Diacylglycerol (DAG)/water and triacylglycerol (TAG)/water emulsions were prepared using beta-lactoglobulin (beta-LG) as an emulsifier. The oil phase (20% in emulsion) was mixed with beta-LG solution (1% beta-LG in water, pH 7) to prepare the emulsions. A fine oil-in-water emulsion was produced from both DAG and TAG oils. The interfacial protein concentration of the TAG emulsion was higher than that of the DAG emulsion. The zeta potential of the DAG oil droplet was higher than that of the TAG oil droplet. The front-surface fluorescence spectroscopy results revealed that tryptophan residues in beta-LG moved to the more hydrophobic environment during the adsorption of protein on the oil droplet surfaces. Changes in secondary structure of beta-LG during the adsorption were determined by FT-IR spectroscopy. Decreases in the beta-sheet content concomitant with increases in the alpha-helix content were observed during the adsorption to the oil droplets, and the degree of structural change was greater for beta-LG in the TAG emulsion than in the DAG emulsion, indicating the increased unfolding of adsorbed beta-LG on the TAG oil droplet surface. Results of interfacial tension measurement supported this speculation, that is, the increased unfolding of the protein at the TAG-water interface. Trypsin- and proteinase K-catalyzed proteolysis was used to probe the topography of the adsorbed beta-LG on the oil droplet surface. SDS-PAGE analyses of liberated peptides after the proteolysis indicated the higher susceptibility of beta-LG adsorbed on the DAG oil droplet surface than on the TAG oil droplet surface. On the basis of all the results, we discussed the conformation of the adsorbed beta-LG on the two oil droplet surfaces.  相似文献

A new nonlinear optical method is presented to detect proteins binding to a gold surface without using fluorescent-dye labeling. After exposure of the protein-binding surface to a gold nanosphere solution, the nanospheres are immobilized above a gold surface with a nanogap supported by the protein. The gold nanospheres immobilized on the gold surface show strong localized surface plasmon (LSP) resonance, and the formation of this structure results in a marked increase in the optical second harmonic (SH) activity of the gold surface arising from a large enhancement of the electric field localized adjacent to the nanospheres on the LSP resonance. The SH image, therefore, gives a high contrast ratio, 7.0:1, of protein-binding spots to control spots. The contrast ratio is much greater than those obtained by linear reflectivity imaging.  相似文献

Apatite-type silicate supported precious metal catalysts were prepared and investigated for their catalytic activity in selective catalytic NO reduction. Single-phase La9.33Si6O26 and La8.33ASi6O25.5 (A=Ca, Sr, Ba) were obtained by a sol-gel method. Pd/La9.33Si6O26 catalyst exhibited high activity for oxidation of C3H6, comparable to Pd/Al2O3 catalyst, although the specific surface area of La9.33Si6O26 was lower than that of Al2O3. In addition, Pt/La9.33Si6O26 catalyst exhibited higher activity for selective catalytic reduction of NO than Pt/Al2O3 catalyst. Substitution of Ba2+ for La3+ of La9.33Si6O26 led to increased catalytic activity at low temperature.  相似文献

What is the smallest protein? This is actually not such a simple question to answer, because there is no established consensus among scientists as to the definition of a protein. We describe here a designed molecule consisting of only 10 amino acids. Despite its small size, its essential characteristics, revealed by its crystal structure, solution structure, thermal stability, free energy surface, and folding pathway network, are consistent with the properties of natural proteins. The existence of this kind of molecule deepens our understanding of proteins and impels us to define an "ideal protein" without inquiring whether the molecule actually occurs in nature.  相似文献

Synthetic ion channels and pores attract current attention as multicomponent sensors in complex matrixes. This application requires the availability of reactive signal amplifiers that covalently capture analytes and drag them into the pore. pi-Basic 1,5-dialkoxynaphthalenes (1,5-DAN) are attractive amplifiers because aromatic electron donor-acceptor (AEDA) interactions account for their recognition within pi-acidic naphthalenediimide (NDI) rich synthetic pores. Focusing on amplifier design, we report here the synthesis of a complete collection of DAN and dialkoxyanthracene amplifiers, determine their oxidation potentials by cyclic voltammetry, and calculate their quadrupole moments. Blockage experiments reveal that subtle structural changes in regioisomeric DAN amplifiers can be registered within NDI pores. Frontier orbital overlap in AEDA complexes, oxidation potentials, and, to a lesser extent, quadrupole moments are shown to contribute to isomer recognition by synthetic pores. Particularly important with regard to practical applications of synthetic pores as multianalyte sensors, we further demonstrate that application of the lessons learned with DAN regioisomers to the expansion to dialkoxyanthracenes provides access to privileged amplifiers with submicromolar activity.  相似文献

New supramolecular copper complexes with pyrazinotetrathiafulvalene (pyra-TTF) as the ligand, [Cu(II)Cl2(pyra-TTF)] (1) and (pyra-TTF) 2[Cu(I)3Cl4(pyra-TTF)] (2), have been synthesized by the diffusion method. Complex 1 is a black block crystal with a three-dimensional (3-D) supramolecular network; the linear chain [-Cu(II)Cl2-(pyra-TTF)-] n extends along the b axis, where the coordinated pyra-TTF donors are stacked in a head-to-tail and ring-over-bond configuration to construct two-dimensional (2-D) sheets, and between the sheets, there are C...Cl(-) or H...Cl(-) contacts. Even though the electron spin resonance (ESR) measurement reveals the nearly Cu(II) state, complex 1 is a semiconductor with sigmaRT=1.0 x 10(-4) S cm(-1) and Ea=0.33 eV. The high-frequency conductivity measurement also confirmed the intrinsic slight carrier doping from Cu(II) to the pyra-TTF donor. This slight doping enhances not only the real and imaginary dielectric constants but also the antiferromagnetic interaction between Cu(II) spins following the 2-D Heisenberg model with 2J=-20 K. In contrast, complex 2 is a very thin black needle. This needle crystal has two crystallographically independent pyra-TTF molecules, which are coordinated and noncoordinated donors. The coordinated donors composed a supramolecular chain [Cu(I)3Cl4(pyra-TTF)(0)]n , whereas the noncoordinated donors formed conducting alpha'-type pyra-TTF(+0.5) sheets. This complex is semiconducting with sigmaRT=0.1 S cm(-1) and Ea=0.15 eV. Both complexes 1 and 2 demonstrate that the pyra-TTF molecule works not only as an oxidized donor by Cu(II) to construct conducting sheets but also as a ligand coordinated to a Cu cation to form supramolecuar chains.  相似文献

A novel photoresponsive functional monomer bearing diaminopyridine and azobenzene moieties was synthesized and applied to the preparation of photo-regulated molecularly imprinted polymers, which can recognize porphyrin derivatives through hydrogen bonding. The binding affinity of the imprinted cavities was regulated by UV irradiation, suggesting that azobenzene groups located inside the binding sites worked as photosensitizers and the trans-cis isomerization could regulate the affinity for the target compounds. Repetitive binding of the target compound to trans-IP and cis-IP was directly monitored by slab optical waveguide spectroscopy and the photo-mediated regulation of binding affinity was successfully confirmed.  相似文献

We developed new model core potentials (MCPs) for s-block elements from Na to Ra, in which the outer core (n-1)s and (n-1)p electrons are treated explicitly together with the ns electrons. By adding suitable correlating functions, we demonstrated that the present MCP basis sets show excellent performance in describing the electronic structures of atoms and molecules, bringing about accurate ionization potentials of atoms and very good spectroscopic constants of ionic and covalent molecules. The results obtained with the new MCPs are very close to the ones obtained using the all-electron correlation consistent basis sets of Dunning.  相似文献

In this article we study and obtain a classification of Ising vectors in vertex operator algebras (VOAs) associated to binary codes and times root lattices, where an Ising vector is a conformal vector with central charge 1/2 generating a simple Virasoro sub-VOA. Then we apply our results to study certain commutant subalgebras related to root systems. We completely classify all Ising vectors in such commutant subalgebras and determine their full automorphism groups. C. H.
