Two new triterpene saponins, albidosides H (1) and I (2), along with the three known saponins were isolated from the barks of Acacia albida. Their structures were elucidated on the basis of extensive 1D- and 2D-NMR studies and mass spectrometry. Albidosides H (1) and I (2) were assayed for their cytotoxicity against HeLa and HL60 cells using MTT method.  相似文献

Mushroom terpenoids are biologically and chemically diverse fungal metabolites. Among them, melleolides are representative sesquiterpenoids with a characteristic protoilludane skeleton. In this study, we applied a recently established hot spot knock-in method to elucidate the biosynthetic pathway leading to 1α-hydroxymelleolide. The biosynthesis of the sesquiterpene core involves the cytochrome P450 catalyzing stepwise hydroxylation of the Δ6-protoilludene framework and a stereochemical inversion process at the C5 position catalyzed by short-chain dehydrogenase/reductase family proteins. The highlight of the biosynthesis is that the flavoprotein Mld7 catalyzes an oxidation-triggered double-bond shift accompanying dehydration and acyl-group-assisted substitution with two different nucleophiles at the C6 position to afford the Δ7-protoilludene derivatives, such as melleolide and armillarivin. The complex reaction mechanism was proposed by DFT calculations. Of particular importance is that product distribution is regulated by interaction with the cell membrane.  相似文献

We prove tail triviality of determinantal point processes \( \mu \) on continuous spaces. Tail triviality has been proved for such processes only on discrete spaces, and hence we have generalized the result to continuous spaces. To do this, we construct tree representations, that is, discrete approximations of determinantal point processes enjoying a determinantal structure. There are many interesting examples of determinantal point processes on continuous spaces such as zero points of the hyperbolic Gaussian analytic function with Bergman kernel, and the thermodynamic limit of eigenvalues of Gaussian random matrices for \(\hbox {Sine}_2 , \hbox {Airy}_2 , \hbox {Bessel}_2 \), and Ginibre point processes. Our main theorem proves all these point processes are tail trivial.  相似文献

Using a modified method developed from Vonk's method, detailed values of crystallinity and crystal disorder were obtained by wide angle X-ray diffraction (WAXD). In Vonk's method, the crystallinity (w) is determined by extrapolation of the WAXD experimental curve back to zero scattering angle, while the distortion factor (k) is determined by the inclination of the experimental curve at zero scattering angle. In our new method, both w and k are determined at the same time by using the least squares method. In order to show the efficiency of our method, the new fitting procedure was applied to the experimental values of polyethylene and polyethylene terephthalate as measured by Vonk, and the values of w and k determined by our new method were compared with those found by Vonk's method. The new fitting method has the advantage that it uses the whole experimental curve. Moreover, our modified Vonk's method enables us to discuss crystal distortions as arising from first-kind (thermal) disorder and second-kind (paracrystalline) disorder.  相似文献

O3‐NaFe1/2Co1/2O2 shows initial capacity of 160 mAh/g and an average operating voltage of 3.1 V (vs. Na) with good cyclability, and is a promising candidate of the cathode materials for sodium‐ion secondary batteries (SIBs). Here, we found that the cyclability of the slowly‐cooled sample is much worse than that of quenched one, even though the former sample keeps the O3‐type structure. The energy dispersive X‐ray spectroscopy (EDX) images suggest that the slow‐cooled sample (Nax Fe1–yCoy O2) is inhomogeneous in the Fe concentration (1 – y), perhaps triggered by the Na deficiency (1 – x). We ascribed the poor cyclability in the slowly‐cooled sample to the concentration inhomogeneity (Δy). Phage display is the most widely used technique to discover de novo peptides that bind to target proteins. However, it is associated with some challenges such as compositional bias. In this study, to overcome these difficulties, we devised a ‘pattern enrichment analysis.’ In this method, two samples (one obtained by affinity selection, the other simply amplified without selection) are prepared, and the two sequence datasets read on next-generation sequencer are compared to find the three-residue pattern most enriched in the selected sample. This allows us to compare two sequence datasets with high coverage and facilitates the identification of peptide sequences and the key residues for binding. We also demonstrated that this approach in the combination with structured peptide libraries allowed spatial mapping of the enriched sequence patterns. Here, we prepared a phage library displaying chemically stapled helical peptides with the X1C2X3X4X5X6X7X8C9X10 sequence, where X is any amino acid. To validate our method, we performed screening against the HDM2 protein. The results showed that the hydrophobic residues (Phe, Tyr, Trp and Leu) that are key to interactions with HDM2 were clearly identified by the pattern enrichment analysis. We also performed selection targeting the SARS-CoV-2 spike RBD in the same manner. The results showed that similar patterns were enriched among the hit peptides that inhibited the protein–protein interaction.

To effectively identify helical peptide binders from a phage library, we developed “pattern enrichment analysis,” which finds the enriched three-residue patterns by comparison with a control sample.  相似文献

The oxidation of [MII(3,5-DTBCat)(DTBbpy)] (M=Ni ( [Ni] ), Pd ( [Pd] ), and Pt ( [Pt] ); 3,5-DTBCat=3,5-di-tert-butylcatecholato; DTBbpy=4,4′-di-tert-butyl-2,2′-bipyridine) afforded the dimeric {[NiII(3,5-DTBSQ)(DTBbpy)](PF6)}2 ( {[Ni](PF6)}2 ; 3,5-DTBSQ=3,5-di-tert-butylsemiquinonato) and monomeric semiquinonato (SQ) complexes [MII(3,5-DTBSQ)(DTBbpy)](PF6) (M=Pd ( [Pd](PF6) ) and Pt ( [Pt](PF6) )). The negative solvatochromic properties of the SQ complexes allowed us to estimate the relative order of their dipole moments: [Pd](PF6) > [Pt](PF6) > {[Ni](PF6)}2 . The complexes [Pd](PF6) and [Pt](PF6) adopt monomeric structures and are stable in CH2Cl2 and toluene, whereas they gradually disproportionate at room temperature to [M] and 3,5-di-tert-butylbenzoquinone (3,5-DTBBQ) in polar solvents such as THF, MeOH, EtOH, DMF, or DMSO. The results of spectroscopic studies suggested that the oxidized nickel complex adopts a monomeric structure ( [Ni](PF6) ) in CH2Cl2, but a dimeric structure ( {[Ni](PF6)}2 ) in the other investigated solvents. In polar solvents, {[Ni](PF6)}2 may disproportionate to [Ni] and 3,5-DTBBQ at 323 K, thereby demonstrating a significant solvent- and metal-dependence in temperature. The relative activities of {[Ni](PF6)}2 and [M](PF6) toward disproportionation are related to the electrochemically estimated Kdis values in CH2Cl2 and DMF. The present work demonstrates that solvent polarity and the dipole moments of the SQ complexes promote disproportionation, which can be controlled by a judicious choice of the metal ion, solvent, and temperature.  相似文献
