Reversed phase‐high performance liquid chromatography (RP‐HPLC), thin layer chromatography (TLC) densitometry and first derivative spectrophotometry (1D) techniques are developed and validated as a stability‐indicating assay of ezetimibe in the presence of alkaline induced degradation products. RP‐HPLC method involves an isocratic elution on a Phenomenex Luna 5μ C18 column using acetonitrile: water: glacial acetic acid (50:50:0.1 v/v/v) as a mobile phase at a flow rate of 1.5 mL/min. and a UV detector at 235 nm. TLC densitometric method is based on the difference in Rf‐values between the intact drug and its degradation products on aluminum‐packed silica gel 60 F254 TLC plates as stationary phase with isopropanol: ammonia 33% (9:1 v/v) as a developing mobile phase. On the fluorescent plates, the spots were located by fluorescence quenching and the densitometric analysis was carried out at 250 nm. Derivative spectrophotometry, the zero‐crossing method, ezetimibe was determined using first derivative at 261 nm in the presence of its degradation products. Calibration graphs of the three suggested methods are linear in the concentration ranges 1–10 mcg/mL, 0.1–1 mg/mL and 1–16 mcg/mL with a mean percentage accuracy of 99.05 ± 0.54%, 99.46 ± 0.63% and 99.24 ± 0.82% of bulk powder, respectively. The three proposed methods were successfully applied for the determination of ezetimibe in raw material and pharmaceutical dosage form; the results were statistically analyzed and compared with those obtained by the reported method. Validation parameters were determined for linearity, accuracy and precision; selectivity and robustness and were assessed by applying the standard addition technique.  相似文献

Sarcophine (1), a cembranoid diterpene is known to inhibit the process of tumorigenesis. Sarcophine can be isolated in large amounts from the Red Sea soft coral Sarcophyton glaucum and hence is an ideal target for semisynthetic or biocatalytic modifications. Hydroxylated derivatives of 1 were reported to improve its anticancer activity. Despite the promising results and ready availability, there are limited attempts towards further diversifying the library of sarcophine derivatives. Hence, the current study targets the epoxide ring to generate sulfur-containing derivatives of sarcophine by reacting it with ammonium thiocyanate and Lawesson's reagent. Structure elucidation of the products was based on extensive 1D and 2D NMR and high resolution mass spectrometry, in addition to mechanistic considerations. The effect of these derivatives on highly malignant +SA mammary epithialial cell proliferation is reported. Anti-inflammatory potential of sarcophine and its derivatives is also demonstrated.  相似文献

Three spectrophotometric methods including Vierordt's method, derivative, ratio spectra derivative, and thin layer chromatography (TLC)-UV densitometric method were developed for simultaneous determination of drotaverine HCl (DRT) and nifuroxazide (NIF) in presence of its impurity, 4-hydroxybenzohydrazide (4-HBH). In Vierordt's method, (E(1 cm)(1%)) values were calculated at 227 and 368 nm in the zero-order spectra of DRT and NIF. By derivative spectrophotometry, the zero-crossing method, drotaverine HCl was determined using the second derivative at 245 nm and the third derivative at 238 nm, while nifuroxazide was determined using the first derivative at 399 nm and the second derivative at 411 nm. The ratio spectra derivative spectrophotometry is basedon the measure of the amplitude at 459 nm for DRT and at 416 nm for NIF in the first derivative of the ratio spectra. Calibration graphs of the three spectrophotometric methods were plotted in the range 1-10 mug/ml of DRT and 2-20 mug/ml of NIF. TLC-UV densitometric method was achieved on silica gel plates using ethyl acetate : methanol : ammonia 33% (10 : 1 : 0.1 v/v/v) as the mobile phase. The Rf values were 0.74, 0.50, 0.30+/-0.01 for DRT, NIF and 4-HBH, respectively. On the fluorescent plates, the spots were located by fluorescence quenching and the densitometrical area were measured at 308 and 287 nm with linear range 0.2-4 mug/spot and 0.6-12 mug/spot for DRT and NIF, respectively. The proposed methods have been successfully applied to the commercial pharmaceutical formulation without any interference of excipients. Mean recoveries, relative standard deviations and the results of the proposed methods were compared with those obtained by applying the alternate methods.  相似文献

The development of chemical reactions in nanospaces is of paramount importance for the development of active nanodevices, particularly in nanofluidics. It has been shown in a previous paper that phospholipid vesicles can be incorporated without spontaneous bilayer rupture into poly-L-glutamic acid/poly(allylamine) (PGA/PAH) multilayered polyelectrolyte films. The aim of the present study was to use such a system as an "embedded submicronic reactor" able to trigger precipitation of calcium phosphates within closed spaces through an enzymatic reaction, the enzyme also being encapsulated in the vesicle interior. To this aim, large unilamellar vesicles (LUVs) were produced containing calcium ions as active ions in the mineralization process, spermine as an activator of crystal growth, and alkaline phosphatase as a catalyst to convert phosphate esters into phosphates. After stabilization by adding a layer of poly-(D-lysine), these vesicles were embedded in a (PGA-PAH)n film. A paranitrophenyl phosphate containing solution was then put in contact with this film. It is shown by means of infrared spectroscopy in the attenuated total reflection mode that, consecutively to this contact, calcium phosphates are growing inside the embedded vesicles. By using scanning near-field fluorescence microscopy, it is demonstrated that the alkaline phosphatase enzymes are most probably located inside the vesicles after their embedding. In addition, atomic force microscopy was used to show, after chemical removal of the organic top layer of the film, that the inorganic platelets produced after the precipitation reaction are localized in volumes of similar size and shape as that of the vesicles into which the phosphate ester hydrolysis and subsequent precipitation reaction did occur.  相似文献

We report resonance associated with polyurea layers sandwiched between identical acrylic and polycarbonate plates when subjected to laser-generated stress waves of several nanoseconds in duration. Transmitted stress wave amplitudes almost 16 times the incident stress wave amplitudes are observed. An elastodynamics simulation identified the thickness of the polyurea layer as the key parameter controlling the amplitude of the transmitted stress wave. This resonance effect was found absent when the polyurea was sandwiched between the steel and aluminum plates of similar thickness. However, for these samples, a dramatic reduction in the amplitudes of the transmitted stress waves was recorded. A finite element analysis of the wave propagation through the sandwiched polyurea layer in these samples tied the large amplitude reduction to the large acoustic impedance mismatch and the viscoelastic dissipation within the polyurea layer.  相似文献

The quaternaries \(In_{1 - x} Ga_{x} As_{y} P_{1 - y}\) are the main promising elements for the fabrication of optoelectronic devices. The adjustment of their physical parameters is assumed by the change of the molar fraction \(x\) and \(y\). These parameters can be affected by the variation of temperature and pressure. To make the theoretical diagnosis of these materials, it is fundamental to know the energy gap ‘\(\varvec{E}_{\varvec{g}}\)’ and the lattice parameter ‘\(a\)’, over a wide range of chemical compositions \(0 \le x \le 0.47\) and \(0 \le y \le 1\), at different temperatures and pressures. We show that by using the Artificial Neural Network method optimized by the Levenberg Maquardt algorithm ANN-LM, it is possible to obtain results very close to the experiment. The scatter plot and error calculation show that the ANN-LM model provides more accurate values of the lattice parameter than those calculated by Vegard’s law. On the other hand, the energy gap values \(Eg (x, y, T)\) estimated, using the ANN-LM model, proved to be close to the experimental values that those calculated by the empirical equations. In addition, the ANN-LM method allowed us to estimate with great accuracy the values of the energy gap at different temperatures and pressures \(Eg (P, T)\). Our work provides crucial information on the physical properties of the quaternary without the use of approximations, and without taking into account the hypothesis of a perfect agreement between \(InGaAsP\) and \(InP\) substrate.  相似文献

The sensitization of the excited triplet state of a novel symmetrical Bis(dialkylamino)phenoxazinium salt was developed in the presence of Hg2+. This effect was used to determine the concentration of Hg2+ in different water samples. The phenoxazinium salt sensor was characterized by different spectroscopic tools such as: UV, FTIR, NMR and fluorescence spectra. The sensor has an emission band at 347 nm in DMSO. Hg2+ in DMSO at pH 5.6 can remarkably quench the fluorescence intensity of the sensor at 347 nm and a new band was appeared at 436 nm due to the strong complex formation between Hg2+ and sensor. The quenching of the band intensity at 347 and the enhancement of the intensity of the new band at 436 were used to determine the Hg2+ in different waste water samples. The dynamic range found for the determination of Hg2+ concentration is 8.7?×?10-10 – 1.4?×?10-6 mol L?1 with a detection limit of 5.8?×?10?10 mol L?1 and quantification detection limit of 1.8?×?10-9 mol L-1.  相似文献
