Short and concise total asymmetric syntheses of (−)-isoretronecanol and (−)-trachelantamidine are reported. Oxidative cleavage of tert-butyl (S,S,S,Z)-7-[N-benzyl-N-(α-methylbenzyl)amino]cyclohept-3-ene-1-carboxylate, followed by hydrogenolysis promoted in situ cyclisation/reduction, which provided rapid access to the bicyclic core within (−)-isoretronecanol. Analogous treatment of the C(1)-epimer gave (−)-trachelantamidine. Overall, the syntheses of (−)-isoretronecanol and (−)-trachelantamidine were completed in eight and seven steps and 20 and 9.5% yield, respectively, from commercially available starting materials.  相似文献

In this work, we present an optical transit DEP flow cytometer for parallel single-cell analysis. Each cell's dielectric property is inferred from velocity perturbations due to DEP actuation in a microfluidic channel. Dual LED sources facilitate velocity measurement by producing two transit shadows for each cell passing through the channel. These shadows are detected using a 256-pixel linear optical array detector. Massively parallel analysis is possible as each pixel of the detector can independently analyze the passing cells. A wide channel (∼18 mm) was employed to carry many particles simultaneously, and the system was capable of detecting the velocity of over 200 cells simultaneously. We have achieved analysis rates for 10 µm diameter polystyrene spheres response exceeding 250 per second. With appropriate calibration, this DEP cytometer can quantitatively measure the dielectric response. The dielectric response (Clausius–Mossotti factor) of viable CHO cells was measured over the frequency range of 100 kHz to 6 MHz, and the obtained response matches the previously measured values by our group. The DEP cytometer uses simple modular components to achieve high throughput label-free single-cell dielectric analysis and can begin analyzing particles within 10 s after starting to pump the sample into the channel.  相似文献

The catalytic enantioselective synthesis of α‐fluorinated chiral tertiary alcohols from (hetero)aryl methyl ketones is described. The use of a bifunctional iminophosphorane (BIMP) superbase was found to facilitate direct aldol addition by providing the strong Brønsted basicity required for rapid aryl enolate formation. The new synthetic protocol is easy to perform and tolerates a broad range of functionalities and heterocycles with high enantioselectivity (up to >99:1 e.r.). Multi‐gram scalability has been demonstrated along with catalyst recovery and recycling. 1H NMR studies identified a 1400‐fold rate enhancement under BIMP catalysis, compared to the prior state‐of‐the‐art catalytic system. The utility of the aldol products has been highlighted with the synthesis of various enantioenriched building blocks and heterocycles, including 1,3‐aminoalcohol, 1,3‐diol, oxetane, and isoxazoline derivatives.  相似文献

A compact experimental setup that integrates laser-induced incandescence (LII) and one-angle elastic light scattering (1A-ELS) to measure the size of polydisperse soot aggregates is described. A 532 nm laser and a detection angle of 35 degrees were employed, which provided sensitivity for aggregate radius of gyrations (R g) of R g≤200 nm. Both lognormal and self-preserving distribution functions are compared with width parameters derived from both aggregation theory and transmission electron microscopy (TEM) measurements. Using these distributions, mean aggregate sizes derived from the scattering measurements are compared. The LII+1A-ELS technique is validated with a two-angle elastic light scattering (2A-ELS) approach with an additional detection angle at 145 deg. Unlike LII+1A-ELS, the 2A-ELS technique has the advantage of not requiring knowledge of soot optical properties. Good agreement is found between the two techniques for a given distribution. A fundamental discrepancy exists between distributions derived from TEM and those according to aggregation theory, limiting the accuracy of both 2A-ELS and LII+1A-ELS. The dependence of both techniques on laser fluence and hence soot temperature is examined and discussed.  相似文献

Polyethylene glycol (PEG) molecules with masses below 1300amu are electrosprayed (ES) from solution, mobility-selected at high resolution in a differential mobility analyzer (DMA), collected on a grid and imaged by transmission electron microscopy (ES–DMA–TEM). The DMA resolves individual n-mers, and selects only one out of the many present in the original sample. Ion identity is established from parallel mass spectra (ES-MS). The images reveal spherical particles 1.46nm in diameter, in good agreement with the known ion mass and bulk density. The DMA-selection technique opens new paths for the study of very small particles.  相似文献
