A novel liquid chromatography-tandem mass spectrometry (LC-MS/MS) method has been established to quantify metabolic intermediates, including lactate (Lac), pyruvate (Pyr), acetoacetate (ACAC) and 3-hydroxybutyrate (3-HB) in blood. Samples were deproteinized with methanol-acetonitrile solution, and analytes were separated on an adamantyl group-bonded reverse phase column and detected in multiple reaction monitoring mode. Total analysis time was 4 min per sample. Method validation results displayed that limits of quantification were 10.0 μmol L−1 for Lac and Pyr, and 5.0 μmol L−1for ACAC and 3-HB. The within- and between-run coefficients of variation were in the range of 1.2–6.4% for all analytes. The recoveries were ranged from 95.6 to 111.5%. The reference values of analytes were determined for the pediatric population. Duo to instability of Lac, Pyr and ACAC in vitro, a comprehensive stability assay was performed to determine optimal conditions for sample collection, pretreatment and storage. Results showed that precipitation of protein in blood at bedside combined with low storage temperature could effectively preserve the integrity of Lac, Pyr and 3-HB, but the precipitated protein accelerated degradation of ACAC. Isolation of supernatant fluid slowed degradation of ACAC. Supernatant samples could store at −20 °C for 10 days. The use of plasma or serum to determine these intermediates was not recommended. In this study, 450 samples from patients were analyzed, and 7 patients were diagnosed as congenital lactic acidosis. With the advantages of rapid, accurate and reliable, this method is very suitable for congenital lactic acidosis screening and researches related to energy metabolism.  相似文献

The prevention of postprandial hyperglycemia and diabetic complications is crucial for diabetes management. Inhibition of α-glucosidase to slow carbohydrate metabolism is a strategy to alleviate postprandial hyperglycemia. In addition, suppression of non-enzymatic glycation can diminish the advanced glycation end products and reduce the oxidative stress and inflammation, thereby preventing the diabetic complications. In this study, an anti-oxidative proteoglycan (named FYGL) extracted from Ganoderma lucidum was investigated in vitro for its inhibitory effect on α-glucosidase and non-enzymatic glycation using molecular kinetics, intrinsic fluorescence assay, and bovine serum albumin glycation models. The molecular kinetics and fluorescence assay revealed that FYGL decreases α-glucosidase activity by forming a FYGL–α-glucosidase complex. To evaluate the anti-glycation effect, fructose-glycated and methylglyoxal-glycated BSA models were analyzed by spectroscopic and SDS-PAGE methods. Results showed that FYGL inhibited the glycation at every stage and suppressed glycoxidation, possibly due to its anti-oxidative capacity and FYGL–BSA complex formation. Furthermore, we demonstrated in vivo that FYGL could alleviate postprandial hyperglycemia in db/db mice as well as AGE accumulation and vascular injury in diabetic rats. Overall, FYGL possesses anti-postprandial hyperglycemia and anti-glycation functions and would be potentially used in clinic for diabetes and related complication management.  相似文献

Nine a-halo-m-trifluoromethgl cinnamides were synthesized from trifluoromethyl benzene,via bromination,Grignard reaction,condensation,halogen addition reactions etc,as indicated in the synthetic scheme.Their configurations were confirmed by 1H NMR.Their pharmacological results showed that the introduction of a-halogen group do not enhance their anticonvulsant activity as expected.  相似文献

目的在糖尿病下肢血管病变患者的诊断过程中应用彩色多普勒超声(彩超)技术,探讨其诊断效果。方法回顾性分析东莞常安医院自2014年4月至2016年4月期间所收治的55例糖尿病下肢血管病变患者并将其作为实验组,另收治的55例健康志愿者作为对照组,给予两组患者彩色多普勒超声(彩超)技术诊断并对比诊断结果。结果实验组患者的检出率是85.45%;实验组患者的足背动脉血管内径为(1.44±0.33)mm,腘动脉血管内径为(4.99±0.63)mm,均显著低于对照组,P0.05,有统计学意义。结论在糖尿病下肢血管病变患者的诊断过程中应用彩色多普勒超声(彩超)技术检出率较高,可以为该疾病的有效预防以及早期诊断、早期治疗提供参考依据,值得临床应用。  相似文献

In this letter, strong asymmetric coupling of two parallel exclusion processes: effect of unequal injection rates will be investigated. It is a generalization of the work of Xiao et al. (Phys. Lett. A 8, 374 (2009)), in which the particles only move on two lanes with rate 1 toward right. We can obtain the diverse phase diagram and density profiles of the system. The vertical cluster mean-field approach and extensively Monte Carlo simulations are used to study the system, and theoretical predictions are in excellent agreement with simulation results.  相似文献

分别以亚硫酰氯(SOCl2)、硫酰氯(SO2Cl2)、二甲亚砜(DMSO)、苯磺酰氯(BSC)和苯基乙烯基亚砜(PVSO)作为添加剂, 加入至1 mol•L-1 LiPF6/碳酸乙烯酯(EC)+碳酸二甲酯(DMC)(体积比为1∶1)电解液中, 采用恒电流沉积-溶出法、循环伏安法(CV)、电化学阻抗谱(EIS)、X射线光电子能谱(XPS)测试技术, 研究了添加剂对电化学金属锂沉积-溶出可逆性以及金属锂负极/电解液界面性质的影响. 电化学测试结果表明, 添加SO2Cl2和BSC能显著改善金属锂电极的循环性能, 100% DOD(锂全溶出)条件下, 循环效率稳定在90%左右. 电化学测试、添加剂分子电子亲和能计算并结合XPS测试结果表明, 较早反应成膜的SO2Cl2和BSC添加剂可以在一定程度上抑制电解液溶剂的分解, 使锂电极循环效率和寿命得以提高. 洞察和比较添加剂的结构揭示, 含磺酰基的化合物明显优于含亚磺酰基的化合物.  相似文献

A unique thermophilic microbial community developed initially from swine waste was investigated in this study. Cellulase activities were observed when this community was inoculated to media containing either cellulose or carboxymethylcellulose at 57 °C. Through constructing a clone library for the 16S ribosomal DNA, it was revealed that this community was mainly composed of three genera: Thermobacillus, Brevibacillus, and Anoxybacillus. New findings regarding the thermo- and pH stability of crude cellulases secreted by Brevibacillus sp. JXL were presented. Recent study on the growth characteristics of Anoxybacillus sp. 527 was discussed.  相似文献

建立了高效液相色谱-三重四极杆串联质谱(HPLC-MS/MS)测定焙烤食品及其塑料包装中31种邻苯二甲酸酯(PAEs)的方法。焙烤食品采用乙酸乙酯超声提取,提取液经冷冻(-18℃)、低温高速离心净化,剪碎后的塑料包装材料采用体积比为1:1:1的甲醇-丙酮-正己烷混合溶剂进行液液超声萃取后进入HPLC-MS/MS分析。采用MGⅢC18色谱柱(2.0 mm×100 mm,5 μm),选择反应监测(SRM)模式测定。实验结果表明,31种邻苯二甲酸酯的特征离子质量色谱峰的峰面积与其质量浓度在各自的质量浓度范围内线性关系良好(r2≥0.9958)。样品加标回收率除邻苯二甲酸二月桂酯(DLP)为70.9%~109.7%外,其余30种化合物为80.1%~113.0%,相对标准偏差(n=6)为1.0%~13.7%。31种PAEs的检出限在0.02~8.15 μg/kg之间,定量限在0.07~27.17 μg/kg之间。应用该方法检测了面包、饼干、糕点、馅料4大类焙烤食品及其塑料包装中31种邻苯二甲酸酯的含量。该方法具有操作简单、快速、准确度和精密度高等优点,满足日常检测的要求。  相似文献
