The elimination of H2O from serine residues in peptides was found to be feasible by the following two steps: O-acylation with AcCl/pyrideine in CH2Cl2 and treatment with an imidine base (DBU or DBN) in THF in the presence of large amounts of LiClO4 (10 examples). Other Li salts such as LiBr and LiCl can also be used. No epimerization of the amino acid residues in te peptide could be detected under these conditions. Thus, a simple method for the preparation of peptides with dehydroalanine residues in high yield, directly from serine-containing precursors, is available. 相似文献

Computing energies of electronically metastable resonance states is still a great challenge. Both scattering techniques and quantum chemistry based L2 methods are very time consuming. Here we investigate two more economical extrapolation methods. Extrapolating bound states energies into the metastable region using increased nuclear charges has been suggested almost 20 years ago. We critically evaluate this attractive technique employing our complex absorbing potential/Green's function method that allows us to follow a bound state into the continuum. Using the (2)Pi(g) resonance of N2- and the (2)Pi(u) resonance of CO2- as examples, we found that the extrapolation works suprisingly well. The second extrapolation method involves increasing of bond lengths until the sought resonance becomes stable. The keystone is to extrapolate the attachment energy and not the total energy of the system. This method has the great advantage that the whole potential energy curve is obtained with quite good accuracy by the extrapolation. Limitations of the two techniques are discussed. 相似文献

The rf-transmission between two antennas coupling capacitively to a 2-dimensional electron gas at a GaAs-AlGaAs heterojunction is investigated as a function of magnetic field <6 T and frequency <600 MHz at a temperatureT~1.5 K. The collision behaviour of droplets and the collision outcome are investigated for high viscous polymer solutions. For that purpose, two droplet chains produced by piezoelectric droplet generators are directed towards each other at a certain angle so that individual droplet pairs collide. For recording the collision event, one double-image and one high-speed CCD camera were used. One camera is positioned perpendicular to the collision plane recording the outcome of the collision, and the second camera is aligned parallel to the collision plane to assure that the droplet chains are exactly in one plane. A new approach for tracking droplets in combination with an extended particle tracking velocimetry algorithm has been developed. Time-resolved series of pictures were used to analyse the dynamics of droplet collisions. The three different water soluble substances were saccharose and 1-Ethenyl-2-pyrrolidone (PVP) with different molecular weights (K17, K30). The solvent was demineralised water. The solids contents ranged from 20 to 60 %, 5 to 25 % and 5 to 35 %, yielding dynamic viscosities in the range of 2–60 mPa s. Results were collected for different pairs of impact angles and Weber numbers in order to establish common collision maps for characterising the outcomes. Here, relative velocities between 0.5 and 4 m/s and impact parameters in the interval from 0 to 1 for equal-sized droplets (Δ = 1) have been investigated. Additionally, satellite formation will be discussed exemplarily for K30. A comparison with common models of different authors (Ashgriz and Poo in J Fluid Mech 221:183–204, 1990; Estrade et al. in Int J Heat Fluid Flow 20:486–491, 1999) mainly derived for low viscous droplets revealed that the upper limit of their validity is given by an Ohnesorge number of Oh = 0.115 and a capillary number of Ca = 0.577. For higher values of these non-dimensional parameters and hence higher dynamic viscosities, these models are unable to predict correctly the boundaries between collision scenarios. The model proposed by Jiang et al. (J Fluid Mech 234:171–190, 1992), which includes viscous dissipation, is able to predict the boundary between coalescence and stretching separation for higher viscosities (i.e. Oh > 0.115 and Ca > 0.577). However, the model constants are not identical for different solution properties. As a conclusion, an alteration of the collision appearance takes place because of the relative importance between surface tension and viscosity. 相似文献

Single-stage and multi-stage strategies have been evaluated and compared for the purification of human antibodies using liquid–liquid extraction in aqueous two-phase systems (ATPSs) composed of polyethylene glycol 3350 (PEG 3350), dextran, and triethylene glycol diglutaric acid (TEG-COOH). The performance of single-stage extraction systems was firstly investigated by studying the effect of pH, TEG-COOH concentration and volume ratio on the partitioning of the different components of a Chinese hamster ovary (CHO) cells supernatant. It was observed that lower pH values and high TEG-COOH concentrations favoured the selective extraction of human immunoglobulin G (IgG) to the PEG-rich phase. Higher recovery yields, purities and percentage of contaminants removal were always achieved in the presence of the ligand, TEG-COOH. The extraction of IgG could be enhanced using higher volume ratios, however with a significant decrease in both purity and percentage of contaminants removal. The best single-stage extraction conditions were achieved for an ATPS containing 1.3% (w/w) TEG-COOH with a volume ratio of 2.2, which allowed the recovery of 96% of IgG in the PEG-rich phase with a final IgG concentration of 0.21 mg/mL, a protein purity of 87% and a total purity of 43%. In order to enhance simultaneously both recovery yield and purity, a four stage cross-current operation was simulated and the corresponding liquid–liquid equilibrium (LLE) data determined. A predicted optimised scheme of a counter-current multi-stage aqueous two-phase extraction was hence described. IgG can be purified in the PEG-rich top phase with a final recovery yield of 95%, a final concentration of 1.04 mg/mL and a protein purity of 93%, if a PEG/dextran ATPS containing 1.3% (w/w) TEG-COOH, 5 stages and volume ratio of 0.4 are used. Moreover, according to the LLE data of all CHO cells supernatant components, it was possible to observe that most of the cells supernatant contaminants can be removed during this extraction step leading to a final total purity of about 85%. 相似文献

Nanofluids, comprising Y2O3-stabilized (partially; 3?mol%) ZrO2 nanoparticles (up to 2?vol%) dispersed in water and ethylene glycol, were developed. A suitable surfactant and stirring routine ensured uniformity and stability of dispersion. Thermal conductivity ratio of the nanofluids, measured using an indigenously developed thermal comparator device, showed a significant increase of up to 100% with only 1.5?vol% nanoparticles. Furthermore, it was analytically shown that shape factor and aspect ratio significantly influence the effective thermal conductivity ratio of nanofluids with comparable vol% nanoparticles. The study demonstrated that a nanofluid of nano-ZrO2 dispersed in water and ethylene glycol could be usefully applied in closed-loop single-phase heat transfer conditions, such as in refrigerators or microchannels. 相似文献

Methanol is a natural ingredient with major occurrence in fruit spirits, such as apple, pear, plum or cherry spirits, but also in spirits made from coffee pulp. The compound is formed during fermentation and the following mash storage by enzymatic hydrolysis of naturally present pectins. Methanol is toxic above certain threshold levels and legal limits have been set in most jurisdictions. Therefore, the methanol content needs to be mitigated and its level must be controlled. This article will review the several factors that influence the methanol content including the pH value of the mash, the addition of various yeast and enzyme preparations, fermentation temperature, mash storage, and most importantly the raw material quality and hygiene. From all these mitigation possibilities, lowering the pH value and the use of cultured yeasts when mashing fruit substances is already common as best practice today. Also a controlled yeast fermentation at acidic pH facilitates not only reduced methanol formation, but ultimately also leads to quality benefits of the distillate.
