Peptides with an N-alkoxy or N-aryloxy amino acid at their N-terminus were synthesized and successfully ligated with a peptide thioester by silver ion activation under a slightly acidic condition without requiring protection of the side chain amino groups. The N-methoxy group was easily cleaved by the SmI2 reduction in CH3OH aq. to obtain the desired peptide with a native peptide bond. This method was successfully applied to the synthesis of the human atrial natriuretic peptide showing the efficiency of the novel ligation.  相似文献

Steady-state polarization measurements of lithium titanium oxide (LTO; Li[Li1/3Ti5/3]O4) were carried out using the 0-V lithium-ion cells consisting of two identical LTO-electrodes with a parallel-plate symmetrical electrode configuration. The sinusoidal voltage with the peak amplitude of 1.0 V was imposed at 0.1 Hz upon the 0-V cells and the current response was measured as a function of time. The steady-state polarization, obtained by plotting the current versus applied voltage, was linear in current up to approximately 60 mA cm?2 or 4 A g?1 based on the LTO weight and suggested the resistance polarization only for the lithium insertion electrode of the LTO. The method was also applied to lithium aluminum manganese oxide (LAMO; Li[Li0.1Al0.1Mn1.8]O4) and the resistance polarization of the LAMO-electrode was determined for currents up to approximately 25 mA cm?2 or 2 A g?1 based on the LAMO weight. The validity of the results was examined for the polarization measurements of the 2.5-V lithium-ion battery consisting of LTO and LAMO, and the significance of the polarization measurements of lithium insertion electrodes for high-power applications was discussed.  相似文献

The effect of the interaction between DNA and the photosensitizer on photosensitized singlet oxygen (1O2) generation was investigated using DNA-binding alkaloids, berberine and palmatine. These photosensitizers were bound to DNA by electrostatic force. Near-infrared luminescence measurement demonstrated that the photoexcited alkaloids can generate 1O2 only when the photosensitizers are bound to DNA. A fluorescence decay study showed significant enhancement of the lifetime of their photoexcited state with the DNA binding. A calculation study suggested that the electrostatic interaction with DNA inhibits the quenching of the photoexcited state of these alkaloids via intramolecular electron transfer, leading to the prolongation of the lifetime of their excited state. This effect should enhance their intersystem crossing and the yield of energy transfer to molecular oxygen. The results show that the electrostatic interaction with DNA significantly affects the 1O2 generation activity of a photosensitizer. In addition, this interaction may be applied to the control and the design of photosensitizers for medical applications such as photodynamic therapy.  相似文献

Sol-Gel Processing of Transparent PLZT((Pb,La)(Zr,Ti)O3) Fibers   总被引:1,自引:0,他引:1  
PLZT((Pb,La)(Zr,Ti)O3) ceramic fibers were prepared by the sol-gel method from a solution of lead acetate trihydrate, lanthanum isopropoxide, zirconium n-propoxide and titanium isopropoxide containing 2-methoxyethanol as the solvent. The as-drawn gel fibers were pretreated in an autoclave and preheated in flowing H2O vapor prior to the heat-treatment for crystallization. The effects of autoclaving and preheating in flowing H2O vapor on the structure and composition of the resultant fibers were investigated. Optically transparent single phase perovskite PLZT fibers of 5–15 m in thickness and 5 cm in length were successfully prepared when the autoclave-treatment and the preheating in flowing H2O vapor were conducted prior to crystallization. These treatments are thought to promote hydrolysis of the organic groups and polycondensation reaction in the gel fibers, leading to the successful formation of dense, crack-free transparent PLZT fibers.  相似文献

We investigated the chemical modifications of the nitroquinazoline derivative (1) through the replacement of the NH group at the C(4)-position with several N-alkyl groups to increase the lipophilicity at the C(4)-position. Among them, we found that the N-methyl analogue (5a) showed a 2-fold loss in the inhibitory activity toward tumor necrosis factor-alpha (TNF-alpha) production in vitro as compared with the NH analogue (1); however, 5a exhibited an oral inhibitory activity on TNF-alpha production with an ED50 value of 26 mg/kg, whereas 1 did not. Moreover, the oral bioavailability of 5a was higher than that of 1 (1, F=1%; 5a, F=21%), and the calculated ClogP value for 5a was higher than that for 1. These results suggest that the improved lipophilicity of 5a compared with that of 1 reflects its greater inhibitory activity on TNF-alpha production in vivo as well as oral bioavailability.  相似文献

Methoxy and fluorine analogs substituted on the terminal carbon of the pentyl chain of N-(piperidinyl)-1-(2,4-dichlorophenyl)-4-methyl-5-(4-pentylphenyl)-1H-pyrazole-3-carboxamide (O-1302) were synthesized in a multi-step process from 5-phenyl-1-pentanol, which was based on the 1,5-diarylpyrazole core template of N-(piperidinyl)-5-(4-chlorophenyl)-1-(2,4-dichlorophenyl)-4-methyl-1H-pyrazole-3-carboxamide (SR141716) through condensation of the respective amine with pyrazole carboxylic acid, in order to develop tracers for medical imaging. Their potency for inhibiting the binding of the CB1 antagonist [(3)H]SR141716 was evaluated with the aim of developing positron emission tomography (PET) ligands for the cerebral cannabinoid CB1 receptor. These analogs bearing a piperidinyl carboxamide at the C(3) of the pyrazole ring exhibited affinities comparable to those of the CB1 reference antagonist SR141716, which warrants further investigation using the radiolabeled form for biological imaging studies. A morpholine ring substituted at the C(3) of the pyrazole ring resulted in a reduction of the CB1 affinity.  相似文献

We present a reaction scheme of silver containing 5-substituted uracils (N) bridge formation with two silver ions to a silver-mediated base pair [N-Ag(2)-N] by using the conductor-like polarizable continuum model (CPCM)-B3LYP/aug-cc-pVDZ level of theory. The whole reaction scheme is divided into the following three steps: (1) silver ion binding and deprotonation, (2) silver ion transfer, and (3) dimer formation and structural fluctuation. With a new pK(a) computing scheme proposed in our previous studies, it is found that a silver coordination decreases the pK(a) of N by 2.5-3.0 pK(a) units, which is an important clue for silver-ion selectivity by N. Judging from the calculation, we revealed that the silver ion transfer reaction and the dimerization reaction occur spontaneously. Moreover, both reactions are independent of the C5 ligand in N so that the deprotonation reaction, which is the first step of this scheme, plays a key role in forming the [N-Ag(2)-N] pairing.  相似文献

Firefly emission is a well-known efficient bioluminescence. However, the mystery of the efficient thermal generation of electronic excited states in firefly still remains unsolved, particularly at the atomic and molecular levels. We performed SA-CASSCF(12,12)/6-31G* and CASPT2(12,12)/6-31G*//SA-CASSCF(12,12)/6-31G* calculations to elucidate the reaction mechanism of bioluminescence from the firefly dioxetanone in the gas phase. Adiabatic transition state (TS) for the O-O bond cleavage and the minimum energy conical intersection (MECI) were located and characterized. The unique topology of MECI featuring a seam of a sloped conical intersection for the firefly dioxetanone, which was uncovered for the first time, emerges along the reaction pathway to provide a widely extended channel to diabatically access the excited-state from the ground state.  相似文献
