The appearance of unexpected peaks in capillary electrophoresis (CE) is common and can lengthen the time of method development as assay conditions and experimental parameters are varied to understand and mitigate the effects of the additional peaks. Additional peaks can arise when a single-analyte zone is split into multiple zones. Understanding the underlying mechanism of these phenomena, recognizing conditions that favor its presence, and knowing how to confirm and eliminate the effect are important for efficient method optimization. In this study, we examine how the overlap of analyte zones with the sample plug can lead to peak splitting. This is explored experimentally using dual detection CE, which enables both the sample plug and analyte zones to be independently and simultaneously measured from the same detection volume. Simulations performed via COMSOL Multiphysics confirm the origin of the splitting and help guide experiments to reduce and eliminate the effect. Our findings show that this peak splitting mechanism can arise in separations of both small and large molecules but is, especially, prevalent in separations of slowly migrating macromolecules. This effect is also more prevalent when using a short length-to-detector, as is commonly found in microfluidic applications. A simple diffusion-less model is introduced to develop strategies for reducing peak splitting that avoids modifying the apparatus, such as by lengthening the separation length, which can be difficult. Decreasing the sample plug length and slowing the electroosmotic flow can both reduce this effect, which is confirmed experimentally. 相似文献

Fourier Transform mid-infrared and Raman spectroscopies were used to investigate the cation/polymer, cation/urea bridge, cation/anion and hydrogen bonding interactions in poly(oxyethylene) (POE)/siloxane di-ureasil networks prepared by the sol–gel route and doped with lithium bis(trifluoromethanesulfonyl)imide (LiTFSI). Materials with compositions 200 ?n ? 5 (where n expresses the molar ratio OCH2CH2/Li+) were studied. The Li+ ions coordinate to the urea carbonyl oxygen atoms over the whole range of salt concentration considered. Bonding to the ether oxygen atoms of the POE chains occurs at n ? 40, although a significant fraction of the POE chains remain non-coordinated. In these high salt content samples, the cations interact with the anions forming contact ion pairs. “Free” ions are probably the main charge carriers at the room temperature conductivity maximum of these ormolytes. 相似文献

Laser Induced Breakdown Spectroscopy (LIBS) is an advanced analytical technique for elemental determination based on direct measurement of optical emission of excited species on a laser induced plasma. In the realm of elemental analysis, LIBS has great potential to accomplish direct analysis independently of physical sample state (solid, liquid or gas). Presently, LIBS has been easily employed for qualitative analysis, nevertheless, in order to perform quantitative analysis, some effort is still required since calibration represents a difficult issue. Artificial neural network (ANN) is a machine learning paradigm inspired on biological nervous systems. Recently, ANNs have been used in many applications and its classification and prediction capabilities are especially useful for spectral analysis. In this paper an ANN was used as calibration strategy for LIBS, aiming Cu determination in soil samples. Spectra of 59 samples from a heterogenic set of reference soil samples and their respective Cu concentration were used for calibration and validation. Simple linear regression (SLR) and wrapper approach were the two strategies employed to select a set of wavelengths for ANN learning. Cross validation was applied, following ANN training, for verification of prediction accuracy. The ANN showed good efficiency for Cu predictions although the features of portable instrumentation employed. The proposed method presented a limit of detection (LOD) of 2.3 mg dm− 3 of Cu and a mean squared error (MSE) of 0.5 for the predictions. 相似文献

The spontaneous formation of vesicles by the salt-free surfactant hexadecyltrimethylammonium octylsulfonate (TASo) and the features of an unusual vesicle-micelle transition are investigated in this work. In a previous work, we have shown that this highly asymmetric catanionic surfactant displays a rare lamellar miscibility gap in the concentrated regime. Here, we analyze in detail the aggregation behavior in the dilute regime (less than 3 wt % surfactant) as a function of both concentration and temperature. The phase diagram is dominated by a two-phase region consisting of a dispersion of a swollen lamellar phase (Lalpha') in the excess solvent phase (L1). Stable vesicles form in this two-phase region, and upon temperature increase, a transition to a single solution phase containing only elongated micelles occurs. The structural characterization of the aggregates and the investigation of their equilibrium properties have been carried out by light microscopy, cryo-TEM, water self-diffusion NMR, and SANS. Similarly to the lamellar-lamellar coexistence, the changes in microstructure at high dilution and high temperature can be understood from solubility differences, electrostatic interactions, and preferred aggregate curvature. Surface charge in the aggregates stems from the higher solubility of the octylsulfonate (So-) ion as compared to that of the hexadecyltrimethylammonium ion (TA+). Upon temperature increase, the ratio of free So(-) relative to the neutral TASo increases. Consequently, the surface charge density of the aggregates increases, and this ultimately induces a transition to a higher-curvature morphology (elongated micelles). Vesicles can also be spontaneously formed by cooling solutions from the micellar region, and the mean size obtained is practically independent of cooling rate, suggesting that dissociation/charge effects also control this process. 相似文献

It is reported the synthesis of magnetic nanoparticles via the chemical co-precipitation of Fe 3+ ions and their preparation by coating them with polyaniline. The electronic micrograph analysis showed that the mean diameter for the nanoparticles is ~15 nm. FTIR, powder X-ray diffraction and Mössbauer spectroscopy were used to understand the chemical, crystallographic and 57Fe hyperfine structures for the two samples. The nanoparticles, which exhibited magnetic behavior with relatively high spontaneous magnetization at room temperature, were identified as being mainly formed by maghemite (γFe2O3). The coated magnetic nanoparticles (sample labeled “mPANI”) presented a real ability to bind biological molecules such as trypsin, forming the magnetic enzyme derivative (sample “mPANIG-Trypsin”). The amount of protein and specific activity of the immobilized trypsin were found to be 13±5 μg of protein/mg of mPANI (49.3 % of immobilized protein) and 24.1±0.7 U/mg of immobilized protein, respectively. After 48 days of storage at 4 °C, the activity of the immobilized trypsin was found to be 89 % of its initial activity. This simple, fast and low-cost procedure was revealed to be a promising way to prepare mPANI nanoparticles if technological applications addressed to covalently link biomolecules are envisaged. This route yields chemically stable derivatives, which can be easily recovered from the reaction mixture with a magnetic field and recyclable reused. 相似文献

Some diuretic substances are controlled and monitored by the World Anti-Doping Agency as prohibited substances for use by athletes, such as triamterene (TRT). Thus, this work describes a voltammetric method based on graphite-epoxy composite electrode modified by tosyl-functionalized magnetic particles (GECE/MPs-To) for determination of TRT diuretic in urine sample. The TRT presented an oxidation peak at +1.24 V at GECE/MPs-To with irreversible behavior. Controlled potential electrolysis of the TRT at +1.26 V indicated the two electrons are transferred during amine group oxidation and the main product was identified by LC-MS/MS. The anodic peak current is 25 % higher at the modified electrode, suggesting that TRT is adsorbed on the magnetic particles. Using optimized conditions by using multivariate optimization of the parameters inherent of the square wave voltammetry, a calibration curve was constructed with a linear relationship for TRT from 0.500 to 99.8 μmol L?1. The limits of detection and quantification were 1.47 and 4.91?×?10?7 mol L?1, respectively. The proposed method was applied to urine sample and validated by LC-MS/MS technique where the values found and compared between the two techniques showed no significant difference at 95 % confidence. 相似文献
