, for the discovery of new anthelmintic candidates. The presence of organic dyes from industrial wastewater can cause pollution and exacerbate environmental problems; therefore, in the present work, activated carbon was synthesized from locally available oil palm trunk (OPT) biomass as a low-cost adsorbent to remove synthetic dye from aqueous media. The physical properties of the synthesized oil palm trunk activated carbon (OPTAC) were analyzed by SEM, FTIR-ATR, and XRD. The concurrent effects of the process variables (adsorbent dosage (g), methylene blue (MB) concentration (mg/L), and contact time (h)) on the MB removal percentage from aqueous solution were studied using a three-factor three-level Box–Behnken design (BBD) of response surface methodology (RSM), followed by the optimization of MB adsorption using OPTAC as the adsorbent. Based on the results of the analysis of variance (ANOVA) for the three parameters considered, adsorbent dosage (X1) is the most crucial parameter, with an F-value of 1857.43, followed by MB concentration (X2) and contact time (X3) with the F-values of 95.60 and 29.48, respectively. Furthermore, the highest MB removal efficiency of 97.9% was achieved at the optimum X1, X2, and X3 of 1.5 g, 200 mg/L, and 2 h, respectively. 相似文献
There has been growing interest among food scientists in producing a toxin-free fat as an end product with varying physical or nutritional properties of interest to the food industry. Oleoresin is a rich source of bioactive compounds which consumers can easily add to a large variety of food. Dabai (Canarium odontophyllum) pulp oleoresin (DPL) was extracted using supercritical carbon dioxide (SC-CO2) extraction, a green extraction technology. This study investigates the quality of SC-CO2 extracted DPL in discovering its potential as a new alternative fat. The extraction experiment was carried out at a pressure of 40 MPa and a temperature of 40 °C. DPL is a saturated fatty acid (SFA)-rich fat due to its high SFA composition (47.72 ± 0.01%). In addition, the low content of peroxide value (PV) (5.60 ± 0.09 mEq/kg) and free fatty acids (FFA) (3.40 ± 0.03%) indicate the quality and stability of DPL for various applications besides food consumption. DPL also has a low slip melting point (SMP) (20.20 ± 0.03 °C), and HPLC-FID revealed that DPL contained 0.13 ± 0.02 mg/100 g of vitamin E (α-tocopherol), indicating its potential application as a solid fat with a bioactive compound. It has been proved that having halogen or methoxy groups substituted in various positions of the phenyl ring enhances the activity of chalcones many times. The title compounds, C21H20O5 and C19H15BrO3, respectively, were chosen for crystallographic study in order to determine their structures and conformations. The promising biomedical applications of silver complexes stimulated the researchers to test these compounds against cancer. The present research work was designed to achieve this goal. In this work, a series of 5-methyl benzimidazole based N-Heterocyclic carbene ligands and respective silver(I) complexes were synthesized and tested on cancer cell lines to assess their anticancer activity. Unsymmetrically substituted benzimidazole was found unique in its reactivity and generation of a single product during NHC ligand formation was only possible after two successive alkylations with same alkyl halide. The corresponding Ag(I)-NHC adducts were obtained by in situ deprotonation of the NHC ligands. Synthesized compounds were characterized by various physcio-chemical and spectroscopic methods. Single crystal X-ray diffraction study of complex 7 revealed its mononuclear structure. Preliminary in vitro anticancer study of azolium salts and respective Ag(I)-NHC complexes against human breast cancer (MDA-MB-231), colon cancer (HCT-116) and normal endothelial cells (EA.hy926) cells revealed that all the compounds are more cytotoxic to cancer cells than normal cells and the complexes are relatively more potent compared to the corresponding NHC ligands. It was found that increased chain length and presence of methyl substituent on benzimidazole ring enhance the biopotency of Ag(I)-NHC complexes. The synthesized compounds were further studied for pro-apoptotic mechanism of action via Rhodamine 123 test. The present study was aimed to investigate the variation of stable isotopic ratios of carbon, nitrogen, hydrogen, and oxygen in wheat kernel along with different processed fractions from three geographical origins across 5 years using isotope ratio mass spectrometry (IRMS). Multiway ANOVA revealed significant differences among region, harvest year, processing, and their interactions for all isotopes. The region contributed the major variability in the δ13C ‰, δ2H ‰, δ15N ‰, and δ18O‰ values of wheat. Variation of δ13C ‰, δ15N ‰, and δ18O ‰ between wheat whole kernel and its products (break, reduction, noodles, and cooked noodles) were ?0.7‰, and no significant difference was observed, suggesting the reliability of these isotope fingerprints in geographical traceability of wheat‐processed fractions and foods. A significant influence of wheat processing was observed for δ2H values. By applying linear discriminant analysis (LDA) to the whole dataset, the generated model correctly classified over 91% of the samples according to the geographical origin. The application of these parameters will assist in the development of an analytical control procedure that can be utilized to control the mislabeling regarding geographical origin of wheat kernel and its products. 相似文献
Poly(limonene carbonate) (PLC) has been highlighted as an attractive substitute to petroleum derived plastics, due to its utilisation of CO2 and bio-based limonene as feedstocks, offering an effective carbon capture and utilisation pathway. Our study investigates the techno-economic viability and environmental sustainability of a novel process to produce PLC from citrus waste derived limonene, coupled with an anaerobic digestion process to enable energy cogeneration and waste recovery maximisation. Computational process design was integrated with a life cycle assessment to identify the sustainability improvement opportunities. PLC production was found to be economically viable, assuming sufficient citrus waste is supplied to the process, and environmentally preferable to polystyrene (PS) in various impact categories including climate change. However, it exhibited greater environmental burdens than PS across other impact categories, although the environmental performance could be improved with a waste recovery system, at the cost of a process design shift towards energy generation. Finally, our study quantified the potential contribution of PLC to mitigating the escape of atmospheric CO2 concentration from the planetary boundary. We emphasise the importance of a holistic approach to process design and highlight the potential impacts of biopolymers, which is instrumental in solving environmental problems facing the plastic industry and building a sustainable circular economy. 相似文献
The mineral extraction activities may disturb the natural radioactivity, therefore current study aims to generate baseline data of natural radionuclides and anthropogenic 137Cs before the start of industrial activities. Gamma spectrometry and gross alpha and beta counting systems were used for activity measurement in environmental samples. In soil, the mean activity of 232Th, 226Ra, 40K and 137Cs were determined as 79 (66–117), 47 (34–80), 823 (602–1159) and 1.3 (1.1–4.5) Bq kg?1, respectively. The average annual effective dose rate (128.7 µSv h?1) in the study area was twice higher than world’s average value. Indoor hazard index was greater than unity at two places; therefore, proper ventilation is proposed during construction.
Direct attachment of gold nanoparticles to a green support without the use of an external reducing agent and using it for removing toxic pollutants from wastewater, i. e., reduction of nitroarene to amine, are described. A novel approach involving the reduction of gold by the jute plant (Corchorus genus) stem-based (JPS) support itself to form nanoparticles (AuNPs) to be used as a catalytic system (‘dip-catalyst’) and its catalytic activity for the hydrogenation of series of nitroarenes in aqueous media are presented. AuNPs/JPS catalyst was characterized using SEM, UV-Vis, FTIR, TEM, XPS, and ICP-OES. Confined area elemental mapping exhibits uniform and homogeneous distribution of AuNPs on the support surface. TEM shows multi-faceted AuNPs in the range of 20–30 nm. The reactivity of AuNPs/JPS for the transfer hydrogenation of nitroarene as well as hydrogenation of quinoline under molecular H2 pressure was evaluated. Sodium borohydride, when used as the hydrogen source, demonstrates a high catalytic efficiency in the transfer hydrogenation reduction of 4-nitrophenol (4-NP). Quinoline is quantitatively and chemoselectively hydrogenated to 1,2,3,4-tetrahydroquinoline (py-THQ) using molecular hydrogen. Reusability studies show that AuNPs are stable on the support surface and their selectivity is not affected. 相似文献
