Jack bean urease was immobilized on polyethylenimine-coated cotton cloth by adsorption following by crosslinking with dimethyl suberimidate. Of the various methods used, crosslinking with dimethyl suberimidate was found to stabilize the enzyme with minimal inactivation. Cloth-bound urease showed a shift in pH optimum towards the acidic side without appreciable change in temperature optimum and thermostability. Cloth-bound urease could be used repeatedly for urea hydrolysis without appreciable loss in activity. Alternatively, urease cloth could be stitched in the form of a bag containing ammonia adsorbent and used for urea hydrolysis with simultaneous removal of ammonia.  相似文献

Studies have shown that the increase of cell metabolism depends on the low level laser therapy (LLLT) parameters used to irradiate the cells. However, the optimal laser dose to up-regulate pulp cell activity remains unknown. Consequently, the aim of this study was to evaluate the metabolic response of odontoblast-like cells (MDPC-23) exposed to different LLLT doses. Cells at 20000 cells/cm2 were seeded in 24-well plates using plain culture medium (DMEM) and were incubated in a humidified incubator with 5% CO2 at 37°C. After 24 h, the culture medium was replaced by fresh DMEM supplemented with 5% (stress by nutritional deficit) or 10% fetal bovine serum (FBS). The cells were exposed to different laser doses from a near infrared diode laser prototype designed to provide a uniform irradiation of the wells. The experimental groups were: G1: 1.5 J/cm2 + 5% FBS; G2: 1.5 J/cm2 + 10% FBS; G3: 5 J/cm2 + 5% FBS; G4: 5 J/cm2 + 10% FBS; G5: 19 J/cm2 + 5% FBS; G6: 19 J/cm2 + 10% FBS. LLLT was performed in 3 consecutive irradiation cycles with a 24-hour interval. Non-irradiated cells cultured in DMEM supplemented with either 5 or 10% FBS served as control groups. The analysis of the metabolic response was performed by the MTT assay 3 h after the last irradiation. G1 presented an increase in SDH enzyme activity and differed significantly (Mann-Whitney test, p < 0.05) from the other groups. Analysis by scanning electron microscopy showed normal cell morphology in all groups. Under the tested conditions, LLLT stimulated the metabolic activity of MDPC-23 cultured in DMEM supplemented with 5% FBS and exposed to a laser dose of 1.5 J/cm2. These findings are relevant for further studies on the action of near infrared lasers on cells with odontoblast phenotype.  相似文献

Morphological transformation during evaporation-induced self-assembly of a mixed colloidal suspension in micrometric droplets has been investigated. It has been demonstrated that a buckling-driven shape transition of drying droplets of mixed colloidal suspension takes place during evaporation-induced self-assembly. Further, it is also shown that the distortion modulations get significantly amplified with enhancement in volume fraction of anisotropic soft colloidal component of the mixed colloids. It has been argued that the reduction in elastic modulus of formed shell, at the boundary of a drying droplet, and the anisotropic nature of one of the colloidal components facilitate the deformation process. Hierarchical structures of these assembled colloidal grains have been probed using electron microscopy and scattering techniques.  相似文献

The conventional treatments of effluents containing heavy metals produce significant quantities of byproducts with recalcitrant characteristics, making necessary looking after alternative techniques in order to avoid the production of new contaminated residues. Sorption process of chromium and zinc in vertical columns loaded with sewage sludge and organic solid waste has been studied in this work. The data from the TG curves of the two sorbents presented significant differences when they were submitted to the metal uptake, being noticed the displacement of the thermal events towards lower temperatures for both types of sorbents studied. As it was expected, for both sorbents, an increase in the mass of samples has been observed at the completion of the thermal tests upon metal uptake. Therefore, these facts demonstrate that during the biosorption process a physico-chemical interaction took place between sorbents and metals, as it was evidenced by the more than 100 K increase in the decomposition temperatures as well as the variation of the ΔH values of the samples.  相似文献

Male mitotic and meiotic chromosomes of two species of the genus Deltochilum (Scarabaeidae) were analyzed through conventional staining, C-banding, base-specific fluorochromes, silver nitrate staining (AgNO3) and FISH (45S rDNA). The two species possessed karyotypes with 2n = 14, neo-XY and meta-submetacentric chromosomes. The analysis of constitutive heterochromatin (CH) revealed mainly diphasic chromosomes in the two species, showing heterochromatic long arms. Silver nitrate staining labeled the blocks corresponding to CH in D. (Deltohyboma) aff morbillosum while in D. (Deltohyboma) calcaratum, AgNO3 staining revealed only the CH blocks of the diphasic autosomes. The fluorochrome staining revealed in D. (D.) calcaratum the diphasic autosomes and the sex chromosomes with CMA3+ blocks, and in D. (D.) aff morbillosum, the GC-rich sequences were restricted to the terminal regions of the long arms of the pairs 1 and 2 and the X. The FISH revealed 45S rDNA sites in two autosomic pairs and in the X chromosome. The analyses performed allowed for the identification of cytogenetic markers and the discussion of possible chromosome rearrangements that have been involved in the karyotypic differentiation of these species mainly related to the repetitive genome.  相似文献

The increasing need for renewable energy resources has led to higher demands for biofuel, a scenario where the castor bean (Ricinus communis L.) seed oil represents a promising source of raw material. Despite that, information regarding the genome organization of R. communis is still scarce, impairing the application of modern biotechnological and breeding procedures. The present work brings the first evaluation of the mitotic chromosomes of this species, including 10 potentially interesting accessions for cultivation in semi-arid environments aiming at the biofuel production. The approach included standard staining, fluorochrome staining (CMA/DAPI), fluorescent in situ hybridization (FISH) with rDNA 5S and 45S, as well as silver impregnation. All accessions were diploid with 2n = 2x = 20, displaying mainly metacentric chromosomes, with CMA-positive bands (GC-rich) in all pairs of the complement. After silver impregnation, one to 14 nucleoli were observed, while the FISH with rDNA 45S revealed two large sites and a variety of minor dots, and the DNAr 5S hybridized in a single pair. The observed features were discussed and compared with literature data regarding pachytene bivalents.  相似文献

In the second part of our study, we introduce the concept of global extended exactness of penalty and augmented Lagrangian functions, and derive the localization principle in the extended form. The main idea behind the extended exactness consists in an extension of the original constrained optimization problem by adding some extra variables, and then construction of a penalty/augmented Lagrangian function for the extended problem. This approach allows one to design extended penalty/augmented Lagrangian functions having some useful properties (such as smoothness), which their counterparts for the original problem might not possess. In turn, the global exactness of such extended merit functions can be easily proved with the use of the localization principle presented in this paper, which reduces the study of global exactness to a local analysis of a merit function based on sufficient optimality conditions and constraint qualifications. We utilize the localization principle in order to obtain simple necessary and sufficient conditions for the global exactness of the extended penalty function introduced by Huyer and Neumaier, and in order to construct a globally exact continuously differentiable augmented Lagrangian function for nonlinear semidefinite programming problems.  相似文献
