Proteins separated by two-dimensional (2-D) gel electrophoresis can be visualized using various protein staining methods. This is followed by downstream procedures, such as image analysis, gel spot cutting, protein digestion, and mass spectrometry (MS), to characterize protein expression profiles within cells, tissues, organisms, or body fluids. Characterizing specific post-translational modifications on proteins using MS of peptide fragments is difficult and labor-intensive. Recently, specific staining methods have been developed and merged into the 2-D gel platform so that not only general protein patterns but also patterns of phosphorylated and glycosylated proteins can be obtained. We used the new Pro-Q Diamond phosphoprotein dye technology for the fluorescent detection of phosphoproteins directly in 2-D gels of mouse leukocyte proteins, and Pro-Q Emerald 488 glycoprotein dye to detect glycoproteins. These two fluorescent stains are compatible with general protein stains, such as SYPRO Ruby stain. We devised a sequential procedure using Pro-Q Diamond (phosphoprotein), followed by Pro-Q Emerald 488 (glycoprotein), followed by SYPRO Ruby stain (general protein stain), and finally silver stain for total protein profile. This multiple staining of the proteins in a single gel provided parallel determination of protein expression and preliminary characterization of post-translational modifications of proteins in individual spots on 2-D gels. Although this method does not provide the same degree of certainty as traditional MS methods of characterizing post-translational modifications, it is much simpler, faster, and does not require sophisticated equipment and expertise in MS.  相似文献

Nimbolide [systematic name: (4α,5α,6α,7α,15β,17α)‐7,15:21,23‐diepoxy‐6‐hydroxy‐4,8‐dimethyl‐1‐oxo‐18,24‐dinor‐11,12‐secochola‐2,13,20,22‐tetraene‐4,11‐dicarboxylic acid γ‐lactone methyl ester], C27H30O7, was isolated from the leaves of Azadirachta indica, and its isomer, isonimbolide [systematic name: (4α,5α,6α,7α,15α)‐7,15:21,23‐diepoxy‐6‐hydroxy‐4,8‐dimethyl‐1‐oxo‐18,24‐dinor‐11,12‐secochola‐2,16,20,22‐tetra­ene‐4,11‐dicarboxylic acid γ‐lactone methyl ester], was prepared from a novel rearrangement reaction of nimbolide, using boron trifluoride etherate and tetra­butyl­ammonium bromide. The reaction conditions are probably responsible for the ether cleavage, double‐bond rearrangement and reformation of the ether linkage. As a result, there are conformational changes in two cyclo­pentane rings and the side‐chain –CH2COOMe group. In isonimbolide, an (24) hydrogen‐bond motif is observed.  相似文献

Summary Volatilization of arsenic, selenium and antimony for sample introduction in atomic absorption spectrometry has been performed by pumping an acidic sample through an anion exchanger in the tetrahydroborate (III) form packed as a bed in the liquid channel of a gas-liquid separation membrane cell. The hydrides generated in the heterogeneous reaction between bound tetrahydroborate (III) ions and the analytes are rapidly transferred with the aid of the concomitantly generated hydrogen gas through the gas-permeable membrane into the gas phase and swept to the spectrometer by an additional hydrogen gas flow. This instant transfer of the hydrides to the gas phase kinetically discriminates the reaction of the hydride with metal borides and metal colloids, whose formation by reaction with tetra-hydroborate (III) is slower than the hydride reaction. The susceptibility to interference by transition metal ions is thus markedly reduced, as compared with both batch hydride generation methods and a previously presented heterogeneous reaction scheme. The detection limits for arsenic, selenium, and antimony were 1.2, 3.7, and 10 g/l, respectively. The calibration graphs were linear from the detection limit up to 125 g/l for arsenic, 150 g/l for selenium, and 250 g/l for antimony. The relative standard deviations at concentration levels of 10 and 100 g/l were 1.8 and 0.7% for arsenic and 2.3 and 1.2% for selenium. Corresponding figures for 50 and 100 g/l antimony were 2.5 and 1.6%.  相似文献

Reaction of 4-amino-2-fluoropyridine (2-F-4-AP) with copper halides produced the neutral coordination complexes: (2-F-4-AP)2CuX2 (X = Cl(1), Br(2)). 1 crystallizes in the orthorhombic space group Pccn in a distorted square planar geometry. Magnetic susceptibility data were fit to the uniform chain Heisenberg model resulting in C = 0.439(6)emu-K/mole-Oe and J = ?28(1) K. 2 crystallizes in the monoclinic space group C2/m and is closer to distorted tetrahedral. Intermolecular Br?Cu contacts generate a square layer. Magnetic data show very weak ferromagnetic interactions [C = 0.42(1)emu-K/mol-Oe, J = 0.71(2) K]. Similarly, reaction of 2-F-4-AP with copper halides and aqueous HX in alcohol solvents produced the salts (2-F-4-APH)2CuX4 (X = Cl(3), Br(4)). 3 crystallizes in the triclinic space group P-1. Crystal packing reveals short Cl?Cl contacts which generate a structural ladder. However, analysis of the magnetic data suggests that only the rails of the ladder produce a viable magnetic superexchange pathway; the uniform Heisenberg chain model provides C = 0.449(1)emu-K/mol-Oe and J = -6.9(1) K. 4 is isostructural and is also best fit by a chain model [J = ?2.7(4) K]. The brominated complex (2-F-3-Br-4-APH)2CuBr4·2H2O, 5, (2-F-3-Br-4-APH = 4-amino-3-bromo-2-fluoropyridinium) was serendipitously produced as a byproduct of the synthesis of 4 and was characterized by single-crystal X-ray diffraction.  相似文献

Medicinal plants have been traditionally used to treat cancer in Ethiopia. However, very few studies have reported the in vitro anticancer activities of medicinal plants that are collected from different agro-ecological zones of Ethiopia. Hence, the main aim of this study was to screen the cytotoxic activities of 80% methanol extracts of 22 plants against human peripheral blood mononuclear cells (PBMCs), as well as human breast (MCF-7), lung (A427), bladder (RT-4), and cervical (SiSo) cancer cell lines. Active extracts were further screened against human large cell lung carcinoma (LCLC-103H), pancreatic cancer (DAN-G), ovarian cancer (A2780), and squamous cell carcinoma of the esophagus (KYSE-70) by using the crystal violet cell proliferation assay, while the vitality of the acute myeloid leukemia (HL-60) and histiocytic lymphoma (U-937) cell lines was monitored in the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) microtiter assay. Euphorbia schimperiana, Acokanthera schimperi, Kniphofia foliosa, and Kalanchoe petitiana exhibited potent antiproliferative activity against A427, RT-4, MCF-7, and SiSo cell lines, with IC50 values ranging from 1.85 ± 0.44 to 17.8 ± 2.31 µg/mL. Furthermore, these four extracts also showed potent antiproliferative activities against LCLC-103H, DAN-G, A2780, KYSE-70, HL-60, and U-937 cell lines, with IC50 values ranging from 0.086 to 27.06 ± 10.8 µg/mL. Hence, further studies focusing on bio-assay-guided isolation and structural elucidation of active cytotoxic compounds from these plants are warranted.  相似文献

In this paper we propose a framework for dynamic routing systems based on their degree of dynamism. Next, we consider its impact on solution methodology and quality. Specifically, we introduce the Partially Dynamic Travelling Repairman Problem and describe several dynamic policies to minimize routing costs. The results of our computational study indicate that increasing the dynamic level results in a linear increase in route length for all policies studied. Furthermore, a Nearest Neighbour policy performed, on the average, uniformly better than the other dispatching rules studied. Among these, a Partitioning policy produced only slightly higher average route lengths.  相似文献

We have studied the enhancement of spontaneous emission rates for InAs quantum dots embedded in GaAs microdisks in a time-resolved photoluminescence experiment. Inhomogeneous broadening of the quantum dot energy levels and random spatial distribution of the quantum dots in a microdisk lead to a broad distribution of the spontaneous emission rates. Using a nonnegative least-norm algorithm, we extract the distribution of spontaneous emission rates from the temporal decay of emission intensity. The maximum spontaneous emission enhancement factor exceeds 10.  相似文献

By using Monte Carlo (MC) methods we study the deconfining transition of QCD with color group SU(3) at finite temperature. The fermion polarization effects are kept in account by implementing the pseudo-fermion method. We use two different lattice volumes (63 × 4, 83 × 4) to check finite size effects. Even in the presence of light quarks we find a discontinuous transition. The existence of this transition is observed both creating metastable states and using mixed phase runs.  相似文献
