研究了在反相高效液相色谱中的烷基醇-甲酸-水、烷基醇-三氟醋酸-水和烷基醇-磷酸盐-水三种流动相,9种生物大分子的分离体系中Z的表征。发现当蛋白质完全变性时,Z值与其分子量呈正比,而与置换剂分子大小呈反比。Z值会因流动相中离子对试剂的存在而减小,且浓度愈大,减小程度愈甚。对于在异丙醇-水溶液中的不同种类离子对试剂而言,对蛋白构象变性影响的减小顺序为44%(V/V)甲酸>0.05mol/L磷酸盐缓冲液(pH<2.5)>0.1%(V/V)三氟醋酸。  相似文献

The dye Brilliant Green (BG) is shown to be an excellent fluorogenic probe for the detection of double-stranded DNA (ds-DNA). Typically, the detection limit of a 21-mer is as low as 6 nM ((3σ), and the method can clearly distinguish between fully complementary ds-DNA and a ds-DNA with a single mismatch. Labeling is not required, hybridization and detection occur in homogenous aqueous solution in short time, and the dye is easily accessible, commercially available, and affordable.  相似文献

间充质干细胞(MSCs)具有高度自我更新能力、多分化潜能、体外易分离和培养的特性,是细胞治疗和组织工程重要的种子细胞来源,但如何大规模地获得具有可再生活性的MSCs一直是限制其临床应用的关键因素,近几年发展起来的贴壁动物细胞动态培养技术为MSCs的大规模体外扩增提供了一条重要的途径。本综述结合动物细胞扩增载体的发展现状,主要介绍了用于间充质干细胞三维动态培养的明胶载体、海藻酸盐载体、壳聚糖载体和其他多糖载体等常规载体及其表面修饰和改性方法,并进一步介绍了以非酶解途径回收扩 增细胞的新型干细胞载体的研究进展。随着新型载体材料的涌现以及人们对干细胞生长和扩增特点的了解,采用三维动态培养技术安全而有效地大规模体外扩增MSCs的必要性将得到进一步的确认。  相似文献

微电子器件的发展趋势是小型化和多功能化,这就对半导体材料的加工技术提出了更高的要求。与传统的加工技术相比,近年发展起来的贵金属粒子辅助化学刻蚀半导体材料制备微结构技术因操作简单、不需要精密设备、反应迅速和可批量生产等优点引起了国内外学者的广泛关注。本文以Si为主,详细介绍了Ag辅助化学刻蚀半导体材料的机理、反应现象及影响因素,总结了各种微结构的制备技术及其应用。此外,对Ge,Si1-xGex和GaAs等其他半导体材料的贵金属粒子辅助化学刻蚀技术也进行了综述。同时分析了贵金属粒子辅助化学刻蚀半导体目前存在的问题,并对未来的研究方向进行了展望。  相似文献

Three new complexes of the formula M(2)L(2) derived from 2-(4-quinolyl)nitronyl nitroxide (4-QNNN) and M(hfac)(2) [M = Mn(II), Co(II), and Cu(II)], (4-QNNN)(2).[Mn(hfac)(2)](2) (1), (4-QNNN)(2).[Co(hfac)(2)](2).2H(2)O (2), and (4-QNNN)(2).Cu(hfac)(2).Cu'(hfac)(2) (3), were synthesized and characterized structurally as well as magnetically. Complexes 1 and 2 are four-spin complexes with quadrangle geometry, in which both the nitrogen atoms of quinoline rings and oxygen atoms of nitronyl nitroxides are involved in the formation of coordination bonds. For complex 3, however, the nitrogen atoms of quinoline rings are coordinated with Cu(II) ion to afford a three-spin complex, which is further linked to another molecule of Cu(hfac)(2) (referred to as Cu'(hfac)(2)) to form a 1D alternating chain. The magnetic behaviors of the three complexes were investigated. For complex 1, as the nitronyl nitroxides and Mn(II) ions are strongly antiferromagnetically coupled, consequently its temperature dependence of magnetic susceptibility was fitted to the model of spin-dimer with S = 2, yielding the intradimer magnetic exchange constant of J = -0.82 cm(-1). For complex 2, the temperature dependence of the magnetic susceptibility in the T > 50 K region was simulated with the model of two-spin unit with S(1) = 3/2 and S(2) = 1/2, leading to J = -321.9 cm(-1) for the magnetic interaction due to Co(II).O coordination bonding, D = -16.3 cm(-1) (the zero-field splitting parameter), g = 2.26, and zJ = -3.8 cm(-1) for the magnetic interactions between Co(II) ions and nitronyl nitroxides through quinoline rings and those between nitronyl nitroxides due to the short O.O short contacts. The temperature dependence of magnetic susceptibility of 3 was approximately fitted to a model described previously affording J(1) = -6.52 cm(-1) and J(2) = 3.64 cm(-1) for the magnetic interaction between nitronyl nitroxides and Cu(II) ions through the quinoline unit via spin polarization mechanism and the weak O.Cu coordination bonding, respectively.  相似文献

The V2O5/SiO2 and V2O5-P2O5/SiO2 catalysts were prepared by the Sol-gel method. The surface composition,structure,chemisorption and reactivity of the catalysts were systematically studied by using the techniques of XRD,TPR,IR,TPD and icro-reactor. The results showed that the active component was dispersed uniformly on the surface of amorphous SiO2 . There existed Lewis base sites(V=O and V-O-V)and Lewis acid sites (Vn+)on the surface of these catalysts. An addition of PO43- could lower the activity of surface terminal oxygen of Lewis base sites V=O. Iso-butane could chemisorbed on surface terminal oxygen of Lewis base sites V=O by one or two H atoms in -CH3 to form molecular adsorption states. Reaction products of i-C4H10 on the V-P-O/SiO2 catalyst were mainly i-C4H8,MAL,MAA and COx . Conversion of i-C4H10 was 5% and selectivity of partial oxidation products is above 72% at 300℃. The addition of PO43- could improve the selectivity of partial oxidation products on the VSiO catalyst because PO43- could reduce the activity of lattice oxygen in V = O bond,and weaken the adsorption intensity of i-C4H10 .  相似文献

Alternaria sp. ND-16, a bacterium isolated from soil sample, was identified as a strain of Alternaria mali based on the morphology and comparison of internal transcribed spacer rDNA gene sequence studies. Furthermore, it is demonstrated that this strain has xylanase activity, and the activity can be optimized under suitable growing conditions where wheat bran and urea are the primary sources of carbon and nitrogen. Partially purified xylanase from Alternaria sp. ND-16 is shown to have an optimal pH of 6.0 and optimal temperature of 50 °C, making this enzyme potentially suitable for industrial applications. It is also demonstrated that Na+ and Mn2+ show strong inhibition of the xylanase while K+, Li+, Fe2+, Cu2+, and Zn2+ have no significant effect on the activity.  相似文献

Both calorimetric determination of displacement adsorption enthalpies ΔH and measurement of adsorbed amounts of lysozyme (Lyz) denatured by 1.8 mol L−1 guanidine hydrochloride (GuHCl) on a moderately hydrophobic packings at 298 K, pH 7.0 and various salt concentrations were carried out. Based on the thermodynamics of stoichiometric displacement theory (SDT) the fractions of thermodynamic functions, which related to four subprocesses of denatured protein refolding on the surface, were calculated and thermodynamic analysis that which one of the subprocesses plays major role for contribution to the thermodynamic fractions was made in detail. The moderately hydrophobic surface can provide denatured Lyz energy and make it gain more conformation with surface coverage or salt concentration increment. The displacement adsorptions of denatured Lyz onto PEG-600 surface are exothermic, more structure-ordered and enthalpy driven processes.  相似文献
