We measured the affinity of more than 20 sugars with concanavalin A (ConA) by an optical biosensor (surface plasmon resonance sensor) using asialofetuin (ASF) as an immobilized binding partner of ConA. We determined kinetic parameters of the effects of sugars on the dissociation of ConA from ASF quantitatively, and the structural requirements of the functional groups of sugars for binding with ConA. We found that the affinity of ConA for sugars is dependent on its conformation induced by interaction with the binding partner. In addition, the results showed that optical biosensor system is well mimics the interaction of ConA with sugars in biomembrane. 相似文献

A novel method for the direct analysis of condensed tannin components in bark was developed on the basis of matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) combined with a simple solid sampling technique. The MALDI mass spectra obtained from the wood (bark) powder sample clearly showed a series of peaks corresponding to the sodium ion adducts of condensed tannin oligomers up to around m/z 3000. The results indicate that the condensed tannins in the bark sample used in this work mostly consist of combinations of flavan-3-ol units such as profisetinidin (PF), prorobinetinidin (PR) and prodelphinidin (PD), at least up to 10-mers (m/z approximately 3000). 相似文献

A flow-injection system is described for the simultaneous determination of calcium and magnesium with simultaneous injection of two sample plugs and a masking agent plug and with a single detector. The system utilizes the simultaneous injection of an ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetra-acetic acid plug and two small sample plugs into the same carrier stream and which are merged downstream with 3,3'-bis[N,N-bis(carboxymethyl)aminomethyl)]-o-cresolphthalein solution for spectrophotometric determination of calcium and magnesium. The results for the analysis of natural waters by the proposed method correspond well with those obtained by the conventional titration method with ethylenediaminetetra-acetic acid, and showed good reproducibility. To study the thermodynamic and mechanical stability of toroidal isomers of C 240, we use a semi-empirical tight-binding theory and calculate their electronic structure, cohesive energy and vibrational spectra within the harmonic approximation. From these, we deduce their free energy at temperatures up to 1500K. The results are also compared to the isomer with icosahedral symmetry. Structure of biluciferyl (luciferyl radical dimer), which is produced by chemical oxidation of . luciferin with such as ferricyanide, was determined to be the symmetric 5,5′-dimer of . luciferin. It gives light in the presence of . luciferase, although the bioluminescent rate is very low. Synthetic, structural, spectroscopic, and kinetic studies have been carried out on the Pd(II) complexes of new 2N1O-donor ligands containing a pendent indole, 3-(N-2-pyridylmethyl-N-2-hydroxy-5-methoxybenzylamino)ethylindole (HMeO-iepp), 3-(N-2-pyridylmethyl-N-2-hydroxy-5-nitrobenzylamino)ethylindole (HNO2-iepp), and (N-2-pyridylmethyl-3-indolylethylamino)acetic acid (Hiepc) (H denotes a dissociable proton). [Pd(MeO-iepp)Cl] (2), [Pd(NO2-iepp)Cl] (3), and [Pd(iepc)Cl] (4) were prepared and revealed by X-ray analysis to have a pyridine nitrogen, an amine nitrogen, a phenolate or carboxylate oxygen, and a chloride ion in the coordination plane. UV absorption and 1H NMR spectral changes indicated that all the complexes could be converted to the indole-binding complexes where the O donor was replaced by the indole C2 atom by cyclopalladation in DMSO or DMF in the temperature range of 40-60 degrees C. Formation of the indole-binding complex species obeyed the first-order kinetics, from which the activation parameters were estimated. The formation rate was dependent on the properties of the O-donor group, a lower pKa value of its conjugate acid causing faster conversion to the indole-binding species in the order 2 (methoxyphenolate) < 3 (nitrophenolate) < 4 (carboxylate). On the other hand, the ratio of the indole-binding complex to the O-donor complex as a result of the conversion was greater for the complexes with a higher pKa value of the ligand OH group, the order being 2 > 3 > 4. 相似文献

In the spectrophotometric determination of aluminium and iron with ferron (7-iodo-8-quinolinol-5-sulphonic acid, H(2)L), the addition of cationic surfactants greatly improves the linearity of the calibration curve and widens the useful pH range. The effect of cetyltrimethylammonium chloride (CTMAC) on the stepwise stability constants (K(1),K(2) and K(3)) of the ferron complexes of aluminium and iron (ML(+), ML(-)(2) and ML(3-)(3)) and on the acid-dissociation constants (K(a1) and K(a2)) of ferron has been studied in connection with the role of the surfactant. CTMAC greatly increases the value of K(3) while exerting little effect on K(1) and K(2), thus rendering ML(3-)(3) the predominant species even at very low concentration of free L(2-). It also has some effect on the acid-dissociation constants of ferron, but sometimes it acts to decrease the free L(2-) concentration. At is therefore concluded that the improvements due to addition of surfactant should be attributed to the increased K(3) value. The presence of surfactant micelles is not essential, because the surfactant has a favourable effect when present at well below its critical micelle concentration, and because the continuous variations plots show a peak at a point corresponding to the composition M: L: Q (Q = cationic surfactant) = 1:3:3. 相似文献

A linearized optimal control method in combination with mixed quantum/classical molecular dynamics simulation is used for numerically investigating the possibility of controlling photodissociation wave packets of I(2)(-) in water. Optimal pulses are designed using an ensemble of photodissociation samples, aiming at the creation of localized dissociation wave packets. Numerical results clearly show the effectiveness of the control although the control achievement is reduced with an increase in the internuclear distance associated with a target region. We introduce effective optimal pulses that are designed using a statistically averaged effective dissociation potential, and show that they semiquantitatively reproduce the control achievements calculated by using optimal pulses. The control mechanisms are interpreted from the time- and frequency-resolved spectra of the effective optimal pulses. 相似文献

Isotachophoresis carried out in a 0.25 mm i.d. fused-silica capillary tube yielded high resolution, compared with that in a fluorinated ethylene-propylene polymer tube. The use of an ultraviolet-visible multichannel spectrophotometer with photodiode array as detector together with a cross flow cell (volume 0.01 μl) was investigated.
