The reaction of 1-alkynylcyclobutanols with aryl iodides in the presence of Pd(OAc)2 and Et3N in acetonitrile at 80°C for 24 h gives 2-disubstituted methylenecyclopentan-1-ones in modest to good yields. The tandem insertion-ring expansion process proceeds via the formation of an alkynyl π-complex, followed by migration of a carbon-carbon bond of the tert-alkanol to form the cyclopentanones stereoselectively.  相似文献

A new very sensitive and selective chromogenic reagent, meso-tetra-(3,5-dibromo-4-hydroxylphenyl)porphyrin [T(DBHP)P], was synthesized and studied for the determination of trace lead in detail. In 0.10 mol l-1 NaOH medium, lead reacts with T(DBHP)P to form a 1:2 yellow complex, which gives a maximum absorption at 479 nm; 0-0.48 microgram ml-1 Pb(II) obeyed Beer's law. The molar absorptivity of the complex and Sandell's sensitivity are 2.5 x 10(5) 1 mol-1 cm-1 and 0.000812 microgram cm-2, respectively. Second-derivative spectrophotometry is better than conventional spectrophotometry in sensitivity and selectivity, and its limit of quantification, limit of detection and relative standard deviation are 0.70 ng ml-1, 0.21 ng ml-1 and 1.0%, respectively. Ca (3250-fold), Mg (2000-fold), Sr (1000-fold), Ba (750-fold), Al (1000-fold), Bi (500-fold), Fe (2000-fold), Co (750-fold), Ni (1000-fold), Cu (750-fold), Zn (1250-fold), Cd (2500-fold) and Ag (550-fold) do not interfere with the determination of lead. The chromogenic system is remarkably superior to other reagents, especially porphyrin compounds. The influence caused by oxygen in air or in solution can be easily eliminated by adding Na2SO3. The reaction is very stable, the stability constant of the complex being 1.2 x 10(45). The chromogenic reaction is completed within 1 min at room temperature when 8-hydroxylquinoline is used as catalyst. The proposed method has been applied to the direct determination of trace lead in clinical samples. The accuracy and precision are both very satisfactory.  相似文献

Software maintenance is indispensable in the software development process. Developers need to spend a lot of time and energy to understand the software when maintaining the software, which increases the difficulty of software maintenance. It is a feasible method to understand the software through the key classes of the software. Identifying the key classes of the software can help developers understand the software more quickly. Existing techniques on key class identification mainly use static analysis techniques to extract software structure information. Such structure information may contain redundant relationships that may not exist when the software runs and ignores the actual interaction times between classes. In this paper, we propose an approach based on dynamic analysis and entropy-based metrics to identify key classes in the Java GUI software system, called KEADA (identifying KEy clAsses based on Dynamic Analysis and entropy-based metrics). First, KEADA extracts software structure information by recording the calling relationship between classes during the software running process; such structure information takes into account the actual interaction of classes. Second, KEADA represents the structure information as a weighted directed network and further calculates the importance of each node using an entropy-based metric OSE (One-order Structural Entropy). Third, KEADA ranks classes in descending order according to their OSE values and selects a small number of classes as the key class candidates. In order to verify the effectiveness of our approach, we conducted experiments on three Java GUI software systems and compared them with seven state-of-the-art approaches. We used the Friedman test to evaluate all approaches, and the results demonstrate that our approach performs best in all software systems.  相似文献

An N-heterocyclic carbene(NHC)-catalyzed enantioselective Mannich reaction of the remote γ-carbon of cyclopropylcarbaldehydes is disclosed for the first time. Diastereo-and enantiomerically enriched multicyclic δ-lactam compound is afforded as the main product from 8 possible stereo-specific isomers through dynamic kinetic asymmetric transformation(DYKAT) processes. The β-sheet is one of the common protein secondary structures, and the aberrant aggregation of β-sheets is implicated in various neurodegenerative diseases. Cross-strand interactions are an important determinant of β-sheet stability. Accordingly, both diagonal and lateral cross-strand interactions have been studied. Surprisingly, diagonal cross-strand ion-pairing interactions have yet to be investigated. Herein, we present a systematic study on the effects of charged amino acid side-chain length on a diagonal ion-pairing interaction between carboxylate- and ammonium-containing residues in a β-hairpin. To this end, 2D-NMR was used to investigate the conformation of the peptides. The fraction folded population and the folding free energy were derived from the chemical shift data. The fraction folded population for these peptides with potential diagonal ion pairs was mostly lower compared to the corresponding peptide with a potential lateral ion pair. The diagonal ion-pairing interaction energy was derived using double mutant cycle analysis. The Asp2-Dab9 (Asp: one methylene; Dab: two methylenes) interaction was the most stabilizing (−0.79 ± 0.14 kcal/mol), most likely representing an optimal balance between the entropic penalty to enable the ion-pairing interaction and the number of side-chain conformations that can accommodate the interaction. These results should be useful for designing β-sheet containing molecular entities for various applications.  相似文献

The aim is to investigate the effect of lotus (Nelumbo nucifera Gaertn.) seedpod extract (LSE) on acetaminophen (APAP)-induced hepatotoxicity. LSE is rich in polyphenols and has potent antioxidant capacity. APAP is a commonly used analgesic, while APAP overdose is the main reason for drug toxicity in the liver. Until now, there has been no in vitro test of LSE in drug-induced hepatotoxicity responses. LSEs were used to evaluate the effect on APAP-induced cytotoxicity, ROS level, apoptotic rate, and molecule mechanisms. The co-treatment of APAP and LSEs elevated the survival rate and decreased intracellular ROS levels on HepG2 cells. LSEs treatment could significantly reduce APAP-induced HepG2 apoptosis assessed by DAPI and Annexin V/PI. The further molecule mechanisms indicated that LSEs decreased Fas/FasL binding and reduced Bax and tBid to restore mitochondrial structure and subsequently suppress downstream apoptosis cascade activation. These declines in COX-2, NF-κB, and iNOS levels were observed in co-treatment APAP and LSEs, which indicated that LSEs could ameliorate APAP-induced inflammation. LSE protected APAP-induced apoptosis by preventing extrinsic, intrinsic, and JNK-mediated pathways. In addition, the restoration of mitochondria and inflammatory suppression in LSEs treatments indicated that LSEs could decrease oxidative stress induced by toxic APAP. Therefore, LSE could be a novel therapeutic option for an antidote against overdose of APAP.  相似文献
