Background: Element-tagged immunoassay coupled with inductively coupled plasma mass spectrometry (ICP-MS) detection has the potential to revolutionize immunoassay analysis for multiplex detection. However, a further study referring to the standard evaluation and clinical sample verification is needed to ensure its reliability for simultaneous analysis in clinical laboratories. Methods: Carcinoembryonic antigen (CEA) and α-fetoprotein (AFP) were chosen for the duplex immunoassay. The performance of the assay was evaluated according to guidelines from the Clinical and Laboratory Standards Institute (CLSI). Moreover, reference intervals (RIs) of CEA and AFP were established. At last, 329 clinical samples were analyzed by the proposed method and results were compared with those obtained with electrochemiluminescent immunoassay (ECLIA) method. Results: The measurement range of the assay was 2–940 ng/mL for CEA and 1.5–1000 ng/mL for AFP, with a detection limit of 0.94 ng/mL and 0.34 ng/mL, respectively. The inter-assay and intra-assay imprecision were all less than 6.58% and 10.62%, respectively. The RI of CEA and AFP was 0–3.84 ng/mL and 0–9.94 ng/mL, respectively. Regarding to clinical sample detection, no significant difference was observed between the proposed duplex assay and the ECLIA method. Conclusions: The ICP-MS-based duplex immunoassay was successfully developed and the analytical performance fully proved clinical applicability. Well, this could be different with other analytes. 相似文献

采用常压金属有机物化学气相沉积技术(AP-MOCVD),以二乙基锌(DEZn)为Zn源,去离子水(H2O)为氧源,N2作载气,在外延ZnO薄膜的反应气氛中通入少量氢气,在c-Al2O3衬底上生长出了ZnO∶H薄膜。用X射线双晶衍射和光致发光谱对ZnO∶H薄膜的结晶性能和光学性质进行表征。结果表明,ZnO∶H薄膜(002)和(102)面的Ω扫描半峰全宽分别为46.1 mrad和81.4 mrad,表明该薄膜具有良好的结晶性能;室温下,ZnO∶H薄膜具有较强的紫外光发射(380 nm),在低温10 K光致发光谱中观测到位于3.3630 eV处与氢相关的中性施主束缚激子峰(I4)及其位于3.331 eV处的双电子卫星峰(TES)。采用退火的方法,通过观测I4峰的强度变化,研究了氢在ZnO∶H薄膜中的热稳定性。发现随着退火温度的升高,I4峰的强度逐渐减弱,表明在高温下退火,氢会从ZnO薄膜中逸出。 相似文献

5-Hydroxymethyl-2′-deoxycytidine (5hmdC) phosphoramidite and triphosphate are important building blocks in 5hmdC-containing DNA synthesis for epigenetic studies. However, efficient and practical methods for the synthesis of these compounds are still limited. The current research provides an intensively improved synthetic method that enables the preparation of commercially available cyanoethyl-protected 5hmdC phosphoramidite with an overall yield of 39% on 5 g scale. The molecular weight distribution is an important factor that affects the properties of polymers. A control algorithm based on the moment-generating function was proposed to regulate the molecular weight distribution for polymerization processes in this work. The B-spline model was used to approximate the molecular weight distribution, and the weight state space equation of the system was identified by the subspace state space system identification method based on the paired date of B-spline weights and control inputs. Then, a new performance criterion mainly consisting of the moment-generating function was constructed to obtain the optimal control input. The effectiveness of the proposed control method was tested in a styrene polymerization process. The molecular weight distribution of the styrene polymers can be approximated by the B-spline model effectively, and it can also be regulated towards the desired one under the proposed control method. 相似文献

The interaction between[Hg(SCN)4]2- and hemoglobin(Hb) under conditions that simulate a physiological environment was investigated by UV-vis spectroscopy,fluorescence spectroscopy,resonance Rayleigh scattering(RRS) spectroscopy and circular dichroism(CD) spectroscopy.The results obtained from the change of UV-vis and CD spectra,the quenching of Hb fluorescence and the enhancement of RRS intensity proved that a 10:1 type complex was formed between[Hg(SCN)4]2- and Hb.The possible mechanism suggested for the interaction was that ten Hg(SCN)4]2- anions entered the four subunits of a Hb molecule to react with some residues to form an adduct by coordination and electrostatic forces.The coordination of[Hg(SCN)4]2- with Trp was the major cause of the fluorescence quenching of Hb. 相似文献
