HIV-1 cell entry is mediated by sequential interactions of the envelope protein gp120 with the receptor CD4 and a coreceptor, usually CCR5 or CXCR4, depending on the individual virion. Considerable efforts on exploiting the HIV coreceptors as drug targets have led to the new class of coreceptor antagonists. While these antiretroviral drugs aim at preventing virus/coreceptor interaction by binding to host proteins, neutralizing antibodies directed against the coreceptor-binding sites on gp120 have attracted attention as possible vaccine candidates. However, both approaches are complicated by the multiple protective mechanisms of gp120 which allow for rapid escape from selective pressures exerted by drugs or antibodies. Thus, advances in rational drug and vaccine design rely heavily on improved insights into the relation between genotype and phenotype, the evolution of coreceptor usage, and, ultimately the structural biology of coreceptor usage and inhibition. The third variable (V3) loop of gp120, crucially involved in all these aspects, will be a major focus of this review.  相似文献

Whereas most conventional DNA probes are flat disklike aromatic molecules, we explored the possibility of developing quadruplex sensors with nonplanar conformations, in particular, the propeller‐shaped tetraphenylethene (TPE) salts with aggregation‐induced emission (AIE) characteristics. 1,1,2,2‐Tetrakis[4‐(2‐triethylammonioethoxy)phenyl]ethene tetrabromide (TPE‐ 1 ) was found to show a specific affinity to a particular quadruplex structure formed by a human telomeric DNA strand in the presence of K+ ions, as indicated by the enhanced and bathochromically shifted emission of the AIE fluorogen. Steady‐state and time‐resolved spectral analyses revealed that the specific binding stems from a structural matching between the AIE fluorogen and the DNA strand in the folding process. Computational modeling suggests that the AIE molecule docks on the grooves of the quadruplex surface with the aid of electrostatic attraction. The binding preference of TPE‐ 1 enables it to serve as a bioprobe for direct monitoring of cation‐driven conformational transitions between the quadruplexes of various conformations, a job unachievable by the traditional G‐quadruplex biosensors. Methyl thiazolyl tetrazolium (MTT) assays reveal that TPE‐ 1 is cytocompatible, posing no toxicity to living cells.  相似文献

We synthesized a group of silole regioisomers 1(x,y), whose photoluminescence varied dramatically with its regiostructure. By internally hindering the intramolecular rotation, we succeeded in creating a novel silole (1(3,4)) that is strongly luminescent in solutions and whose fluorescence quantum yield in acetone is as high as 83%. We revealed that 1(3,4) was a sensitive chemosensor capable of optically discriminating nitroaromatic regioisomers of p-, o-, and m-nitroanilines. Against general belief, crystal formation of 1(2,4) blue-shifted its emission color and boosted its emission efficiency. The light-emitting diode based on the crystal of 1(2,4) emitted a strong blue light (464 nm) in a high current efficiency (5.86 cd/A).  相似文献

New hyperbranched polyarylenes (HPAs) and linear polyacetylenes (LPAs) containing chromophoric moieties were synthesized in high yields (up to 97%) by coablt- and tantalum-catalyzed polycyclotrimerizations and tungsten-catalyzed metathesis polymerizations, respectively. The polymers possessed high molecular weights (Mw up to 113 000 Da) and were completely soluble in common organic solvents. The HPAs emitted strong UV light of 400 nm in high quantum yields (ΦF up to 0.98) and limited intense laser pulses, whose limiting threshold and signal suppress power were better than those of C60, a well-known optical limiter. The electroluminescence (EL) devices of the LPAs emitted blue light of ∼460 nm and exhibited maximun brightness, current efficiency, and external quantum efficiency of 1118 cd/m2, 1.53 cd/A, and 0.85%, respectively.  相似文献

The United Nations University/International Center for Theoretical Physics Plasma Focus Facility (UNU/ICTP PFF), a small plasma focus, has proven very useful as a training device for research initiation and as a test bed for applications. In this work, its performance in terms of a soft X-ray (SXR) source is examined. The total SXR yield when operated in neon is measured using low-cost detectors, including a calorimeter and a five-channel filtered PIN system. For a charging voltage of 14 kV with 2.9 kJ stored energy, the optimum operating pressure in neon is found to be in the range of 2.7-3.3 torr, resulting in a total SXR yield of 6J/shot into 4π steradians measured by the calorimeter in agreement with the PIN detectors. Spectral data shows that 64% of the total SXR yield is contributed by the Ly-α and He-α lines and 36% by the rest, mainly the radiative recombination. The low total SXR yield (0.2 % of stored energy) is consistent with numerical computations of focus dynamics, which reveals that 3% of stored energy is converted into plasma energy. This low conversion rate into plasma energy is due to the large inductance of 110 nH of this simple single-capacitor device. A new compound with intramolecular charge transfer (ICT) property—5,6-Bis-[4-(naphthalene-1-yl-phenyl-amino)-phenyl]-pyrazine-2,3-dicarbonitrile(BNPPDC) was synthesized. The new compound was strongly fluorescent in non-polar and moderately polar solvents, as well as in thin solid film. The absorption and emission maxima shifted to longer wavelength with increasing solvent polarity. The fluorescence quantum yield also increased with increasing solvent polarity from non-polar to moderately polar solvents, then decreased with further increase of solvent polarity. This indicates both “positive” and “negative” solvatokinetic effects co-existed. Using this material as hole-transporting emitter and host emitter, we fabricated two electroluminescent (EL) devices with structures of A (ITO/BNPPDC (45 nm)/1,3,5-tris(N-phenylbenzimidazol-2-yl)benzene (TPBI) (45 nm)/Mg:Ag (200 nm) and B (ITO/N,N′-diphenyl-N,N′-bis-(3-methylphenyl) (1,1′-diphenyl)4,4′-diamine (TPD) (50 nm)/BNPPDC (20 nm)/1,3,5-tris(N-phenylbenzimidazol-2-yl)benzene (TPBI) (45 nm)/Mg:Ag (200 nm). The devices showed green-yellow EL emission with good efficiency and high brightness. For example, the device A exhibited a high brightness of 17400 cd/m2 at a driving voltage of 11 V and a very low turn-on voltage (2.9 V), as well as a maximum luminous efficiency 3.61 cd/A. The device B showed a similar performance with a high brightness of 12650 cd/m2 at a driving voltage of 13 V and a maximum luminous efficiency 3.62 cd/A. In addition, the EL devices using BNPPDC as a host and 4-(dicyanomethylene)-2-t-butyl-6-(1,1,7,7-tetramethyljulolidyl-9-enyl)-4H-pyran (DCJTB) as a dopant (configuration: ITO/TPD (60 nm)/BNPPDC:DCJTB (2%) (30 nm)/TPBI (35 nm)/Mg:Ag (200 nm)) showed a good performance with a brightness of 150 cd/m2 at 4.5 V, a maximum brightness of 12600 cd/m2 at 11.5 V, and a maximum luminous efficiency of 3.30 cd/A.  相似文献
