Sample pre-concentration can be a critical element to improve sensitivity of integrated microchip assays. In this work a converging Y-inlet microfluidic channel with integrated coplanar electrodes was used to investigate transverse DNA and protein migration under uniform direct current (DC) electric fields to assess the ability to concentrate a sample prior to other enzymatic modifications or capillary electrophoretic separations. Employing a pressure-driven flow to perfuse the microchannel, negatively charged samples diluted in low and high ionic strength buffers were co-infused with a receiving buffer of the same ionic strength into a main daughter channel. Experimental results demonstrated that, depending of the buffer selection, different DNA migration and accumulation dynamics were seen. Charged analytes could traverse the channel width and accumulate at the positive bias electrode in a low electroosmotic mobility, high electrophoretic mobility, high ionic strength buffer or migrated towards an equilibrium position within the channel in a high electroosmotic mobility, high electrophoretic mobility, low ionic strength buffer. The various migration behaviours are the result of a balance between the electrophoretic force and a drag force induced by a recirculating electroosmotic flow generated across the channel width due to the bounding walls. Under continuous flow conditions, DNA samples were concentrated several-fold by balancing these transverse electrokinetic forces. The electrokinetic trapping technique presented here is a simple technique which could be expanded to concentrate or separate other analytes as a preconditioning step for downstream processes. 相似文献

By using calcium fluorite and barium fluorite as test materials, we demonstrated that homovalent "dopants" can greatly affect ionic conductivity through locally changing the defect density. Whilst this doping is a state-of-the-art effect in the case of dopants that replace native ions of different charge (heterovalent dopants), it is a rather surprising effect at a first glance for substitutional dopants of the same charge; here, the phenomenon is not electrostatic, but elastic in nature. As a consequence of size mismatch, the smaller Ca atoms in the BaF(2) lattice favored the formation of interstitial sites that were located close to the Ca atoms, whilst doping larger Ba species into the CaF(2) phase favored vacancy formation. In terms of conductivity, and in agreement with the different mobilities, the first doping effect was favorable, whilst the other decreased conductivity. The concentration effects were formalized by a heterogeneous Frenkel reaction that was distinguished from the mean Frenkel reaction by additional (elastic) trapping that became more pronounced the lower the temperature. It was very revealing to relate this phenomenon to CaF(2)-BaF(2) multilayers and composites. In very general terms, these effects in the solid solutions were understood as being the atomistic limit of the interfacial charge-transfer that occurred at the hetero-interface of the crystallites or films, and reflected the transition from heterogeneous doping (higher-dimensional doping) to homogeneous doping (zero-dimensional doping). 相似文献

A microfluidic device for continuous, real time blood plasma separation is introduced. The principle of the blood plasma separation from blood cells is supported by the Zweifach-Fung effect and was experimentally demonstrated using simple microchannels. The blood plasma separation device is composed of a blood inlet, a bifurcating region which leads to a purified plasma outlet, and a concentrated blood cell outlet. It was designed to separate blood plasma from an initial blood sample of up to 45% inlet hematocrit (volume percentage of cells). The microfluidic network was designed using an analogous electrical circuit, as well as analytical and numerical studies. The functionality of this device was demonstrated using defibrinated sheep blood. During 30 minutes of continuous blood infusion through the device, all the erythrocytes (red blood cells) traveled through the device toward the concentrated blood outlet while only the plasma was separated at the bifurcating regions and flowed towards the plasma outlet. The device has been operated continuously without any clogging or hemolysis of cells. The experimentally determined plasma selectivity with respect to blood hematocrit level was almost 100% regardless of the inlet hematocrit. The total plasma separation volume percent varied from 15% to 25% with increasing inlet hematocrit. Due to the device's simple structure and control mechanism, this microdevice is expected to be used for highly efficient continuous, real time cell-free blood plasma separation from blood samples for use in lab on a chip applications. 相似文献

We study infrared divergences due to ultraviolet–infrared mixing in quantum field theory on Moyal space with Lorentzian signature in the Yang–Feldman formalism. Concretely, we are considering the ${\phi^4}$ and the ${\phi^3}$ model in arbitrary even dimension. It turns out that the situation is worse than in the Euclidean setting, in the sense that we find infrared divergences in graphs that are finite there. The Bunte salt derivative was prepared from bovine insulin-A isolated from natural sources, and was purified by chromatography. Natural abundance 40.55 MHz 15N NMR spectra were measured in water and dimethyl sulphoxide. The latter solvent gave the best resolution, and 20 of the 22 possible amide nitrogen signals were detected.
