Development of Microtiter Plate Culture Method for Rapid Screening of ε-Poly-L-Lysine-Producing Strains

ε-Poly-l-lysine (ε-PL) produced by Streptomyces albulus possesses a broad spectrum of antimicrobial activity and is widely used as a food preservative. To extensively screen ε-PL-overproducing strain, we developed an integrated high-throughput screening assay using ribosome engineering technology. The production protocol was scaled down to 24- and 48-deep-well microtiter plates (MTPs). The microplate reader assay was used to monitor ε-PL production. A good correlation was observed between the fermentation results obtained in both 24-(48)-deep-well MTPs and conventional Erlenmeyer flasks. Using this protocol, the production of ε-PL in an entire MTP was determined in <5 min without compromising on accuracy. The high-yielding strain selected through this protocol was also tested in Erlenmeyer flasks. The result showed that the ε-PL production of the high-yielding mutants was nearly 45% higher than that of the parent stain. Thus, development of this protocol is expected to accelerate the selection of ε-PL-overproducing strains.     

On the 4D nonlinear Schrödinger equation with combined terms under the energy threshold

In this paper, we consider the longtime dynamics of the solutions to focusing energy-critical Schrödinger equation with a defocusing energy-subcritical perturbation term under a ground state energy threshold in four spatial dimension. This extends the results in Miao et al. (Commun Math Phys 318(3):767–808, 2013, The dynamics of the NLS with the combined terms in five and higher dimensions. Some topics in harmonic analysis and applications, advanced lectures in mathematics, ALM34, Higher Education Press, Beijing, pp 265–298, 2015) to four dimension without radial assumption and the proof of scattering is based on the interaction Morawetz estimates developed in Dodson (Global well-posedness and scattering for the focusing, energy-critical nonlinear Schrödinger problem in dimension \(d =4\) for initial data below a ground state threshold, arXiv:1409.1950), the main ingredients of which requires us to overcome the logarithmic failure in the double Duhamel argument in four dimensions.     

Analysis of Glycocholic Acid in Human Plasma and Urine from Hepatocellular Carcinoma Patients

Glycocholic acid (GCA) has been identified as endogenous biomarker for hepatocellular carcinoma (HCC). To dissolve protein and liberate GCA from protein, ionic liquids (ILs) that contain chaotropic ions were used for pretreatment of liquid biological samples. Coupling with solid-phase extraction (SPE) and reversed-phase high-performance liquid chromatography (RP-HPLC), the novel sample pretreatment method was applied for quantitative determination of GCA in urine and plasma samples. Compared with the traditional organic solvents pretreatment of biological samples, the proposed method is “greener” and simpler, due to no use of volatile organic solvent, and avoiding centrifugation. Under the optimal conditions, when the GCA-free urine and plasma samples were spiked with GCA at 0.05–1.0 and 0.2–10 μmol L^?1, the recoveries of GCA ranged between 95.8–101.6 and 96.9–100.4%, respectively. These procedures only required 1 mL of urine and 3 mL of 3 mM ILs aqueous solution and 100 μL of plasma and 4 mL of 2 mM ILs aqueous solution, respectively. The proposed method has been successfully validated on a small sample size of 14 HCC patients and 14 healthy volunteers. For HCC patients, the mean concentration of GCA was 24.79 ± 6.86 and 31.98 ± 11.12 μmol L^?1 in urine and plasma samples, and was about 3 times and 24 times as much as in healthy people, respectively. The proposed method opens up a new possibility in early diagnosis of HCC.     

Recent progress toward the total synthesis of humulanolides

The humulanolides are a series of sesquiterpene lactones, most of which have unique and challenging structure. The humulanolides have exhibited anticancer activity. The combinations of fascinating structural motifs and promising pharmacological properties have prompted significant interest in the synthetic community. In this review, we provide a summary of recent progress regarding the total synthesis of humulanolides.     

Transition-metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols with trifluoromethanesulfonyl chloride and difluoromethanesulfonyl chloride

Triphenylphosphine-mediated metal-free trifluoromethylthiolation and difluoromethylthiolation of thiols by CF₃SO₂Cl and CHF₂SO₂Cl to synthesize trifluoromethyl disulfides and difluoromethyl disulfides, respectively, was achieved at room temperature. Iodine generated in situ from iodide facilitated this reaction via the formation of iodotriphenylphosphonium iodide which could serve as a reducing agent in this transformation. Readily available reagents and mild reaction conditions without transition-metals allow this protocol to be more practical than traditional methods.     

One step synthesis of indirubins by reductive coupling of isatins with KBH4

One step synthesis of the natural product indirubin by reductive coupling of isatin with KBH₄ is described and a possible mechanism for the reaction is proposed. Eleven indirubin derivatives were obtained easily from the corresponding substituted isatin.     

The colorimetric and ratiometric fluorescent detection of cyanide and sulfide in live cells,application for logic gate and bioimging

We described the design and synthesis of a colorimetric and ratiometric fluorescent probe based on a conjugated π-electron system that displayed differential fluorescence responses towards cyanide and sulfide and demonstrated its utility in intracellular ion imaging and logic gate.     

Synthesis of 2,3,6-trisubstituted pyridines by transition-metal free cyclization of 1,3-diynes with amino acids

Amino acids are firstly employed in transition-metal free heterocyclization reaction of 1,3-diynes in the presence of K₃PO₄ and DMSO at 120 °C. This method produces 2,3,6-trisubstituted pyridines with up to 86% yield. The –CO₂H group on the amino acids is crucial for this heterocyclization reaction. The mechanism of such a heterocyclization reaction is discussed, as well.
