Enzyme cascades combining epimerization and isomerization steps offer an attractive route for the generic production of rare sugars starting from accessible bulk sugars but suffer from the unfavorable position of the thermodynamic equilibrium, thus reducing the yield and requiring complex work‐up procedures to separate pure product from the reaction mixture. Presented herein is the integration of a multienzyme cascade reaction with continuous chromatography, realized as simulated moving bed chromatography, to overcome the intrinsic yield limitation. Efficient production of D ‐psicose from sucrose in a three‐step cascade reaction using invertase, D ‐xylose isomerase, and D ‐tagatose epimerase, via the intermediates D ‐glucose and D ‐fructose, is described. This set‐up allowed the production of pure psicose (99.9 %) with very high yields (89 %) and high enzyme efficiency (300 g of D ‐psicose per g of enzyme).  相似文献

Treatment of CuCl with the lithiated silyl amides RSi(NLiPh)3 (R = Me, Ph, Vin) in THF as solvent led to the formation of the novel CuI cluster compounds [Li(THF)4]2[Cu10{RSi(NPh)3}4]. For each of the three compounds the X‐ray crystal structure analysis revealed similar Si4N12Cu10 cores which are based on cubane like Cu8 cores bearing two additional peripheral copper atoms. The copper atoms are coordinated nearly linearly by the μ5‐bridging silyl amide ligands with Cu–N distances in the range of 187.1(3) to 194.5(4) pm and N–Cu–N angles of 171.6(1) to 178.7(1)°. For each of the compounds the structural parameters are very similar which indicates that the structures are barely influenced by the different steric requirements of the organic groups bound to silicon.  相似文献

Antioxidants are important for maintaining the appropriate balance between oxidizing and reducing species in the body and thus preventing oxidative stress. Many natural compounds are being screened for their possible antioxidant activity. It was found that a mushroom pigment Norbadione A, which is a pulvinic acid derivative, shows an antioxidant activity; the same was found for other pulvinic acid derivatives and structurally related coumarines. Based on the results of in vitro studies performed on these compounds as a part of this study quantitative structure–activity relationship (QSAR) predictive models were constructed using multiple linear regression, counter-propagation artificial neural networks and support vector regression (SVR). The models have been developed in accordance with current QSAR guidelines, including the assessment of the models applicability domains. A new approach for the graphical evaluation of the applicability domain for SVR models is suggested. The developed models show sufficient predictive abilities for the screening of virtual libraries for new potential antioxidants.  相似文献

Macrolide antibiotics, such as azithromycin and erythromycin, are in widespread use for the treatment of bacterial infections. Macrolides are taken up and excreted mainly by bile. Additionally, they have been implicated in biliary system diseases and to modify the excretion of other drugs through bile. Despite mounting evidence for the interplay between macrolide antibiotics and bile acids, the molecular details of this interaction remain unknown. Herein, we show by NMR measurements that macrolides directly bind to bile acid micelles. The topology of this interaction has been determined by solvent paramagnetic relaxation enhancements (solvent PREs). The macrolides were found to be bound close to the surface of the micelle. Increasing hydrophobicity of both the macrolide and the bile acid strengthen this interaction. Both bile acid and macrolide molecules show similar solvent PREs across their whole structures, indicating that there are no preferred orientations of them in the bile micelle aggregates. The binding to bile aggregates does not impede macrolide antibiotics from targeting bacteria. In fact, the toxicity of azithromycin towards enterotoxic E. coli (ETEC) is even slightly increased in the presence of bile, as was shown by effective concentration (EC50) values.  相似文献

A convenient synthetic approach for the preparation of uniform metallopolymer‐containing hollow spheres based on 2‐(methacryloyloxy)ethyl ferrocenecarboxylate (FcMA) as monomer by sequential starved feed emulsion polymerization is described. Core/shell particles consisting of a noncrosslinked poly(methyl methacrylate) core and a slightly crosslinked ferrocene‐containing shell allows for the simple dissolution of core material and, thus, monodisperse metallopolymer hollow spheres are obtained. Since PFcMA is incorporated in the particle shell, herein investigated hollow spheres can be addressed by external triggers, i.e., solvent variation and redox chemistry in order to change the particle swelling capability. PFcMA‐containing core/shell particles and hollow spheres are characterized by transmission electron microscope (TEM), scanning electron microscopy, cryogenic TEM, thermogravimetric analysis, and dynamic light scattering in terms of size, size distribution, hollow sphere character, redox‐responsiveness, and composition. Moreover, the general suitability of prepared stimulus‐responsive nanocapsules for the use in catch‐release systems is demonstrated by loading the nanocapsules with malachite green as model payload followed by release studies.

Grinding methods were evaluated aiming to determine yield and composition of essential oils (EOs) from Baccharis articulata (Lam) Pers., called as carqueja, a native plant from South Brazil. Cryogenic, knife (with and without cooling) and ball mills were used. The major constituents found in EOs were β-pinene, caryophyllene, spathulenol and caryophyllene oxide, but differences in 21 compounds were observed. All grinding processes reduced monoterpenes and oxygenated sesquiterpenes with concentration of hydrocarbon sesquiterpenes. The sesquiterpenes (hydrocarbons and oxygenated) were found from 63.55 to 86.02% while the monoterpene hydrocarbons’ concentration ranged from 13.98 to 36.45%. Plants milled with knife mill provided EOs with profile similar to those not grinded. Despite the lower working temperature, cryogenic milling resulted in smaller EO yield (0.40 ± 0.03%) in comparison to other grinding methods (yields from 0.50 ± 0.02 to 0.56 ± 0.03%) and different chromatographic profile. Microscopic analysis showed the smaller particle size provided by cryogenic grinding, which leads to glandular trichome rupture and consequently to loss of EOs. This study showed that grinding should be carefully evaluated to provide reproducible results in essential oil analysis.  相似文献

An Euler tour of a hypergraph (also called a rank‐2 universal cycle or 1‐overlap cycle in the context of designs) is a closed walk that traverses every edge exactly once. In this paper, using a graph‐theoretic approach, we prove that every triple system with at least two triples is eulerian, that is, it admits an Euler tour. Horan and Hurlbert have previously shown that for every admissible order >3, there exists a Steiner triple system with an Euler tour, while Dewar and Stevens have proved that every cyclic Steiner triple system of order >3 and every cyclic twofold triple system admits an Euler tour.  相似文献
