KRAS homo-dimerization has been implicated in the activation of RAF kinases, however, the mechanism and structural basis remain elusive. We developed a system to study KRAS dimerization on nanodiscs using paramagnetic relaxation enhancement (PRE) NMR spectroscopy, and determined distinct structures of membrane-anchored KRAS dimers in the active GTP- and inactive GDP-loaded states. Both dimerize through an α4–α5 interface, but the relative orientation of the protomers and their contacts differ substantially. Dimerization of KRAS-GTP, stabilized by electrostatic interactions between R135 and E168, favors an orientation on the membrane that promotes accessibility of the effector-binding site. Remarkably, “cross”-dimerization between GTP- and GDP-bound KRAS molecules is unfavorable. These models provide a platform to elucidate the structural basis of RAF activation by RAS and to develop inhibitors that can disrupt the KRAS dimerization. The methodology is applicable to many other farnesylated small GTPases.  相似文献

A detailed exploration of the synthesis of (?)‐morphine based on sequential [3,3]‐sigmatropic rearrangements is described. The sequential Claisen/Claisen rearrangements of an allylic vicinal diol resulted in the stereoselective formation of the two contiguous carbon centers, including a sterically encumbered quaternary carbon, in a single operation. The two ethyl esters generated in this reaction were successfully differentiated during a subsequent Friedel–Crafts‐type cyclization. The (?)‐morphine double bond was introduced at a late stage in our first‐generation synthesis, but was formed at an earlier stage in the second‐generation synthesis, resulting in a more efficient route to the end product.  相似文献

Graft copolymers of poly(tulipalin A) (PT) and poly(DL‐lactide) (PDLLA) (PT‐g‐PDLLA) having various graft lengths and ratios were synthesized by free‐radical copolymerization of α‐methylene‐γ‐butyrolactone (MBL) and PDLLA macromonomers (HEMA‐PDLLA) terminated by 2‐hydroxyethyl methacrylate (HEMA)‐terminated. HEMA‐PDLLA were synthesized by ring opening polymerization (ROP) of DL‐lactide in the presence of HEMA. Both HEMA‐PDLLA and the copolymers were characterized by NMR spectroscopy and gel permeation chromatography (GPC). The thermal properties of the graft copolymers were found to depend on the graft length and the ratio. The copolymers consisting of PDLLA side chains of Mn = 500 Da showed a single Tg between Tgs of the two component polymers, suggesting a miscible state of PT and PDLLA. In contrast, the copolymers consisting of PDLLA side chains of Mn = 1100, 2000, and 7000 Da showed two isolated Tg, suggesting two segregated domains. The AFM phase images of the copolymers supported the single and phase‐separated morphologies for the former and latter systems, respectively. © 2011 Wiley Periodicals, Inc. We describe the potential of microchip electrophoresis with a Hitachi SV1210, which can be used to evaluate the integrity of total RNA, for the analysis of mRNA expression. The ribonuclease (RNase) protection assay was performed by using microchip electrophoresis with cyanine 5 (Cy5) labeled 248-base antisense RNA probe (riboprobe) encoding adipose-type fatty acid binding protein (A-FABP) as the riboprobe. The fluorescence intensity corresponding to the protected RNA fragment increased in a dose-dependent manner with respect to the complementary strand RNA. Results were obtained in 120 s, and the same amount of Cy5-labeled antisense riboprobe as used in the conventional method can be used. Furthermore, 8 times more sensitive detection of mRNA by microchip electrophoresis could be obtained. An obvious increase in the mRNA expression of A-FABP, which is known as a differentiation marker of adipocytes, occurred during the adipocyte differentiation of 3T3-L1 cells. These results clearly indicate the potential of microchip electrophoresis for the analysis of mRNA expression in cells.  相似文献

Synthesis and inhibitory activities against α-l-fucosidase of phenylalkyl-substituted polyhydroxypiperidines have been described. Among the newly synthesized compounds, 2,4,6-trichloro derivative (16q) showed very high inhibitory activity against α-l-fucosidase with IC50 value of 0.005 μM, and Ki values of 0.0011 μM, respectively.  相似文献

Recently, we have reported a metal-macrocycle framework (MMF) with five enantiomerically paired molecular binding pockets that exhibit site-selective guest arrangement on the nano-channel surface in soaking experiments using a variety of guest molecules. The guest inclusion is based largely on molecular exchange between solvent molecules such as CH3CN and guest molecules on the surface. Herein, we report that the molecular arrangement on the nano-channel surface varies with size, shape and/or chemical properties of functional groups of guests, mono-substituted benzene derivatives, such as benzonitrile, acetophenone and nitrobenzene. In their inclusion complexes, polar nitrile, acetyl and nitro groups serve as molecular anchors to a macrocyclic cavity through hydrogen bonding. Notably, benzonitrile and benzenesulphonic acid bind only to one pair of enantiomeric binding pockets. Such a highly site-selective binding would enable further multi-component surface modifications in the MMF.  相似文献
