The solid‐phase combinatorial synthesis of cyclodepsipeptide destruxin E has been demonstrated. The combinatorial synthesis of cyclization precursors 8 was achieved by using a split and pool method on SynPhase Lanterns. The products were successfully macrolactonized in parallel in the solution phase by using 2‐methyl‐6‐nitrobenzoic anhydride and 4‐(dimethylamino)pyridine N‐oxide to afford macrolactones 9 , and the subsequent formation of an epoxide in the side chain gave 18 member destruxin E analogues 6 . Biological evaluation of analogues 6 indicated that the N‐MeAla residue was crucial to the induction of morphological changes in osteoclast‐like multinuclear cells (OCLs). Based on structure–activity relationships, azido‐containing analogues 15 were then designed for use as a molecular probe. The synthesis and biological evaluation of analogues 15 revealed that 15 b , in which the Ile residue was replaced with a Lys(N3) residue, induced morphological changes in OCLs at a sufficient concentration, and modification around the Ile residue would be tolerated for attachment of a chemical tag toward the target identification of destruxin E ( 1 ).  相似文献

Structures and magnetic properties of double perovskite-type oxides Eu2LnTaO6 (Ln=Eu, Dy-Lu) were investigated. These compounds adopt a distorted double perovskite structure with space group P21/n. Magnetic susceptibility, specific heat, and 151Eu Mössbauer spectrum measurements show that the Eu2+ ions at the 12-coordinate sites of the perovskite structure are antiferromagnetically ordered at ∼4 K, and that Ln3+ ions at the 6-coordinate site are in the paramagnetic state down to 1.8 K.  相似文献

It was noted in a graft copolymerization reaction involving a system of poly(vinyl alcohol) fibers (PVA fibers), methyl methacrylate, and water that fibers oxidized with hydrogen peroxide or sodium hypochlorite or ferric ion-adsorbing fibers effectively initiate the reaction as in the case of cellulose fibers. The initiation reaction of the sample was markedly activated by the presence of a pretreatment with an oxidizing agent at a concentration on the order of 10?9 mole/l., but the oxidized samples were sharply deactivated by such means as reduction and oximation. Since model initiators for the present system, such as iron(III) acetylacetonate-adsorbing PVA fibers or ferric ion-adsorbing fibres, with the co-existence of acetylacetone were observed to initiate graft copolymerization effectively, it is believed that carbonyl groups in modified PVA fibers could be a major factor for initiating copolymerization through interaction with metallic ions.  相似文献

Anilinepropylsilica xerogel was obtained by using an appropriate organosilane and tetraethyl orthosilicate as precursor reagents. The gelation was carried out using HF and NaF as catalysts. The presence of Na+ (when NaF was used) resulted in a decrease in the final organic content of the materials. This effect was interpreted as an inhibition of the organosilane polycondensation possibly due to the Na+ interaction with the SiO- groups of the hydrolyzed organosilane. The presence of Na+ also results in morphological changes in the xerogels.  相似文献

Graft copolymerization of methyl methacrylate on cellulosic materials of various carbonyl and aldehyde contents with the use of a ceric salt as an initiator was studied. It was found that the concentration of the ceric salt which gives the maximum per cent grafting is in good agreement with the equivalent of total carbonyl content in the cellulosic material, and the number of grafted chains in copolymers is roughly proportional to it. However, the molar ratio of the number of grafted chains to total carbonyl content is quite small, being approximately 1:50, and the graft copolymerization can be explained kinetically on the assumption that the number of radicals produced on cellulose by the ceric salt leading to branching is very much smaller than the number of radicals destroyed by the ceric salt, and growing radicals can be stabilized by the termination reaction with the ceric salt or with a cellulose radical. Although both aldehyde and carbonyl groups contribute to the formation of grafted chains, the former are effective mainly at low concentrations of the ceric salt; both groups participate in the production of graft copolymers showing the maximum per cent grafting.  相似文献

High-resolution solid-state NMR (SSNMR) of paramagnetic systems has been largely unexplored because of various technical difficulties due to large hyperfine shifts, which have limited the success of previous studies through depressed sensitivity/resolution and lack of suitable assignment methods. Our group recently introduced an approach using "very fast" magic angle spinning (VFMAS) for SSNMR of paramagnetic systems, which opened an avenue toward routine analyses of small paramagnetic systems by (13)C and (1)H SSNMR [Y. Ishii et al., J. Am. Chem. Soc. 125, 3438 (2003); N. P. Wickramasinghe et al., ibid. 127, 5796 (2005)]. In this review, we discuss our recent progress in establishing this approach, which offers solutions to a series of problems associated with large hyperfine shifts. First, we demonstrate that MAS at a spinning speed of 20 kHz or higher greatly improves sensitivity and resolution in both (1)H and (13)C SSNMR for paramagnetic systems such as Cu(II)(DL-alanine)(2)H(2)O (Cu(DL-Ala)(2)) and Mn(acac)(3), for which the spectral dispersions due to (1)H hyperfine shifts reach 200 and 700 ppm, respectively. Then, we introduce polarization transfer methods from (1)H spins to (13)C spins with high-power cross polarization and dipolar insensitive nuclei enhanced by polarization transfer (INEPT) in order to attain further sensitivity enhancement and to correlate (1)H and (13)C spins in two-dimensional (2D) SSNMR for the paramagnetic systems. Comparison of (13)C VFMAS SSNMR spectra with (13)C solution NMR spectra revealed superior sensitivity in SSNMR for Cu(DL-Ala)(2), Cu(Gly)(2), and V(acac)(3). We discuss signal assignment methods using one-dimensional (1D) (13)C SSNMR (13)C-(1)H rotational echo double resonance (REDOR) and dipolar INEPT methods and 2D (13)C(1)H correlation SSNMR under VFMAS, which yield reliable assignments of (1)H and (13)C resonances for Cu(Ala-Thr). Based on the excellent sensitivity/resolution and signal assignments attained in the VFMAS approach, we discuss methods of elucidating multiple distance constraints in unlabeled paramagnetic systems by combing simple measurements of (13)C T(1) values and anisotropic hyperfine shifts. Comparison of experimental (13)C hyperfine shifts and ab initio calculated shifts for alpha- and beta-forms of Cu(8-quinolinol)(2) demonstrates that (13)C hyperfine shifts are parameters exceptionally sensitive to small structural difference between the two polymorphs. Finally, we discuss sensitivity enhancement with paramagnetic ion doping in (13)C SSNMR of nonparamagnetic proteins in microcrystals. Fast recycling with exceptionally short recycle delays matched to short (1)H T(1) of approximately 60 ms in the presence of Cu(II) doping accelerated 1D (13)C SSNMR for ubiquitin and lysozyme by a factor of 7.3-8.4 under fast MAS at a spinning speed of 40 kHz. It is likely that the VFMAS approach and use of paramagnetic interactions are applicable to a variety of paramagnetic systems and nonparamagnetic biomolecules.  相似文献

An intramolecular conjugate addition of γ- and δ-trichloroacetimidoyloxy-α,β-unsaturated esters, a new way to construct 1,2-amino or 1,3-amino alcohol moieties in an acyclic system, is described. Very concise synthesis of d-vancosamine and 3-epi-d-vancosamine derivatives was also achieved utilizing this methodology.  相似文献

A new effective copper catalyzed CO coupling reaction using excess amount of strongly coordinating monodentate ligands was successfully developed. Among the DMAP-type monodentate ligands, 4-pyrrolidinopyridine afforded the best results. The developed reaction is widely applicable for the synthesis of various hindered or acyclic secondary alkyl-aryl ethers. In this study, a novel and remarkable acceleration of the coupling reaction using excess amount of monodentate ligands was discovered.  相似文献
