Micellar liquid chromatography with sodium dodecyl sulphate or Brij-35 as surfactants in the mobile phase was evaluated and compared with reversed phase liquid chromatography using conventional acetonitrile-water eluents for the separation and determination of desferrioxamine (DFO) and its complexes with aluminium (AIDFO) and iron (FeDFO) in uremic serum. Reversed phase liquid chromatography proved to be superior in terms of sensitivity and selectivity. The three solutes investigated were separated with a mobile phase of 13% (v/v) acetonitrile/phosphate buffer (5 mM, pH = 3.5) on a C(18) column and detected by ultraviolet absorption at 210 nm (DFO) and 220 nm (AlDFO and FeDFO). Limits of detection of 0.1 mug ml(-1) and relative standard deviation of 3-4% were obtained. The recovery from serum samples after ultramicrofiltration was around 90%. The method was applied to the determination of DFO, AlDFO and FeDFO in uremic serum.  相似文献

Microcystins, which have their origin in species of cyanobacteria present in freshwaters, have recently been found to be important contaminants of the aquatic environment at trace levels. HPLC and HPCE with UV detection have been applied in the determination of such toxic compounds. Immunoaffinity chromatography for the selective extraction and clean-up of microcystins has been successfully applied to different matrices. Simple protocols for unambiguous determination of these toxins are presented and the immunoaffinity clean-up is compared with conventionally used solid phase extraction procedures. The development and optimisation of an on-line preconcentration procedure based on field amplified sample stacking for the analysis of microcystins by HPCE in the micellar electrokinetic chromatography mode is also described, using borate buffer with the anionic surfactant SDS, as separation electrolyte. Results indicate that sub-nanogram/gram content of microcystins can be detected in water samples, while sub-microgram/gram concentrations can be determined in algae samples.  相似文献

In this communication, by means of stress relaxation experiments, the viscous stress at various strains during tensile deformation of oriented polyolefin samples including high density polyethylene (HDPE), linear low density polyethylene (LLDPE) and isotactic polypropylene (iPP), has been determined. The viscous stress in the oriented samples takes up to 50%-70% of the total stress, which is unusually high compared with their isotropic counterparts. The unusual high viscous stress was discussed based on mainly the existence of shish structure in oriented polyolefins, which could enhance the inter-lamella coupling significantly.  相似文献

The mechanism of tensile deformation in semi-crystalline polymers was studied based on true stress-strain curvesobtained with the aid of a video-controlled tensile set-up. The deformation is affected by both the crystalline and theamorphous phases. However, the relative weights of the two portions change with the deformation stage. At lowdeformations the coupling and coarse slips of the crystalline blocks dominate the mechanical properties, which allows thesystem to maintain a homogeneous strain distribution in the sample. As the stretching increases, at a critical strain the forcegenerated from entangled fluid portions reaches a critical value to destroy the crystallites. The dominant deformationmechanism then changes into a disaggregation - recrystallization process.  相似文献

Transition metal selenides have been widely studied as anode materials of sodium ion batteries(SIBs),however,the investigation of solid-electrolyte-interface(SEI)on these materials,which is critical to the electrochemical performance of SIBs,remains at its infancy.Here in this paper,ZnSe@C nanoparticles were prepared from ZIF-8 and the SEI layers on these electrodes with and without reduced graphene oxide(rGO)layers were examined in details by X-ray photoelectron spectroscopies at varied charged/discharged states.It is observed that fast and complicated electrolyte decomposition reactions on ZnSe@C leads to quite thick SEI film and intercalation of solvated sodium ions through such thick SEI film results in slow ion diffusion kinetics and unstable electrode structure.However,the presence of rGO could efficiently suppress the decomposition of electrolyte,thus thin and stable SEI film was formed.ZnSe@C electrodes wrapped by rGO demonstrates enhanced interfacial charge transfer kinetics and high electrochemical performance,a capacity retention of 96.4%,after 1000 cycles at 5 A/g.This study might offer a simple avenue for the designing high performance anode materials through manipulation of SEI film.  相似文献

Human replication protein A (RPA) is a single-stranded DNA (ssDNA)-binding protein with three subunits. The largest subunit, p70, contains a conserved (cysteine)(4)-type zinc-finger motif that has been implicated in the regulation of DNA replication and repair. Previous studies indicated that the ssDNA-binding activity of RPA could be redox-regulated via reversible oxidation of cysteines in the zinc-finger motif. We exposed recombinant human RPA to hydrogen peroxide and characterized the oxidized protein by liquid chromatography/tandem mass spectrometric (LC/MS/MS) analyses. Our results demonstrated that, upon H(2)O(2) treatment, four cysteines, which reside at the zinc-finger motif of the p70 subunit, could result in the formation of two pairs of intramolecular disulfides, Cys481-Cys486 and Cys500-Cys503; no cysteine sulfinic acid or cysteine sulfonic acid could be found. Moreover, the other 11 cysteines in this protein remained intact. The results demonstrated that the formation of disulfide bonds at the zinc-finger site was responsible for the redox regulation of the DNA-binding activity of RPA.  相似文献

分析型气相色谱仪对低沸点易挥发的有机化合物展现出优异的分离性能,通过在其色谱柱末端加装馏分收集装置,建立了现代制备气相色谱(Prep GC)技术,该技术可用于挥发性成分的快速分离富集。制备气相色谱仪是由分析型气相色谱仪改装而来,其进样系统、分离系统、检测系统、馏分收集系统也在不断地优化升级,以提高目标化合物的回收效率和纯度。Prep GC与现代波谱技术(如紫外可见吸收光谱、红外吸收光谱、拉曼光谱、质谱、X射线衍射、核磁共振波谱)耦合,可对分离富集得到的目标化合物的结构进行精准确证。近年来,与Prep GC在各种挥发性成分分离中的应用相关的报道逐渐增多,展现出良好的应用前景。然而,Prep GC在分离过程中也存在着无法制备热敏性化合物、分离成本高、易引入外源性污染等问题。该文根据近年来国内外研究学者的相关研究工作,对制备气相色谱仪的结构及其在精油单体化合物、昆虫信息素、食品和植物挥发性成分、地质生物标志物及持久性环境污染物的分离等领域的应用研究进展进行综述。最后,还对Prep GC在挥发性成分分离中的应用进行了总结与展望,旨在为拓展Prep GC应用领域提供参考。  相似文献
