The effects of fatty acids, octanoic acid, (2E, 4E)-hexa-2,4-dienoic acid, hexanoic acid, (2E)-but-2-enoic acid, and butyric acid on the activities of mushroom tyrosinase have been investigated. The results showed that the fatty acids can potently inhibit both monophenolase activity and diphenolase activity of tyrosinase, and that the unsaturated fatty acids exhibited stronger inhibitory effect against tyrosinase than the corresponding saturated fatty acids, and the inhibitory effects were enhanced with the extendability of the fatty acid chain. For the monophenolase activity, the fatty acids could not only lengthen the lag period, but also decrease the steady-state activities. For the diphenolase activity, fatty acids displayed reversible inhibition. Kinetic analyses showed that octanoic acid and hexanoic acid were mixed-type inhibitors and (2E,4E)-hexa-2,4-dienoic acid and (2E)-but-2-enoic acid were noncompetitive inhibitors. The inhibition constants have been determined and compared.  相似文献

A strategy based on chemical markers’ fishing and knockout has been proposed for holistic activity and interaction evaluation of the bioactive components in herbal medicines (HMs). It was devised to screen bioactive-compound group that represents the efficacy of HM, estimate the bioactivity contribution of each component and elucidate the interactions of multi-components. This strategy was accomplished through the following steps: (1) screen out the chemical markers (target peaks) in a HM fingerprint using online two-dimensional turbulent flow chromatography/liquid chromatography–mass spectrometry technique, (2) fish target peaks and knockout any interested peak, and (3) evaluate the bioactivities of fishing and knockout portions. After comparison of the bioactivities of samples containing different target peaks, the efficacy of target-peak group, bioactivity contribution of each compound, and the interactions of multi-components are elucidated. Using Acetylcholinesterase (AChE) and Bulbs of Lycoris radiata (L. Herit.) Herb. (BLR) as the experimental materials, four target peaks were screened out as the AChE binders. By target peaks’ fishing and knockout, combined with activity evaluation, we observed that the bioactivity of the four-peak mixture is similar with the global bioactivity of BLR extract, and there are significant suppressive actions among these four target peaks. These results indicate that this proposed strategy is a useful approach for holistic screening of bioactive-compound group and elucidation of the multi-component interactions in HM.  相似文献

A novel hypoxanthine (Hx) microsensor was constructed. In this work, Nafion xanthine oxidase (XOD) and Au colloid were immobilized onto the surface of a Pt microelectrode. The enzyme biosensor displayed a quick and sensitive response to Hx. Under physiological conditions, a low detection limit, with high selectivity and sensitivity for Hx determination were obtained. The oxidation current [investigated using current-time (I-t) plots] was linear with Hx concentration ranging from 2.0 x 10(-7) to 2.0 x 10(-5) mol L(-1) with a calculated detection limit of 1.0 x 10(-7) mol L(-1) (S/N of 3). The biosensor should be promising for in vivo measurement of Hx without interferences and fouling. The change of Hx concentration in cardiac myocytes stimulated by L-arginine (L-Arg) and acetylcholine (Ach) was also studied.  相似文献

Amphiphilic copolymers of 2-acrylamido-2-methyl-1-propane sulfonic acid (AMPS) with dihydrocholesteryl acrylate (pDHCHAt) or tert-octyl acrylamide (ptOAm) as comonomers were synthesized, characterized and used as new polymeric pseudostationary phases in electrokinetic chromatography. High-efficiency separations with theoretical plates around 500,000 to 870,000/m were achieved. Linear solvation energy relationships analysis of 20 solutes was performed to investigate the retention mechanism. Polycyclic aromatic hydrocarbons (PAHs) were separated under 30% v/v acetonitrile. The separation of nine aromatic model solutes by pDHCHAt showed significantly different selectivity from that of sodium dodecyl sulfate. Although only minor difference in selectivity was found for small aromatic compounds on pDHCHAt and poly(AMPS-lauryl acrylate), significantly different retention mechanism was found for PAHs and n-dodecanophenone comparing both copolymers. Significant chemical selectivity difference was found for selected small aromatic compounds on ptOAm and poly(AMPS-stearyl acrylamide).  相似文献

This paper describes a highly enantioselective epoxidation of alpha,beta-unsaturated esters using the fructose-derived ketone 2 as catalyst and Oxone as oxidant. High ee's have been obtained for a number of trans and trisubstituted substrates (82-98% ee). The results described show that it is feasible for dioxiranes to effectively epoxidize electron-deficient olefins with high ee's.  相似文献

A series of KF/Al2O3 catalyzed Michael-addition reactions between malononitrile and α,β-unsaturated cycloketones in DMF solution were studied. At room temperature, 2-cyano-3-aryl-3-(1,2,3,4-tetrahydronaphthalen-1-one-2-yl) propionitrile derivatives were synthesized by the reaction between 2-arylmethylidene-1,2,3,4-tetra-hydronaphthalen-1-one and malononitrile. However, if the temperature was increased to 80℃, 2-amino-3-cyano-4-aryl-4H-benzo[h]chromene derivatives were obtained in high yields. When the α,β-unsaturated ketones were replaced by 2,6-biarylmethylidenecyclohexanone or 2,5-biarylmethylidenecyclopentanone, another series of 2-amino-3-cyano-4H-pyran derivatives was isolated successfully. The structures of the products were confirmed by X-ray diffraction analysis.  相似文献

The dinuclear copper(II) cryptate [Cu2L](ClO4)4 (1) cleaves the C-C bond of acetonitrile at room temperature to produce a cyanide bridged complex of [Cu2L(CN)](ClO4)3.2CH3CN.4H2O (2). A new ursane‐type nortriterpenoid, adenanthusone (=(11α,12α)‐4‐demethyl‐11,12‐epoxy‐3,13‐dihydroxy‐2‐oxoursa‐3,20(30)‐diene‐28‐oic acid γ‐lactone; 1 ) was isolated from Isodon adenanthus. Its structure was determined by NMR spectra and X‐ray crystallographic diffraction analysis. The biogenetic implication of the nortriterpene is discussed.  相似文献
