Polyol-induced extraction (PIE) is applied to the extraction of essential oils, using glycerol as a mass separating agent. In 1:1 acetonitrile (ACN)/water solvent mixtures, two immiscible phases can be generated. PIE as an alternative extraction technique was assessed by the extraction of the main flavor and fragrance compounds that comprise six essential oils. In the extraction of eugenol (4-allyl-2-methoxyphenol) from clove buds, the partition coefficients were determined and the % recovery and thermodynamic data in the temperature range of ?20 to 20°C were calculated. The main components present in each essential oil extract were identified through gas chromatography/mass spectrometry (GC/MS) and the compositional profile was compared to traditional extraction techniques. The optimized extraction conditions (?10°C, 1:1 ACN/water (v/v), 20% glycerol) for eugenol at ?10°C give a partition coefficient (KPC) of 87 and an extraction efficiency of 97% in the acetonitrile-rich phase. The eugenol migration to the organic phase is a spontaneous process (ΔG°?=??9.3?kJ/mol) and an endothermic process (ΔH°?=?9.2?kJ/mol) with entropy being the driving force behind the reaction (ΔS°?=?70?J/K, TΔS°?=?18.4?kJ). The technique was applied to five other essential oils (cinnamon bark, caraway seed, spearmint leaf, peppermint leaf, and anise seed oils) with similar results.  相似文献

Leaves of Rubus fruticosus L., Vaccinum myrtillus L., Ribes nigrum L. and Fragaria vesca L. are considered agro-waste of the berry industry, but they can be a rich source of valuable bioactive compounds used in cosmetic industry. In this study, kombucha-fermented and non-fermented extracts were compared in terms of chemical composition and biological activity. Polyphenol compounds were identified by HPLC/DAD/ESI-MS. The antioxidant potential was analyzed by evaluating the scavenging of intracellular free radicals contained in keratinocytes and fibroblasts and by DPPH and ABTS assay, obtaining a higher radical scavenging capacity for the ferments, especially for R. fruticosus and V. myrtillus ferments. Assessment of the cytotoxicity on skin cell lines showed their positive effect on the viability of fibroblasts and keratinocytes (especially for the ferments after 10 days of fermentation). The potential anti-ageing properties were determined by their ability to inhibit the activity of metalloproteinases, obtaining almost 30% inhibition of collagenase and elastase in the case of fermented V. myrtillus. Moreover, when the samples were applied to the skin, the positive effect of ferments on skin hydration and pH was demonstrated, which indicates that kombucha berry leaf extracts may be an innovative cosmetic ingredient.  相似文献

The condensation of bacteriophage phi29 genomic DNA into its preformed procapsid requires the DNA packaging motor, which is the strongest known biological motor. The packaging motor is an intricate ring-shaped protein/RNA complex, and its function requires an RNA component called packaging RNA (pRNA). Current structural information on pRNA is limited, which hinders studies of motor function. Here, we used site-directed spin labeling to map the conformation of a pRNA three-way junction that bridges binding sites for the motor ATPase and the procapsid. The studies were carried out on a pRNA dimer, which is the simplest ring-shaped pRNA complex and serves as a functional intermediate during motor assembly. Using a nucleotide-independent labeling scheme, stable nitroxide radicals were attached to eight specific pRNA sites without perturbing RNA folding and dimer formation, and a total of 17 internitroxide distances spanning the three-way junction were measured using Double Electron-Electron Resonance spectroscopy. The measured distances, together with steric chemical constraints, were used to select 3662 viable three-way junction models from a pool of 65 billion. The results reveal a similar conformation among the viable models, with two of the helices (H(T) and H(L)) adopting an acute bend. This is in contrast to a recently reported pRNA tetramer crystal structure, in which H(T) and H(L) stack onto each other linearly. The studies establish a new method for mapping global structures of complex RNA molecules, and provide information on pRNA conformation that aids investigations of phi29 packaging motor and developments of pRNA-based nanomedicine and nanomaterial.  相似文献

Herein, a study of the plasma electrolytic oxidation (PEO) of niobium in an anodising bath composed of potassium silicate (K2SiO3) and potassium hydroxide (KOH) is reported. The effects of the K2SiO3 concentration in the bath and the process voltage on the characteristics of the obtained oxide layers were assessed. Compact, barrier-type oxide layers were obtained when the process voltage did not exceed the breakdown potential of the oxide layer. When this threshold was breached, the morphology of the oxide layer changed markedly, which is typical of PEO. A significant amount of silicon, in the form of amorphous silica, was incorporated into the oxide coatings under these conditions compared with the amount obtained with conventional anodising. This surface modification technique led to an improvement in the corrosion resistance of niobium in Ringer’s solution, regardless of the imposed process conditions.  相似文献
