A new series of aryl-based synthetic mimics of antimicrobial peptides (SMAMPs) with antimicrobial activity and selectivity have been developed via systematic tuning of the aromatic groups and charge. The addition of a pendant aromatic group improved the antimicrobial activity against Gram-negative bacteria, while the addition of charge improved the selectivity. SMAMP 4 with six charges and a naphthalene central ring demonstrated a selectivity of 200 against both Staphylococcus aureus and Escherichia coli , compared with a selectivity of 8 for the peptide MSI-78. In addition to the direct antimicrobial activity, SMAMP 4 exhibited specific immunomodulatory activities in macrophages both in the presence and in the absence of lipopolysaccharide, a TLR agonist. SMAMP 4 also induced the production of a neutrophil chemoattractant, murine KC, in mouse primary cells. This is the first nonpeptidic SMAMP demonstrating both good antimicrobial and immunomodulatory activities. 相似文献

基于绿原酸对鲁米诺-H2O2-辣根过氧化物酶化学发光体系具有强烈的抑制作用,建立了绿原酸的化学发光分析法,并探讨了其作用机理.化学发光强度的变化值与绿原酸浓度的对数在5.2×10-9~1.0×10-6 g/mL范围内呈线性关系,检出限(S/N =3)达7.8×10-10 g/mL.该方法成功用于金银花中绿原酸含量的测定,回收率为96.0%~100.7%. 相似文献

The importance of the HSO(2) system in atmospheric and combustion chemistry has motivated several works dedicated to the study of associated structures and chemical reactions. Nevertheless, controversy still exists about a possible connection between the upper and lower energy regions of the potential energy surface (PES) for the ground electronic state of the system. Very recently, a path to connect these regions was proposed based on studies at the CASPT2/aug-cc-pV(T+d)Z level of calculation but the small energy difference between some of the transitions states along that path suggested the necessity of calculations at a higher level of theory. In the present work, we report a CCSD(T)/aug-cc-pV(T+d)Z study of the stationary states associated to the proposed connection path, including assessment of the most reliable complete basis set (CBS) extrapolation scheme for the system. Among the new features, the present calculations show that there are no structures corresponding to the HSO(2)(b) minimum and the TS3 saddle point obtained at the CASPT2 level and that the connection path between the upper and lower energy regions of the PES for the ground electronic state involves only one transition state and most probably more than one electronic state. 相似文献

Three new monosubstituted sucrose fatty acid esters, 1 – 3 , were isolated from Equisetum hiemale L., together with nine known compounds, 4 – 12 . Their structures were elucidated by spectroscopic analyses. Compounds 5, 6 , and 10 – 12 were isolated from the title plant for the first time. An efficient and reusable heterogeneous catalytic assembly of PdCl2 held in ionic liquid brushes has been synthesized and an environmentally‐friendly procedure was developed for coupling aryl iodides with acrylic acid. These reactions were conducted in water under aerobic conditions with water‐insoluble or even solid aryl iodides and they proceeded smoothly and cleanly without any organic co‐solvent or other additives. A 0.5 mol% (based on Pd atom) dose of the catalyst was found to be sufficient for Mizoroki–Heck reaction. The catalyst is easily recovered post reaction, via simple filtration, and reused at least eight times without a noticeable loss of activity. 以卟啉及其衍生物和特异性染料为敏感化学元件, 基于交叉响应原理构建了识别蛋白质的可视6×6阵列. 该阵列以颜色差谱图显示其与蛋白质作用呈现的特异性光谱反应, 采用聚类分析、 主成分分析和欧氏距离对图谱进行了分析. 结果表明, 该阵列可以鉴别模式蛋白牛血清白蛋白(BSA)、 牛血红蛋白(BHb)和卵清白蛋白(Ova)及其混合物, 且有望实现定量分析. 此外, 阵列的高敏感性使其不仅能识别天然蛋白质和不同变性程度的蛋白质, 还能对其热变性过程进行可视化实时监控. 该阵列产生的特殊颜色变化与蛋白质的空间构型、 微环境pH值的差异及溶解度有关. 因此, 该方法不仅能实现对蛋白质的快速识别, 为蛋白质热变性机理的研究提供新途径, 而且在临床医学和食品安全等的实时快速检测方面有潜在的应用价值. 相似文献

Density functional theory calculations (B3LYP) have been carried out to investigate the 4π‐electron systems of 2,4‐disila‐1,3‐diphosphacyclobutadiene (compound 1 ) and the tetrasilacyclobutadiene dication (compound 2 ). The calculated nucleus‐independent chemical shift (NICS) values for these two compounds are negative, which indicates that the core rings of compounds 1 and 2 have a certain amount of aromaticity. However, deep electronic analysis reveals that neither of these two formal 4π‐electron four‐membered ring systems is aromatic. Compound 1 has very weak, almost negligible antiaromaticity, and the amidinate ligands attached to the Si atoms play an important role in stabilizing this conjugated 4π‐electron system. The monoanionic bidentate ligand interacts with the conjugated π system to cause π‐orbital splitting. This ligand‐induced π‐orbital splitting effect provides an opportunity to manipulate the gap between occupied and unoccupied π orbitals in conjugated systems. Conversely, compound 2 is nonaromatic because its core ring does not have a conjugated π ring system and does not fulfill the requirements of a Hückel system. 相似文献

The DNA fragment detection focusing technique has further enhanced the sensitivity and information of DNA targets. The DNA fragment detection method was established by capillary electrophoresis with laser‐induced fluorescence detection and restriction endonuclease chromatographic fingerprinting (CE‐LIF‐REF) in our experiment. The silica capillary column was coated with short linear polyarclarylamide (SLPA) using nongel sieving technology. The excision product of various restricted enzymes of DNA fragments was obtained by REF with the molecular biology software Primer Premier 5. The PBR322/BsuRI DNA marker was used to establish the optimization method. The markers were focused electrophoretically and detected by CE‐LIF. The results demonstrate that the CE‐LIF‐REF with SLPA can improve separation, sensitivity and speed of analysis. This technique may be applied to analysis of the excision product of various restricted enzymes of prokaryotic plasmid (pIRES2), eukaryote plasmid (pcDNA3.1) and the PCR product of codon 248 region of gastric cancer tissue. The results suggest that this method could very sensitively separate the excision products of various restricted enzymes at a much better resolution than the traditional agarose electrophoresis. Double‐stranded DNA and multiwalled carbon nanotube (MWNT) complex modified glassy carbon electrodes (DNA‐MWNT‐GCE) were employed to discriminate penicillamine (PA) enantiomers. Cyclic voltammetry, electrochemical impedance spectroscopy, atomic force microscopy and ultraviolet‐visible spectroscopy were used to characterize the enantioselective phenomenon. The results indicated that the binding effect between L ‐PA and DNA‐MWNTs was stronger than that of D ‐PA and DNA‐MWNTs. In addition, the influencing factors of the modified electrodes were systematically investigated. The modified electrodes exhibited a linear response towards PA enantiomers from 1.0×10?1 to 1.0×10?8 mol L?1 and detection limits of 3.1×10?9 and 3.3×10?8 mol L?1 for L ‐PA and D ‐PA, respectively. 相似文献
