The blood–brain barrier (BBB) is a formidable physical and enzymatic barrier that tightly controls the passage of molecules from the blood to the brain. In fact, less than 2 % of all potential neurotherapeutics are able to cross it. Here, by applying the retro‐enantio approach to a peptide that targets the transferrin receptor, a full protease‐resistant peptide with the capacity to act as a BBB shuttle was obtained and thus enabled the transport of a variety of cargos into the central nervous system.  相似文献

Pyrrolidinophenones represent one emerging class of newly encountered drugs of abuse, also known as ‘new psychoactive substances’, with stimulating psychoactive effects. In this work, we report on the detection of the new designer drug α‐pyrrolidinohexiophenone (α‐PHP) and its phase I and II metabolites in a human urine sample of a drug abuser. Determination and structural elucidation of these metabolites have been achieved by liquid chromatography electrospray ionisation quadrupole time‐of‐flight mass spectrometry (LC‐ESI‐QTOF‐MS). By tentative identification, the exact and approximate structures of 19 phase I metabolites and nine phase II glucuronides were elucidated. Major metabolic pathways revealed the reduction of the ß‐keto moieties to their corresponding alcohols, didesalkylation of the pyrrolidine ring, hydroxylation and oxidation of the aliphatic side chain leading to n‐hydroxy, aldehyde and carboxylate metabolites, and oxidation of the pyrrolidine ring to its lactam followed by ring cleavage and additional hydroxylation, reduction and oxidation steps and combinations thereof. The most abundant phase II metabolites were glucuronidated ß‐keto‐reduced alcohols. Besides the great number of metabolites detected in this sample, α‐PHP is still one of the most abundant ions together with its ß‐keto‐reduced alcoholic dihydro metabolite. Monitoring of these metabolites in clinical and forensic toxicology may unambiguously prove the abuse of the new designer drug α‐PHP. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

The time resolution achievable in in situ high-temperature nuclear magnetic resonance experiments is investigated using laser heating of refractory materials. Three case studies using 27Al in alumina nanoparticles, 29Si in silicon carbide and 23Na in a glass-forming mixture of sodium carbonate and quartz have been conducted to distinguish the cases of (a) a fast-relaxing, high natural abundance nucleus, (b) a probe nucleus with low abundance and low spin–lattice relaxation rate, and (c) a complex and changing system of industrial relevance. The most suitable nucleus for in situ high-temperature studies is one with high abundance but slow relaxation because the differential relaxation time between hot and cold parts of the sample effectively removes the signal from the cold material. There is no "in situ penalty" from the diminishing Boltzmann polarization at high temperature since this effect is balanced by a corresponding increase of the spin–lattice relaxation rate. First-principles modeling, experimental, and thermodynamic methodologies were integrated to facilitate a fundamentally guided investigation of quaternary complex hydride compounds within the bialkali Na-Li-Al-H hydrogen storage system. The integrated approach has broad utility for the discovery, understanding, and optimization of solid-state chemical systems. Density functional theory ground-state minimizations, low-temperature powder neutron diffraction, and low-temperature synchrotron X-ray diffraction were coupled to refine the crystallographic structures for various low-temperature distorted Na2LiAlH6 allotropes. Direct method lattice dynamics were used to identify a stable Na2LiAlH6 allotrope for thermodynamic property predictions. The results were interpreted to propose transformation pathways between this allotrope and the less stable cubic allotrope observed at room temperature. The calculated bialkali dissociation pressure relationships were compared with those determined from pressure-composition-isotherm experiments to validate the predicted thermodynamic properties. These predictions enabled computational thermodynamic modeling of Na2LiAlH6 and competing lower order phases within the Na-Li-Al-H system over a wide of temperature and pressure conditions. The predictions were substantiated by experimental observations of varying Na2LiAlH6 dehydrogenation behavior with temperature. The modeling was used to identify the most favorable reaction pathways and equilibrium products for H discharge/recharge in the Na-Li-Al-H system, and to design conditions that maximize the theoretical hydrogen reversibility within the Na-Li-Al-H system.  相似文献

The ability to monitor and quantify anionic components of aerosols is important for developing a better fundamental understanding of temporal and spatial variations in aerosol composition. Of the many methods that can be used to detect anions, capillary electrophoresis is among the most attractive ones because of its high separation efficiency, high resolving power for ionic compounds, and ability to be miniaturized for in-field monitoring. Here we present a method to baseline resolve common aerosol components nitrate, sulfate, chloride, and over two dozen organic acids in a single separation. A capillary electrophoresis separation utilizing a pH 5.78 piperazine buffer with 1,5-naphthalenedisulfonic acid as a probe for indirect UV absorbance detection was developed for this analysis. Previously, two different buffers were required to adequately separate all of these compounds. Electrophoretic mobilities, limits of detection, and migration time reproducibilities were measured for 38 organic and 8 inorganic anions. For solutions of low conductivity, detection limits for electrokinetic injections were found to be up to two orders of magnitude lower (0.2-0.4 microM) than those for pressure injection (1-45 microM). This separation was optimized and used for routine analysis of aqueous extracts of ambient atmospheric aerosols, but may be extended to other samples containing similar mixtures of anions.  相似文献

Anionic phospholipids phosphatidic acid (PA), phosphatidylglycerol (PG), phosphatidylinositol (PI), and phosphatidylserine (PS) were examined for their effect on 1-palmitoyl-2-oleyl-sn-glycero-3-phosphatidylcholine (POPC)-containing liposomes used as coating material in capillary electrochromatography. Liposome solvent was N-(2-hydroxyethyl)piperazine-N'-(2-ethanesulfonic acid) (HEPES) buffer at pH 7.4 with and without 3 mM of CaCl2. The background electrolyte solution was HEPES buffer at pH 7.4. The net charge, size, and short-term stability of the liposomes were measured with a Zetasizer. Results showed that calcium interacts with all liposomes but most strongly with POPC/PA. The relative migration times, retention factors, and resolution of the model analytes (one cationic, three uncharged ions, and one anionic) were studied. All liposomes successfully coated the silica capillary. Without calcium the strongest interaction and best separation of the analytes were with the POPC/PI and POPC/PS coatings, while interactions with the POPC/PA coating were weak. Calcium enhanced the interactions of the model analytes with all coatings, and the interactions were then strongest with the POPC/PA coating. In the presence of calcium there appears to be a slight reorganization of the coating with increasing number of runs. Our results indicate strong interactions between calcium and the phosphate groups in phospholipids and demonstrate the significant role of the phospholipid polar head group in phospholipid coatings on silica surfaces.  相似文献

A chemical examination of the endophytic fungus Penicillium sp. isolated from the mangrove plant Aegiceras corniculatum resulted in the isolation and characterization of eight new indole triterpenes named shearinines D-K (1-8), along with shearinine A (9), paspalitrem A (10), and paspaline (11). Their stereostructures were determined by extensive spectroscopic data analyses and quantum chemical circular dichroism calculations. The biosynthetic relationship of all 11 alkaloids is suggested. Shearinines D (1), E (2), and (with reduced potency) G (4) exhibit significant in vitro blocking activity on large-conductance calcium-activated potassium channels.  相似文献

The first derivatives of catenated cyclotetraphosphinophosphonium cations, [(PhP)4PPhMe]+ (8a), [(MeP)4PMe2]+ (8b), [(CyP)4PPh2]+ (8d), [(CyP)4PMe2]+ (8e), [(PhP)4PPh2]+ (8f), [(PhP)4PMe2]+ (8g), are synthesized as trifluoromethanesulfonate (triflate, OSO2CF3-) salts through the reaction of cyclopentaphosphines (PhP)5 (4a) or (MeP)5 (4b) with methyl triflate (MeOTf) or by a net phosphenium ion [PR2+, R = Ph, Me; from R2PCl and trimethylsilyltriflate (Me3SiOTf)] insertion into the P-P bond of either cyclotetraphosphine (CyP)4 (3c) or cyclopentaphosphines (PhP)5 (4a) or (MeP)5 (4b). Although more conveniently prepared from 4a, compound 8a[OTf] can also be formed from (PhP)4 (3a) and MeOTf, and derivatives 8f[OTf] and 8g[OTf] are also accessible through reactions of 3a and R2PCl/Me3SiOTf with R = Ph or Me, respectively. A tetrachlorogallate salt of [(PhP)4PPhtBu]+ (8c) has been synthesized by alkylation of 4a with tBuCl/GaCl3. 31P[1H] NMR parameters for all derivatives of 8 have been determined by iterative simulation of experimental data. Derivatives 8a[OTf], 8b[OTf], 8c[GaCl4], 8e[OTf], 8f[OTf], and 8g[OTf] and have been characterized by X-ray crystallography, showing the most favorable all-trans configuration of substituents for the phosphine centers, thus minimizing steric interactions. Each derivative adopts a unique envelope or twist conformation of C1 symmetry. The effective C2 symmetry observed for 8b, d, e, f, and g in solution, signified by their 31P[1H] NMR AA'BB'X spin systems, implies a rapid conformational exchange for derivatives of 8. The core frameworks of the cations in the solid state are viewed as snapshots of different conformational isomers within the solution-phase pseudorotation process.  相似文献

Sixteen novel cephalosporins were synthesized by amination of 2,5-dihydroxybenzoic acid derivatives with the aminocephalosporins cefadroxil, cefalexin, cefaclor, and the structurally related carbacephem loracarbef using laccases from Trametes sp. or Myceliophthora thermophila. All products inhibited the growth of several Gram positive bacterial strains in the agar diffusion assay, among them methicillin-resistant Staphylococcus aureus and vancomycin-resistant enterococci. The products protected mice against an infection with Staphylococcus aureus lethal to the control animals. Cytotoxicity and acute toxicity of the new compounds were negligible. The results show the usefulness of laccase for the synthesis of potential new antibiotics. The biological activity of the new compounds stimulates intensified pharmacological tests.  相似文献
