In order to develop an analytical method for the discrimination of dextromethorphan (an antitussive medicine) from its enantiomer, levomethorphan (a narcotic) in biological samples, chiral analyses of these drugs and their O-demethyl and/or N-demethyl metabolites in rat plasma, urine, and hair were carried out using LC-MS/MS. After the i.p. administration of dextromethorphan or levomethorphan to pigmented hairy male DA rats (5 mg/kg/day, 10 days), the parent compounds and their three metabolites in plasma, urine and hair were determined using LC-MS/MS. Complete chiral separation was achieved in 12 min on a Chiral CD-Ph column in 0.1% formic acid–acetonitrile by a linear gradient program. Most of the metabolites were detected as being the corresponding O-demethyl and N, O-didemethyl metabolites in the rat plasma and urine after the hydrolysis of O-glucuronides, although obvious differences in the amounts of these metabolites were found between the dextro and levo forms. No racemation was observed through O- and/or N-demethylation. In the rat hair samples collected 4 weeks after the first administration, those differences were more clearly detected and the concentrations of the parent compounds, their O-demethyl, N-demethyl, and N, O-didemethyl metabolites were 63.4, 2.7, 25.1, and 0.7 ng/mg for the dextro forms and 24.5, 24.6, 2.6, and 0.5 ng/mg for the levo forms, respectively. In order to fully investigate the differences of their metabolic properties between dextromethorphan and levomethorphan, DA rat and human liver microsomes were studied. The results suggested that there might be an enantioselective metabolism of levomethorphan, especially with regard to the O-demethylation, not only in DA rat but human liver microsomes as well. The proposed chiral analyses might be applied to human samples and could be useful for discriminating dextromethorphan use from levomethorphan use in the field of forensic toxicology, although further studies should be carried out using authentic human samples.  相似文献

Exciton-polariton transmission in quantum dot waveguides is investigated with quantum time-evolution equations in Liouville space for exciton wave packet dynamics. The transmission efficiency of the exciton-polariton wave with the longitudinal and transverse mode transformations strongly depends on the geometric parameters (bending angle and interdot distance) of the waveguides and on configuration of an additional branch attached to the waveguide. We have numerically demonstrated that the transmission efficiency significantly improves by controlling these geometric parameters and the configuration of the branched waveguide. The optimal bending angle for efficient transmission with the longitudinal and transverse mode transformations deviates from the right angle owing to more than nearest-neighbor-site interactions through a shortcut. We have also found that existence of thermal relaxation enables to open a new transmission channel along which the exciton-polariton transmission through the Coulomb interaction is suppressed.  相似文献

This paper presents an efficient and exact formulation for finding stress concentration factors using a zooming analysis. A favorable characteristics of this method is its ability to perform a zooming analysis with computations taking place only in the zooming area under consideration with no need to treat the region outside the zooming area. At the final zooming step, the flexibility of a limited zooming area is determined for all nodes, including the new nodes created in the zooming processes as well as those of the original system before zooming.  相似文献

The effect of pressure on the melting point and volume of fusion of polyethylene was studied by high-pressure dilatometry. Starting materials were crystallized slowly from the melt under pressures of 1500, 3500, 5130 kg/cm2, and 1 atm. It has been shown that the unusual behavior observed at pressures above 4000 kg/cm2 is due to crystallization and melting of two kinds of extended-chain crystals differing in thermal stability. These are designated as ordinary extended-chain and highly extended-chain crystals, respectively. The relation between pressure P and melting temperature Tm of folded-chain, ordinary extended-chain, and highly extended-chain polyethylene was determined precisely. At pressures up to about 3000 kg/cm2, plots of P against Tm for the crystal forms have almost the same curvature and then become parallel. But at pressures above 4000 kg/cm2, ordinary extended-chain crystals show a linear increase of Tm with a constant slope of about 70 atm/deg. Curve for the highly extended-chain crystals changes in slope from 70 to 50 atm/deg at pressures between 3500 and 4300 kg/cm2, and then show a sharp increase of Tm with increasing pressure. Experiments show that the meltingpoint curve of the highly extended-chain crystals overlaps that of the ordinary extended-chain crystals at pressures below 4000 kg/cm2. Annealing experiments with folded-chain and ordinary extended-chain crystals have been made under high pressure. It is suggested that the formation of highly extended-chain crystals occurs stepwise through the formation and reorganization of ordinary extended-chain crystals from the original folded-chain crystals by a mechanism of partial melting and recrystallization at pressures above 4000 kg/cm2.  相似文献

A search for K+ → π+γ, π+γγ, π+γγγ was made detecting pions from stopped kaons in the kinetic energy region between 117 MeV and 127 MeV. New Limits of 1.4 ×10?6, 8.4 × 10?6 and 1.0 × 10?4 for the branching ratio were obtained assuming a phase-space spectrum on the pion energy. The first limit also aplies to another process K+ → π+ + a, where a is a light meson with mass smaller than 100 MeV/c2 and decays into nγ's with lifetime less than 10?9 s.  相似文献

A phosphorus-containing hybrid porphyrin was successfully prepared via the BF3-promoted dehydrative condensation between sigma4-phosphatripyrrane and 2,5-bis[hydroxy(phenyl)methyl]thiophene. The NMR and UV-vis absorption spectra, electrochemical measurements, and DFT calculations revealed that the sigma3-P,N2,S-hybrid porphyrin exhibits high aromaticity as an 18pi-electron system in terms of both geometric and magnetic criteria. A fast method for enclosing all eigenvalues in generalized eigenvalue problems Ax=λBx is proposed. Firstly a theorem for enclosing all eigenvalues, which is applicable even if A is not Hermitian and/or B is not Hermitian positive definite, is presented. Next a theorem for accelerating the enclosure is presented. The proposed method is established based on these theorems. Numerical examples show the performance and property of the proposed method. As an application of the proposed method, an efficient method for enclosing all eigenvalues in polynomial eigenvalue problems is also sketched.  相似文献
