Aristolochic acids (AAs) are a mixture of structural-related compounds, in which aristolochic acid I (AA I) and aristolochic acid II (AA II) are reported to be correlated with Aristolochic acid nephropathy (AAN). In this work, a rapid and sensitive ultra-high-pressure liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was developed to determine AA I and AA II in herbal products and biological fluids. By using gradient elution with a mobile phase composed of a mixture of 10 mM ammonium formate buffer (pH 3.0) and acetonitrile, AAs could be determined within 10 min. Under optimum UHPLC-MS/MS conditions, the limit of detections was 0.14 and 0.26 ng mL−1 for AA I and AA II, respectively. Run-to-run repeatability and intermediate precision of peak area for AA I and AA II were less than 5.74% relative standard deviation (RSD). Accuracy was tested by spiking 10, 100 and 1000 ng mL−1 in rat serum and the recoveries were within 76.5-92.9%. Matrix effects were within 78.8-127.7%. The developed method was successfully applied to determine AA I and AA II in several herbal products and to investigate their pharmacokinetic behavior in female Wister rats. The result shows that the developed UHPLC-MS/MS method is efficient, sensitive, and accurate for the determination of AA I and AA II in herbal products and biological samples.  相似文献

A series of transition metal coordination polymers [Co(H3L)2(4,4′- bpy)(H2O)2]n?n(4,4′-bpy) (1), [Ni(H2L)(4,4′-bpy)(H2O)2]n (2), [Co2(L)(phen)2(H2O)4]n?(H2O)2n (3), and [Ni2(L)(phen)2(H2O)4]n?(H2O)2n (4) have been assembled from a semirigid multicarboxylate ligand 3,3′-(1,4-phenylenebis(oxy))diphthalic acid (H4L) with the help of 4,4′-bipyridine (4,4′-bpy) ligand or 1,10-phenanthroline (phen) ligand. X-ray single crystal diffraction analysis reveals that complex 1 crystallizes in the space group of P − 1 and displays a one-dimensional (1D) chain structure constructed from 4,4′-bpy ligand and H3L ligand, which was further interlinked to form a three-dimensional network via hydrogen bonds. In complex 2, Ni(II) atoms are coordinated by L ligand in monodentate fashion to form alternate left- and right-helices, which are further bridged together by the coordination interactions between Ni(II) atoms and 4,4′-bpy, leading to a 2-fold (4, 4)-connected interpenetrating network. Isostructural complexes 3 and 4 belong to the space group P − 1 and display a 1D chain structure constructed from phen and L ligands, which was further interlinked to form a 2D plane via π–π interactions. In addition, their thermal and luminescent properties were also investigated.  相似文献

A deep geologic disposal is the prime option for the long-term isolation of high-level radioactive waste (HLW) in many countries. For deeply located repositories, a radionuclide released from a failed waste container moves through the engineered and natural barriers before it reaches a biosphere. The pseudo-colloid which a radionuclide is adsorbed on a moving natural humic or fulvic colloid can be generated in a fractured porous medium. The size of a colloid is in general in the order of a hundred nanometer so that its migration velocity in a fracture is higher than that of a radionuclide due to the hydrochromatic effect. A large colloid cannot diffuse easily into a surrounding rock. Also, there are many kinds of actinides and these actinides have decay chains. In this analysis, the canonical form solution is derived for a pseudo-colloid and a solute in a fracture and a surrounding rock with a realistic inlet boundary condition with multi-member decay chains. It is used the Fortran based a computational code which uses a special subroutine for the inversion of Laplace transform. Consequently, the role of the pseudo-colloid in the fractured porous medium is important and also their decay chains aren’t neglected in the performance assessment of the HLW.  相似文献

Three monomeric germatranes, 1-isopropoxy-3,3,7,7,10,10-hexamethyl-2,8,9-trioxa-5-aza-1-germatricyclo[3.3.3.01,5]undecane (1), 1-isopropoxy-3,3,7,7-tetramethyl-2,8,9-trioxa-5-aza-1-germatricyclo[3.3.3.01,5]undecane (2), and 1-isopropoxy-3,3-dimethyl-2,8,9-trioxa-5-aza-1-germatricyclo[3.3.3.01,5]undecane (3) have been synthesized by the reaction of Ge(O-i-Pr)4 in refluxing toluene with corresponding triethanolamines, (HOCH2CH2)nN(CH2CMe2OH)3−n (n = 0, L1H3; n = 1, L2H3; n = 2, L3H3), where the number of CMe2 groups adjacent to a OH functionality varied from 3 (L1H3) to 2 (L2H3), and to 1 (L3H3). These germatranes 1-3 have been characterized by solution 1H and 13C{1H} NMR and the solid state structure of 2 has been determined by single crystal X-ray diffraction.  相似文献

We present a microfluidic device generating three-dimensional (3D) coaxial flow by the addition of a simple hillock to produce an alginate core-shell microcapsule for the efficient formation of a cell spheroid. A hillock tapered at downstream of the two-dimensional focusing channel enables outside flow to enclose the core flow. The aqueous solution in the core flow was focused and surrounded by 1.8% alginate solution to be solidified as a shell. The double-layered coaxial flow (aqueous phase) was broken up into a droplet by the shear flow of oleic acid (oil phase) containing calcium chloride for the polymerization of the alginate shell. The droplet generated from the laminar coaxial flow maintained a double-layer structure and gelation of the alginate solution made a core-shell microcapsule. The shell-thickness of the microcapsule was adjusted from 8-21 μm by the variation of two aqueous flow rates. The inner shape of the shell was almost spherical when the ratio of the water-glycol mixture in the core flow exceeded 20%. The microcapsule was used to form a spheroid of embryonic carcinoma cells (embryoid body; EB) by injecting a cell suspension into the core flow. The cells inside the microcapsule aggregated into an EB within 2 days and the EB formation rate was more than 80% with strong compaction. The microcapsule formed single spherical EBs without small satellite clusters or a bumpy shape as observed in solid microbeads. The microfluidic chip for encapsulation of cells could generate a number of EBs with high rate of EB formation when compared with the conventional hanging drop method. The core-shell microcapsule generated by 3D focusing in the microchannel was effective in forming large number of spherical cell clusters and the encapsulation of cells in the microcapsule is expected to be useful in the transplantation of islet cells or cancer stem cell enrichment.  相似文献

Single nucleotide polymorphisms (SNPs) can serve as important biomarkers for genetic diseases, for which accurate detection of SNPs is essential for early diagnosis. We have developed a novel SNP sensor by combining a Au nanowire-on-film surface-enhanced Raman scattering (SERS) platform with S1 nuclease reaction. The combined sensor system provides reproducible SERS signals only in the presence of perfectly matched target DNAs, to probe DNAs as a result of single-stranded DNA-specific degradation by S1 nuclease. Furthermore, point mutations in DNA causing Wilson disease and Avellino corneal dystrophy were successfully identified by this sensor, thereby indicating its practical ability to diagnose genetic diseases.  相似文献

The gas‐phase free radical initiated peptide sequencing (FRIPS) fragmentation behavior of o‐TEMPO‐Bz‐conjugated peptides with an intra‐ and intermolecular disulfide bond was investigated using MSn tandem mass spectrometry experiments. Investigated peptides included four peptides with an intramolecular cyclic disulfide bond, Bactenecin (RLC RIVVIRVC R), TGF‐α (C HSGYVGVRC ), MCH (DFDMLRC MLGRVFRPC WQY) and Adrenomedullin (16–31) (C RFGTC TVQKLAHQIY), and two peptides with an intermolecular disulfide bond. Collisional activation of the benzyl radical conjugated peptide cation, which was generated through the release of a TEMPO radical from o‐TEMPO‐Bz‐conjugated peptides upon initial collisional activation, produced a large number of peptide backbone fragments in which the S? S or C? S bond was readily cleaved. The observed peptide backbone fragments included a‐, c‐, x‐ or z‐types, which indicates that the radical‐driven peptide fragmentation mechanism plays an important role in TEMPO‐FRIPS mass spectrometry. FRIPS application of the linearly linked disulfide peptides further showed that the S? S or C? S bond was selectively and preferentially cleaved, followed by peptide backbone dissociations. In the FRIPS mass spectra, the loss of ?SH or ?SSH was also abundantly found. On the basis of these findings, FRIPS fragmentation pathways for peptides with a disulfide bond are proposed. For the cleavage of the S? S bond, the abstraction of a hydrogen atom at Cβ by the benzyl radical is proposed to be the initial radical abstraction/transfer reaction. On the other hand, H‐abstraction at Cα is suggested to lead to C? S bond cleavage, which yields [ion ± S] fragments or the loss of ?SH or ?SSH. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献
