1-Deoxy-D-galactohomonojirimycin was synthesized in seven steps from optically pure allenylstannane 4 and L-lactate-derived aldehyde 5 in 48% overall yield. The key step was the Lewis acid catalyzed reaction of 4 and 5 to give the syn-amino alcohol in excellent yield and very high diastereoselectivity.  相似文献

Acclimation of the photosynthetic apparatus of chlorophyll b-less barley mutant chlorina f2 to low light (100 micromolm(-2)s(-1); LL) and extremely high light level (1000 micromolm(-2)s(-1); HL) was examined using techniques of pigment analysis and chlorophyll a fluorescence measurements at room temperature and at 77 K. The absence of chlorophyll b in LL-grown chlorina f2 resulted in the reduction of functional antenna size of both photosystem II (by 67%) and photosystem I (by 21%). Chlorophyll fluorescence characteristics of the LL-grown mutant indicated no impairment of the utilization of absorbed light energy in photosystem II photochemistry. Thermal dissipation of excitation energy estimated as non-photochemical quenching of minimal fluorescence (SV(0)) was significantly higher as compared to the wild-type barley grown under LL. Despite impaired assembly of pigment-protein complexes, chlorina f2 was able to efficiently acclimate to HL. In comparison with chlorina f2 grown under LL, HL-grown chlorina f2 was characterized by unaffected maximal photochemical efficiency of photosystem II (F(V)/F(M), doubled content of both beta-carotene and the xanthophyll cycle pigments and considerably reduced efficiency of excitation energy transfer from carotenoids to chlorophyll a. The enormous xanthophyll cycle pool size was however associated with reduced SV(0) capacity. We suggest that the substantial part of the xanthophyll cycle pigments is not bound to the remaining pigment-protein complexes and acts as filter for excitation energy, thereby contributing to the efficient photoprotection of chlorina f2 grown under HL.  相似文献

The objective of the presented study was to develop and optimize a simple, high-throughput method for the control of 32 mycotoxins (Fusarium and Alternaria toxins, aflatoxins, ergot alkaloids, ochratoxins, and sterigmatocystin) in beer. Due to the broad range of their physicochemical properties, the sample preparation step was simplified as much as possible to avoid analyte losses. The addition of acetonitrile to beer samples enabled precipitation of abundant matrix components. The clean-up efficiency was controlled by ambient mass spectrometry employing a direct analysis in real time (DART) ion source. For determination of analytes, ultra-high-performance liquid chromatography hyphenated with high-resolution mass spectrometry utilizing an orbitrap (U-HPLC-orbitrapMS) or time-of-flight (TOFMS) technology was used. Because of significantly better detection capabilities of the orbitrap technology, the U-HPLC-orbitrapMS method was chosen as a determinative step and fully validated. To compensate matrix effects, matrix-matched calibration was employed. The lowest calibration levels for most of the target mycotoxins ranged from 1 to 8 μg L(-1) beer and the recoveries of analytes were in range from 86 to 124%.  相似文献

The orthogonality of three columns coupled in two series was studied for the congener specific comprehensive two-dimensional GC separation of polychlorinated biphenyls (PCBs). A non-polar capillary column coated with poly(5%-phenyl–95%-methyl)siloxane was used as the first (1D) column in both series. A polar capillary column coated with 70% cyanopropyl-polysilphenylene-siloxane or a capillary column coated with the ionic liquid 1,12-di(tripropylphosphonium)dodecane bis(trifluoromethane-sulfonyl)imide were used as the second (2D) columns. Nine multi-congener standard PCB solutions containing subsets of all native 209 PCBs, a mixture of 209 PCBs as well as Aroclor 1242 and 1260 formulations were used to study the orthogonality of both column series. Retention times of the corresponding PCB congeners on 1D and 2D columns were used to construct retention time dependences (apex plots) for assessing orthogonality of both columns coupled in series. For a visual assessment of the peak density of PCBs congeners on a retention plane, 2D images were compared. The degree of orthogonality of both column series was, along the visual assessment of distribution of PCBs on the retention plane, evaluated also by Pearson's correlation coefficient, which was found by correlation of retention times tR,i,2D and tR,i,1D of corresponding PCB congeners on both column series. It was demonstrated that the apolar + ionic liquid column series is almost orthogonal both for the 2D separation of PCBs present in Aroclor 1242 and 1260 formulations as well as for the separation of all of 209 PCBs. All toxic, dioxin-like PCBs, with the exception of PCB 118 that overlaps with PCB 106, were resolved by the apolar/ionic liquid series while on the apolar/polar column series three toxic PCBs overlapped (105 + 127, 81 + 148 and 118 + 106).  相似文献

A comprehensive characterization of grape skin methanolic and ethanolic extracts prepared by pressurized fluid extraction (PFE) at various temperatures within 40 to 120 °C from two wine grape varieties, St. Laurent and Alibernet was performed. For the first time, an offline combination of PFE and electron paramagnetic resonance (EPR) spectroscopy together with other experimental methods was employed to assess the effect of extraction conditions on numerous extract characteristics including antioxidant or radical-scavenging ability, HPLC profile of anthocyanins, total phenolic compounds content (TPC), tristimulus color values (CIE Lab), and pH values. The properties of extracts depend on the solvent used, the mass of grape skins as well as on the extraction conditions among which the temperature plays a crucial role. In spite of wide interval of extraction temperatures, all extracts still retain their antioxidant and/or radical-scavenging properties, indicating that the extracts prepared by PFE can serve as potential source of functional food supplements or color enhancers.  相似文献

A simple NMR experiment capable of providing well resolved spectra under conditions where either radiation damping or static magnetic field inhomogeneity would broaden otherwise high‐resolution NMR spectra is introduced. The approach involves using a strong pulsed magnetic field gradient and a selective radio‐frequency pulse to encode a predetermined noise pattern into the spatial distribution of magnetization. Following readout in a much smaller field gradient, the noise sequence may be deconvolved from the acquired data and a high‐resolution spectrum is obtained, eliminating the effects of either radiation damping or the static field inhomogeneity. In the presence of field inhomogeneity a field map is also obtained from the same single transient. A quasi‐two‐dimensional version of the experiment eliminates the need for deconvolution and produces improved results with simplified processing, but without requiring a full two‐dimensional experiment. Example spectra are shown for both radiation damping and one‐dimensional field inhomogeneity with improvement in linewidths of more than a factor of 40.  相似文献

Applications and prospects of two-phase, tuneable solvent systems composed of ionic liquids (ILs) and supercritical fluids with an emphasis on supercritical carbon dioxide (scCO(2)) are reviewed. The IL-scCO(2) biphasic systems have increasingly been used in diverse fields of chemistry and technology, and some examples of these applications are mentioned here. Rational design of such applications can obviously benefit from pertinent data on phase equilibria including the partition coefficients of the prospective products and reactants between the two phases. Therefore, a reliable technique to measure the limiting partition coefficients would be of value. Here, the pros and cons of supercritical fluid chromatography in this respect are discussed. An overview of methods for predictive thermodynamic modelling of binary (IL-scCO(2)) and ternary (solute-IL-scCO(2)) systems is also included.  相似文献

Poly(ethylene oxide)-poly(methyl acrylate) diblock copolymers with narrow molecular weight distributions were synthesized using atom transfer radical polymerization. The copolymers were used as micellar templates for the synthesis of mesoporous silicas. The products were characterized using small-angle X-ray scattering, transmission electron microscopy (TEM) and nitrogen adsorption. The obtained silicas exhibited two-dimensional hexagonal structures of cylindrical mesopores, and thus can be classified as SBA-15 silicas. In some cases, the size of ordered domains was very small. The (100) interplanar spacings were 13–17 nm, depending on the size of the diblock copolymer used and on the synthesis conditions. Nitrogen adsorption showed that the silicas exhibited specific surface areas of 350–800 m2 g−1, pore volumes ∼1 cm3 g−1, and narrow pore size distributions. The BJH (nominal) pore diameters were up to ∼20 nm, but actual diameters of cylindrical pores are expected to be somewhat smaller. In many cases, the mesopores exhibited constrictions.  相似文献
