The protein beta(2)-microglobulin (beta(2)m) aggregates to form classical amyloid fibrils in patients undergoing long-term haemodialysis. Amyloid-like fibrils with a cross-beta fold can also be formed from wild-type beta(2)m under acidic conditions in vitro. The morphology of such fibrils depends critically on the conditions used: incubation of beta(2)m in low ionic strength buffers at pH 2.5 results in the formation of long (microm), straight fibrils while, at pH 3.6, short (<500 nm) fibrils form. At higher ionic strengths (0.2-0.4 M) at pH 1.5-3.6, the fibrils have a distinct curved and nodular morphology. To determine the conformational properties of beta(2)m within in vitro fibrils of different morphologies, limited proteolysis of each fibril type using pepsin was performed and the resulting peptide fragments identified by tandem mass spectrometry. For comparison, the proteolytic degradation patterns of monomeric beta(2)m and seven synthetic peptides spanning the entire sequence of the intact protein were similarly analysed. The results show that fibrils with different morphologies result in distinct digestion patterns. While the curved, worm-like fibrils are relatively weakly protected from proteolysis, the long, straight fibrils formed at pH 2.5 at low ionic strength show only a single cut-site at Val9, demonstrating that substantial refolding of the initially acid-denatured and unprotected state of beta(2)m occurs during assembly. The data demonstrate that the organisation of the polypeptide chain in fibrils with different morphological features differs considerably, despite the fact that the fibrils possess a common cross-beta architecture.  相似文献

The asymmetric synthesis of (−)-(S,S)-homaline was achieved in 8 steps from commercially available starting materials using the diastereoselective conjugate addition of the novel lithium amide reagent lithium (R)-N-(3-chloropropyl)-N-(α-methyl-p-methoxybenzyl)amide to methyl cinnamate to install the correct stereochemistry. Subsequent functional group manipulation of the resultant β-amino ester and Sb(OEt)3-mediated macrolactamisation was followed by homodimerisation to give (−)-(S,S)-homaline in 18% overall yield, representing the first asymmetric, and by far the most efficient synthesis of this natural product reported to date.  相似文献

The ammonium-directed olefinic epoxidations of a range of differentially N-substituted cyclic allylic and homoallylic amines (derived from cyclopentene, cyclohexene, and cycloheptene) have been investigated, and the reaction kinetics have been analyzed. The results of these studies suggest that both the ring size and the identity of the substituents on nitrogen are important in determining both the overall rate and the stereochemical outcome of the epoxidation reaction. In general, secondary amines or tertiary amines with nonsterically demanding substituents on nitrogen are superior to tertiary amines with sterically demanding substituents on nitrogen in their ability to promote the oxidation reaction. Furthermore, in all cases examined, the ability of the (in situ formed) ammonium substituent to direct the stereochemical course of the epoxidation reaction is either comparable or superior to that of the analogous hydroxyl substituent. Much slower rates of ring-opening of the intermediate epoxides are observed in cyclopentene-derived and cycloheptene-derived allylic amines as compared with their cyclohexene-derived allylic and homoallylic amine counterparts, allowing for isolation of these intermediates in both of the former cases.  相似文献

We review the generation of broadband THz radiation from femtosecond photo‐induced gas plasmas, with an emphasis on the highly efficient “AC‐bias” case where the plasma is generated and driven by a superposition of fundamental and second‐harmonic optical fields. The dependence on experimental parameters such as pulse energy, air pressure, polarization and focusing are presented, and compared to the predictions from semi‐quantitative models for the THz generation process, namely (i) a microscopic photocurrent model and (ii) a four‐wave mixing model. We also employ these models to the case of few‐cycle pulses, where the observed THz emission is related directly to the carrier‐envelope phase of the pulses, and hence provides a mechanism with which to measure this phase.}  相似文献

A scalable, one‐pot, enantioselective catalytic synthesis of 2‐pyrazolines from beta‐substituted enones and hydrazines is described. Pivoting on a two‐stage catalytic Michael addition/condensation strategy, the use of an aldehyde to generate a suitable hydrazone derivative of the hydrazine was found to be key for curtailing background reactivity and tuning the catalyst‐controlled enantioselectivity. The new synthetic method is easy to perform, uses a new and readily prepared cinchona‐derived bifunctional catalyst, is broad in scope, and tolerates a range of functionalities with high enantioselectivity (up to >99:1 e.r.). The significant scalability of this methodology was demonstrated with the synthesis of more than 80 grams of a pyrazoline product with 89 % catalyst recovery.  相似文献

Monodisperse, crosslinked polystyrene latexes were prepared by the dispersion technique. Some general observations regarding the effect of initial reagent concentrations on final particle size and size distribution are offered, in addition to a detailed discussion concerning the problems encountered with the use of the crosslinker divinylbenzene (DVB) in latex preparation. Particles synthesized in very polar media were found to reach their growth plateau sooner than those made in less polar surroundings. This trend was proposed to be the result of more effective nucleation in polar environments, which increases available surface area, thereby allowing the rapid replacement of monomer consumed within the particle phase during the polymerization. Attempts to favorably influence the growth rate and size distribution of particles during the reaction were unsuccessful, underlining the importance of the nucleation period in defining particle size characteristics. Up to 1% DVB was successfully incorporated in the synthesis of coagulum-free, monodisperse, 5 μm beads, by controlling the entry of the crosslinker into the particle phase during the major particle growth period. Latex stability is proposed to be largely dependent on the mobility of the adsorbed steric stabilizer. © 1995 John Wiley & Sons, Inc.  相似文献
