We have synthesized a group of glucamine and gluosamine-substituted cyanine dyes structurally related to indocyanine green (ICG) and have characterized these compounds with regard to their potential as contrast agents for biomedical optical imaging. The compounds reported herein exhibit increased hydrophilicity and less plasma protein binding (< 50%), and are thus expected to have different pharmacokinetic properties compared with ICG. Furthermore, we measured enhanced fluorescence quantum yields (7-15%) in a physiological environment with respect to ICG. For the derivative with the highest hydrophilicity (5a) the efflux from tumor and normal tissue was monitored by intensity-modulated diffuse optical spectroscopy after intravenous injection into tumor-bearing rats. In comparison with ICG, 5a exhibited a considerably enhanced tissue-efflux half-life (73 min versus less than 10 min for ICG in tumor tissue), a two-fold higher initial tissue absorption coefficient compared to ICG, and finally, it generated an elevated tumor-to-tissue concentration gradient up to 1 h after injection. In conclusion, compounds such as 5a are promising contrast agents for optical imaging, and could facilitate highly sensitive and specific detection of breast cancer or other malignancies by utilizing mechanisms similar to contrast-enhanced magnetic resonance imaging or computerized tomography.  相似文献

The photocatalytic degradation of the organophosphorus insecticide diazinon in aqueous suspensions has been studied by using titanium dioxide as a photocatalyst. The degradation of the insecticide was a fast process and included the formation of several intermediates that were identified using GC/ion-trap mass spectrometry with EI or CI in positive and negative ionization mode and HPLC/electrospray-QqTOF mass spectrometry. Since primarily hydroxy derivatives were identified in these aqueous suspensions, the mechanism of degradation was probably based on hydroxyl radical attack. The initial oxidative pathways of the degradation of diazinon involved the substitution of sulfur by oxygen on the Pz.dbnd6;S bond, cleavage of the pyrimidine ester bond, and oxidation of the isopropyl group. Exact mass measurements of the derivatives allowed the elemental formula of the molecules to be determined confidently. Similarities to the metabolic pathways occurring in living organisms were observed.  相似文献

A simple synthetic route was used for the synthesis of a novel series of alternating copolymers based on substituted 2,7‐distyrylfluorene bridged through alkylene chains. First, 2,7‐dibromofluorene was reacted with 2 equiv of butyllithium, and this was followed by a treatment with 1 equiv of α,ω‐dibromoalkane to yield the intermediate, poly(2,7‐dibromofluorene‐9,9‐diyl‐alt‐alkane‐α,ω‐diyl). ( 1 ) Heck coupling of the latter with 1‐tert‐butyl‐4‐vinylbenzene afforded the target, poly[2,7‐bis(4‐tert‐butylstyryl)fluorene‐9,9‐diyl‐alt‐alkane‐α,ω‐diyl] ( 2 ). The two versions of 2 ( 2a and 2b which have hexane and decane, respectively, as alkane groups) were readily soluble in common organic solvents. Their glass‐transition temperature was relatively low (52 and 87 °C). An intense blue photoluminescence emission with maxima at about 408 and 409 nm was observed in tetrahydrofuran solutions, whereas thin films exhibited an orange emission with maxima at 569 and 588 nm. Very large redshifts of the photoluminescence maxima and Stokes shifts in thin films indicated strong aggregation in the solid state. Both polymers oxidized and reduced irreversibly. Single‐layer light‐emitting diodes with hole‐injecting indium tin oxide and electron‐injecting aluminum electrodes were fabricated. They emitted orange light with external electroluminescence efficiencies of 0.52 and 0.36% photon/electron, as determined in light‐emitting diodes made of 2a and 2b , with alkylenes of (CH2)6 and (CH2)10, respectively. An increase in the external electroluminescence efficiency up to 1.5% was reached in light‐emitting diodes made of polymer blends consisting of 2a and poly(9,9‐dihexadecylfluorene‐2,7‐diyl), which emitted blue‐white light. © 2007 Wiley Periodicals, Inc. Polarization is indicative of material anisotropy, a property that reveals structural orientation information of molecules inside the material. Herein we investigate whether polarization can be detected optoacoustically in scattering and absorbing tissues. Using a laboratory prototype of polarization-sensitive optoacoustic tomography, we demonstrate high-resolution reconstructions of dichroism contrast deep in optically diffusive tissue-mimicking phantoms. The technique is expected to enable highly accurate imaging of polarization contrast in tissues, far beyond the current capabilities of pure optical polarization-imaging approaches.  相似文献

Diffuse optical tomography is a novel imaging technique that resolves and quantifies the optical properties of objects buried in turbid media. Typically, numerical solutions of the diffusion equation are employed to construct the tomographic problem when media of complex geometries are investigated. Numerical methods offer implementation simplicity but also significant computation burden, especially when large three-dimensional reconstructions are involved. We present an alternative method of performing tomography of diffuse media of arbitrary geometries by means of an analytical approach, the Kirchhoff approximation. We show that the method is extremely efficient in computation times and consider its potential as a real-time three-dimensional imaging tool.  相似文献

Fluorochromes have become essential reporter molecules in biological research. We show that the depth-resolved distribution of fluorochromes in small animals can be imaged with 25 fmol sensitivity and 150 microm spatial resolution by means of multispectral photoacoustic imaging. The major advantage of the multispectral approach is the sensitive differentiation of chromophores and fluorochromes of interest based on self-reference measurements, as evidenced in this study by resolving a commonly used fluorochrome (Alexa Fluor 750) in mouse. The suggested method is well suited for enhancing visualization of functional and molecular information in vivo and longitudinally.  相似文献

The sharp ring lemma in two dimensions states that no disc has a radius below c n (R 2) = (F 2n−3−1)−1—where F k denotes the kth Fibonacci number—and that the lower bound is attained in essentially unique Apollonian configurations. In this article, generalizations of the ring lemma to three dimensions are discussed, a version of the ring lemma in three dimensions is proved, and a natural generalization of the extremal two-dimensional configuration—thought to be extremal in three dimensions—is given.
