We have studied the micro-homogeneity of boron carbide powders by inductively coupled plasma optical emission spectrometry (ICP-OES) and total reflection X-ray fluorescence spectrometry (TXRF) using slurry sampling. To get information on the particle size distributions of the powders, the stabilized slurries of boron carbide powders were nebulized, the aerosols were transported into a Batelle impactor and the droplets were collected on the impactor stages bearing TXRF sample holders. In a first series of measurements, parameters of the impaction like the duration of the impaction and the use of glutinous substance on the sample holders were optimized. The different mass size fractions for industrial boron carbide powders were determined by weight measurements of the fractions collected on the different stages. The established particle size distributions were in the range of 0.5 to >16 μm and found similar to those determined by laser diffraction reported elsewhere. Analyses of the mass fractions by slurry sampling TXRF showed that Ca, Ti, Cr, Mn, Fe, Ni and Cu within the measurements errors were homogeneously distributed over the mass fractions between 0.5 and 4 μm and that their concentrations agreed with the bulk composition, as determined with ICP-OES subsequent to digestion. However, light underestimates were found at the 5 (Mn) up to 150 μg g?1 (Fe) level. Finally, boron carbide powders were washed out with nitric acid with different concentrations and leaching solutions and the residues were analyzed by ICP-OES and TXRF respectively. It is shown that up to 60% of the residual trace impurities in the powder studied can be removed by leaching with 34% (v/v) of nitric acid.

The major octabromo isomer of technical octabromo diphenyl ether mixture (technical octaBDE) DE-79 was isolated by RP-HPLC. Three serially coupled columns (each 250 mm long) enabled a good separation of the target compound from other congeners using 100% ACN as eluent. Approximately 100 microg of the target compound was isolated with a purity of >90% and investigated by MS for confirmation of the degree of bromination. 1H-NMR and 2-D 1H-13C correlation NMR spectra unequivocally clarified that the octaBDE in question is 2,2',3,3',4,4',6,6'-octabromodiphenyl ether (BDE 197). Based on annual production rates of technical BDE products (1999/2001), approximately 380 tons of BDE 197 were annually produced which, on the long term, may enter the environment. Compared with other individual BDE congeners, BDE 197 has the seventh highest application rate. Reductive debromination of BDE 197 can lead to four hepta-, 15 hexa-, 23 penta-, and 28 tetra-BDE isomers, respectively. This variety includes all known major BDEs of environmental concern (BDE 47, 85, 99, 100, 153, 154, and 183). The identification of BDE 197 in technical octaBDE DE-79 strongly suggests that research on the environmental fate of BDEs should include this key-BDE congener.  相似文献

In this study the influence of aromatic dopant benzene on the sensitivity of GC-APPI-DMS to gasoline related aromatic compounds was investigated. This influence was investigated on example of four gasolin related fingerprints (toluene, ethylbenzene, o-xylene, and 1,2,4-trimethylbenzene), which were found in high relative abundance in the water-soluble gasoline fraction. The analysis of calibration curves slopes demonstrats that the GC-APPI-DMS sensitivity to gasoline fingerprints can be improved by up to seven times when benzene concentration in nitrogen carrier gas is less than 10 ppmv/v. The estimated detection limits (S/N?=?3) for the analyzed in this study compounds were found to be within the range of 33–105 μg L?1 at benzene concentration in the carrier gas of 2.27 ppmv/v (10 μL injection volume). These limits of detection may be reduced (at the cost of lower resolution) using the larger injection volumes. For example, increase of injection volume to 100 μL at benzene concentration in the carrier gas of 2.27 ppmv/v leads to reduction of LOD values for toluene, ethylbenzene, and o-xylene to 11.1, 13.3, and 5.3 μg L?1, respectively.  相似文献

The circumsporozoite protein (CSP) of Plasmodium falciparum is a leading candidate antigen for inclusion in a malaria subunit vaccine. We describe here the design of a conformationally constrained synthetic peptide, designated UK-39, which has structural and antigenic similarity to the NPNA-repeat region of native CSP. NMR studies on the antigen support the presence of helical turn-like structures within consecutive NPNA motifs in aqueous solution. Intramuscular delivery of UK-39 to mice and rabbits on the surface of reconstituted influenza virosomes elicited high titers of sporozoite crossreactive antibodies. Influenza virus proteins were crucially important for the immunostimulatory activity of the virosome-based antigen delivery system, as a liposomal formulation of UK-39 was not immunogenic. IgG antibodies elicited by UK-39 inhibited invasion of hepatocytes by P. falciparum sporozoites, but not by antigenically distinct P. yoelii sporozoites. Our approach to optimized virosome-formulated synthetic peptide vaccines should be generally applicable for other infectious and noninfectious diseases.  相似文献

Accurate in silico models for predicting aqueous solubility are needed in drug design and discovery and many other areas of chemical research. We present a statistical modeling of aqueous solubility based on measured data, using a Gaussian Process nonlinear regression model (GPsol). We compare our results with those of 14 scientific studies and 6 commercial tools. This shows that the developed model achieves much higher accuracy than available commercial tools for the prediction of solubility of electrolytes. On top of the high accuracy, the proposed machine learning model also provides error bars for each individual prediction.  相似文献

The determination of glucose in microfluidic chips made of glass or PMMA was used as a model for the combination of an enzymatic reaction with the separation of compounds. It was based on the enzymatic oxidation of glucose and the amperometric detection of hydrogen peroxide. Real samples frequently contain compounds, such as ascorbic acid, which may interfere with quantitative glucose determinations. Thus, electrophoretic separation of specific from unspecific signals was envisaged by applying electric fields which are also used to control the flow of liquid via electroosmotic effects. Surface charge densities of the capillaries influence the electroosmotic flow (EOF). They are dependent on the chip material and on the adsorption of components from the background electrolyte. Reversal of the EOF after addition of cetyltrimethylammonium bromide (CTAB) and an increase in EOF after addition of sodium dodecylsulfate (SDS) were observed at lower surfactant concentrations with the PMMA chips rather than with the glass chips. For both chip materials these concentrations were below the critical micelle concentration. Effective separation of H2O2 and ascorbic acid was achieved with low CTAB concentrations, which lead to a reduction, but not to a reversal of the EOF. Reversal of the EOF by higher CTAB concentrations or the increase in cathodic EOF by SDS accelerated ascorbic acid transportation and reduced the differences in migration times. Thus, for the specific determination of glucose, glucose oxidase was added together with low CTAB concentrations to the background electrolyte. This avoided interference from ascorbic acid, and data obtained from the analysis of fruit juices showed a good correlation to data obtained from a reference method.  相似文献

The biological function of the aspartic protease from HIV-1 has recently been related to the conformational flexibility of its structural scaffold. Here, we use a multistep strategy to investigate whether the same mechanism affects the functionality in the pepsin-like fold. (i) We identify the set of conserved residues by using sequence-alignment techniques. These residues cluster in three distinct regions: near the cleavage-site cavity, in the four beta-sheets cross-linking the two lobes, and in a solvent-exposed region below the long beta-hairpin in the N-terminal lobe. (ii) We elucidate the role played by the conserved residues for the enzymatic functionality of one representative member of the fold family, the human beta-secretase, by means of classical molecular dynamics (MD). The conserved regions exhibit little overall mobility and yet are involved into the most important modes of structural fluctuations. These modes influence the substrate-catalytic aspartates distance through a relative rotation of the N- and C-terminal lobes. (iii) We investigate the effects of this modulation by estimating the reaction free energy at different representative substrate/enzyme conformations. The activation free energy is strongly affected by large-scale protein motions, similarly to what has been observed in the HIV-1 enzyme. (iv) We extend our findings to all other members of the two eukaryotic and retroviral fold families by recurring to a simple, topology-based, energy functional. This analysis reveals a sophisticated mechanism of enzymatic activity modulation common to all aspartic proteases. We suggest that aspartic proteases have been evolutionarily selected to possess similar functional motions despite the observed fold variations.  相似文献

EPR, UV/Vis and FTIR spectroscopy as well as thermal analysis (TA/MS) were applied to study the influence of sulfate species present in the anatase support on the specific nature of VOx species in supported VOx/TiO2 catalysts. Those sulfate species modify the local structure of the supported vanadyl species and lead to the formation of two types of VO2+ sites instead of only one type being formed on sulfate‐free anatase. EPR and FTIR spectroscopic measurements revealed that a part of the VO2+ species are directly bound to the surface sulfate species. By TA/MS it was found that SO2 is released at lower temperature from VOx/TiO2 in comparison to the vanadium‐free support. The direct bonding between sulfate and VOx species stabilizes the latter on the surface of VOx/TiO2 resulting in three effects: 1) a higher V site dispersion in comparison to sulfate‐free TiO2, 2) a better resistance of surface vanadyls against diffusion into the bulk of the support and 3) a much faster reoxidation of reduced V sites than observed on sulfate‐free TiO2.  相似文献

The improvement and performance of a micellar electrokinetic capillary chromatography assay for cefepime in human serum and plasma with a 50 μm id fused‐silica capillary elongated from 40 to 60 cm is reported. Sample preparation with dodecylsulfate protein precipitation at pH 4.5, the pH 9.1 separation medium, and the applied voltage were as reported previously [16]. The change resulted in a significant lower current, higher resolution, and increased detection time intervals. The performance of the assay with multilevel internal calibration was assessed with calibration and control samples. Quality assurance data of a 2‐year period assessed under the new conditions demonstrated the robustness of the assay. In serum samples of patients who received both cefepime and sulfamethoxazole, cefepime could not be detected due to the inseparability of the two compounds. The presence of an interference can be recognized by an increased peak width (width > 0.2 min), the appearance of a shoulder or an unresolved double peak. The patient data gathered during a 3‐year period reveal that introduction of therapeutic drug monitoring led to a 50% reduction of the median drug level. The data suggest that therapeutic drug monitoring can help to minimize the risk of major adverse reactions and to increase drug safety on an individual basis.  相似文献

An evaluation of the non-boiling evaporation technique for the preconcentration of Pb, Cd, Cu, Zn at the pg g?1 level in water samples is presented. Various improvements were made to allow efficient control of contamination problems at these extremely low concentrations. They include the choice of FEP Teflon for the evaporation containers and the use of sophisticated cleaning, ageing and pre-conditionning procedures. Detailed calibration graphs were obtained down to the sub-pg g?1 level by processing ultra-low concentration standards. This technique was then applied to the determination of these four metals in snow samples collected in Greenland and Antarctica.  相似文献
