Circulating tumor cells (CTCs) have gained increasing attention as physicians and scientists learn more about the role these extraordinarily rare cells play in metastatic cancer. In developing CTC technology, the critical criteria are high recovery rates and high purity. Current isolation methods suffer from an inherent trade-off between these two goals. Moreover, ensuring minimal cell stress and robust reproducibility is also important for the clinical application of CTCs. In this paper, we introduce a novel CTC isolation technology using selective size amplification (SSA) for target cells and a multi-obstacle architecture (MOA) filter to overcome this trade-off, improving both recovery rate and purity. We also demonstrate SSA-MOA's advantages in minimizing cell deformation during filter transit, resulting in more stable and robust CTC isolation. In this technique, polymer microbeads conjugated with anti-epithelial cell adhesion molecules (anti-EpCAM) were used to selectively size-amplify MCF-7 breast cancer cells, definitively differentiating from the white blood cells (WBCs) by avoiding the size overlap that compromises other size selection methods. 3 μm was determined to be the optimal microbead diameter, not only for size discrimination but also in maximizing CTC surface coverage. A multi-obstacle architecture filter was fabricated using silicon-on-glass (SOG) technology-a first such application of this fabrication technique-to create a precise microfilter structure with a high aspect ratio. The filter was designed to minimize cell deformation as simulation results predicted that cells captured via this MOA filter would experience 22% less moving force than with a single-obstacle architecture. This was verified by experiments, as we observed reliable cell capture and reduced cell deformation, with a 92% average recovery rate and 351 peripheral blood leukocytes (PBL) per millilitre (average). We expect the SSA-MOA platform to optimize CTC recovery rates, purity, and stability, increasing the sensitivity and reliability of such tests, thereby potentially expanding the utilization of CTC technologies in the clinic.  相似文献

HKUST-1, a metal-organic framework (MOF) material containing Cu(II)-paddlewheel-type nodes and 1,3,5-benzenetricarboxylate struts, features accessible Cu(II) sites to which solvent or other desired molecules can be intentionally coordinated. As part of a broader investigation of ionic conductivity in MOFs, we unexpectedly observed substantial proton conductivity with the "as synthesized" version of this material following sorption of methanol. Although HKUST-1 is neutral, coordinated water molecules are rendered sufficiently acidic by Cu(II) to contribute protons to pore-filling methanol molecules and thereby enhance the alternating-current conductivity. At ambient temperature, the chemical identities of the node-coordinated and pore-filling molecules can be independently varied, thus enabling the proton conductivity to be reversibly modulated. The proton conductivity of HKUST-1 was observed to increase by ~75-fold, for example, when node-coordinated acetonitrile molecules were replaced by water molecules. In contrast, the conductivity became almost immeasurably small when methanol was replaced by hexane as the pore-filling solvent.  相似文献

The bactericidal activity of TiO2 nanoparticles under visible light is very important in regards to its practical applications. In this paper, we synthesized vanadium-pentoxide-loaded TiO2 nanoparticles (V2O5?CTiO2) using a chemical vapor condensation method, followed by the impregnation method, and characterized its physicochemical properties through X-ray diffraction patterning, X-ray photoelectron spectroscopy analysis, Raman spectra analysis, and Fourier transform infrared analysis. In addition, the antibacterial activity of V2O5?CTiO2 nanoparticles against E. coli was evaluated and compared with pure TiO2 nanoparticles. In these experiments, the population of E. coli was shown to be significantly reduced by V2O5?CTiO2 nanoparticles under illumination with fluorescent light, whereas pure TiO2 nanoparticles showed about 3.3-fold lower antibacterial activity than the V2O5?CTiO2 nanoparticles. This result was most likely due to the change in surface conditions of the TiO2 nanoparticles, which was due to the loading of vanadium pentoxide on the TiO2 nanoparticles. Furthermore, both photocatalysts showed similar antibacterial activity under UV-A (352?nm) irradiation.  相似文献

This study was performed to demonstrate the importance of selecting an appropriate membrane when developing immunochromatographic assays (ICAs) for the sensitive detection of low-molecular weight compounds. Based on our findings, we propose a theoretical basis for selecting such a membrane. When eluting the sample solution for the competitive ICA using colloidal gold label for low-molecular analytes, the degree of binding inhibition is proportional to the collision frequency between the antibody-colloidal gold (Ab–CG) and analyte before Ab–CG binding to the capture antigen and a higher concentration of pesticides around the Ab–CG leads to a greater degree of inhibition. Therefore, we propose that the relative migration speed of the analyte and Ab–CG on the test strip is critically important for selecting a membrane in the development of sensitive competitive ICAs. We developed a novel method to estimate such a relative migration speed. We demonstrated the applicability of this proposal by using it to select an appropriate membrane for the development of an ICA of the pesticide diazinon.  相似文献

Negative-ion photoelectron spectroscopy and ab initio simulations are used to study the variation in magnetic structure in Mn(x)O(y) (x = 3, 4[semicolon] y = 1, 2) clusters. The ferrimagnetic and antiferromagnetic ground-state structures of Mn(x)O(y) are 0.16-1.20 eV lower in energy than their ferromagnetic isomers. The presence of oxygen thus stabilizes low-spin isomers relative to the preferred high-spin ordering of bare Mn(3) and Mn(4). Each cluster has a preferred overall magnetic moment, and no evidence is seen of competing states with different spin multiplicities. However, non-degenerate isomags, which possess the same spin multiplicity but different arrangements of local moments, do contribute additional features and peak broadening in the photoelectron spectra. Proper accounting for all possible isomags is shown to be critical for accurate computational prediction of the spectra.  相似文献

We study both classical and quantum relation between two Hamiltoniansystems which are mutually connected by time-dependent canonical transformation. One is ordinary conservative system and the other istime-dependent Hamiltonian system. The quantum unitary operatorrelevant to classical canonical transformation between the two systems are obtained through rigorous evaluation. With the aid of the unitary operator, we have derived quantum states of the time-dependent Hamiltonian system through transforming the quantum states of the conservative system. The invariant operators of the two systems are presented and the relation between them are addressed. We showed that there exist numerous Hamiltonians, which gives the same classical equation of motion. Though it is impossible to distinguish the systems described by these Hamiltonians within the realm of classical mechanics, they can be distinguishable quantum mechanically.  相似文献

The copolymerization of ethylene and norbornene (N) was carried out with iPr(Cp)(Flu)ZrCl2 catalyst and modified methylaluminoxane (MMAO) cocatalyst. The catalytic activity was dependent on the structure of MMAO, i.e., MMAO-4 exhibited higher catalyst activity than MMAO-3A containing more i-butyl groups. The glass transition temperature (Tg) and the composition of the produced copolymer were not affected by MMAO type. With styrene derivatives as 3rd monomer, Tg of copolymer increased while the catalytic activity decreased. With the addition of 3rd monomer, not only the content of 3rd monomer but also the content of N increased.  相似文献

Summary: N,N‐Diphenylacrylamide was polymerized in a living fashion with triisobutylaluminum in THF at 0 °C. The polymerization results showed an increase of molecular weight proportional to the amount of monomer consumed and a first‐order kinetics at −78 °C. The intermediates obtained with excess initiator at −78 °C revealed that the polymerization was initiated through 1,4‐addition of hydride from a triisobutyl group in the triisobutylaluminum and then proceeded through aluminum‐oxygen bond interchange.
