A highly sensitive, rapid assay method was developed and validated for the estimation of lorglumide in mouse plasma using liquid chromatography coupled to tandem mass spectrometry with electrospray ionization in positive‐ion mode. The assay procedure involves extraction of lorglumide and phenacetin (internal standard, IS) from mouse plasma with simple protein precipitation. Chromatographic separation was achieved using an isocratic mobile (0.2% formic acid solution–acetonitrile, 20:80, v/v) at a flow‐rate of 0.5 mL/min on an Atlantis dC18 column maintained at 40 °C with a total run time of 4.0 min. The MS/MS ion transitions monitored were 459.2 → 158.4 for lorglumide and 180.1 → 110.1 for IS. Method validation was performed as per FDA guidelines and the results met the acceptance criteria. The lower limit of quantitation achieved was 0.42 ng/mL and the linearity range extended from 0.42 to 500 ng/mL. A highly sensitive and specific LC-MS/MS-ESI method was developed for simultaneous quantification of albenadazole (ABZ) and ricobendazole (RBZ) in rat plasma (50 μL) using phenacetin as an internal standard (IS). Simple protein precipitation was used to extract ABZ and RBZ from rat plasma. The chromatographic resolution of ABZ, RBZ and IS was achieved with a mobile phase consisting of 5 m m ammonium acetate (pH 6) and acetonitrile (20:80, v/v) at a flow rate of 1 mL/min on a Chromolith RP-18e column. The total chromatographic run time was 3.5 min and the elution of ABZ, RBZ and IS occurred at 1.66, 1.50 and 1.59 min, respectively. A linear response function was established for the ranges of concentrations 2.01-2007 and 6.02-6020 ng/mL for ABZ and RBZ, respectively. The intra- and inter-day precision values for ABZ and RBZ met the acceptance as per FDA guidelines. ABZ and RBZ were stable in battery of stability studies, viz. bench-top, auto-sampler and freeze-thaw cycles. The developed assay was applied to a pharmacokinetic study in rats. 相似文献

An approach has been developed for the quantitative determination of concentrations of centchroman ( I), a nonsteroidal once‐a‐week oral contraceptive, and its major metabolite (7‐desmethyl centchroman, II) using dried blood spots (DBS) on paper, rather than conventional plasma samples. The assay employed simple solvent extraction of the DBS sample circle (6 mm) requiring small blood volumes (30 μL) followed by reversed‐phase HPLC separation, combined with multiple reaction monitoring mass spectrometric detection. The calibration plot in matrix using d ‐trans‐hydroxy chroman as internal standard (IS) was linear (r2 = 0.998) over ranges of 1.5–240 and 4.5–720 ng/mL for I and II, respectively. The recoveries of both I and II were always >60% with quantification limits (signal‐to‐noise ratio = 10) of 1.5 and 4.5 ng/mL for I and II, respectively. The intra‐day and inter‐day precision (%RSD) and accuracy (%bias) variations in blood spots for both I and II were better than 13%. Moreover, both I and II were stable in DBS for at least 3 months when stored at room temperature. The developed method was successfully applied to the pharmacokinetic interaction study after oral administration of centchroman with and without co‐administration of carbamazepine in female Sprague–Dawley rats using serial sampling and results were comparable with the plasma concentrations reported earlier. The high dose rate electron beams are increasingly being used for radiation processing of various products worldwide. A comprehensive dosimetric evaluation of an in-house developed 10 MeV industrial electron beam irradiator was carried out in static as well as in dynamic mode of irradiations. Radiochromic B3 film and graphite calorimeter were used for dosimetric measurements. The dose rate from the electron beam was also calculated using the empirical relation prescribed in the ASTM report E2232-02. The measured electron beam profile indicates the dose rate variation within 8% in the irradiated product boxes. The most probable energy determined from the depth dose distribution in PMMA, Al and water was found in agreement with the intended energy of the electron beam. Measured dose rate using radiochromic film and graphite calorimeter were found in good agreement with each other and also found comparable with the theoretically estimated dose rates. Experimentally measured dose rates were considered for the trial irradiation of medical and industrial products. Dosimetric data obtained through this study confirms the suitability of the irradiator for routine radiation processing of various products. 相似文献

Due to the importance of neuraminidase in the pathogenesis of influenza virus infection, it has been regarded as the most important drug target for the treatment of influenza. Resistance to currently available drugs and new findings related to structure of the protein requires novel neuraminidase 1 (N1) inhibitors. In this study, a consensus QSAR model with defined applicability domain (AD) was developed using published N1 inhibitors. The consensus model was validated using an external validation set. The model achieved high sensitivity, specificity, and overall accuracy along with low false positive rate (FPR) and false discovery rate (FDR). The performance of model on the external validation set and training set were comparable, thus it was unlikely to be overfitted. The low FPR and low FDR will increase its accuracy in screening large chemical libraries. Screening of ZINC library resulted in 64,772 compounds as probable N1 inhibitors, while 173,674 compounds were defined to be outside the AD of the consensus model. The advantage of the current model is that it was developed using a large and diverse dataset and has a defined AD which prevents its use on compounds that it is not capable of predicting. Structural and optical properties of the Tb-doped ZnO nanoparticles with average diameter ≈4 nm have been systematically investigated.
Our X-ray diffraction studies show a contraction of the ZnO lattice with the increase of the Tb mole-fraction x for x ≤ 0.02 and an expansion beyond x ≈ 0.02. The photoluminescence spectra are found to be comprised of a near band edge ultra violet luminescence (UVL) and a
broad green luminescence (GL) band. Under the atmospheric condition, the intensity of the GL band is found to increase with
the Tb mole-fraction over the entire doping range. On the other hand, under the vacuum condition, it has been observed that
the GL intensity decreases with the increase of x up to x ≈ 0.02 but further increase of x leads to a gradual revival of the GL emission. Our study suggests that for x ≤ 0.02, GL results due to the physisorption of certain groups on the surface of the nanoparticles (GL-groups). It is also
found that in this Tb mole-fraction regime, Tb incorporates mostly on the surface of the nanoparticles and affects the UVL
to GL intensity ratio by influencing the attachment of the GL-groups. However, for x > 0.02, GL originates not only from the GL-groups but also from certain point defects, which are likely to be generated due
to the incorporation of Tb in the core of the nanoparticles. A simple rate equation model is introduced to get a quantitative
understanding about the variation of the density of the centers responsible for the GL emission as a function of x under the atmospheric and the vacuum conditions. 相似文献

A certified reference material, ERM-FD100, for quality assurance and validation of various nanoparticle sizing methods, was developed by the Institute for Reference Materials and Measurements. The material was prepared from an industrially sourced colloidal silica containing nanoparticles with a nominal equivalent spherical diameter of 20?nm. The homogeneity and stability of the candidate reference material was assessed by means of dynamic light scattering and centrifugal liquid sedimentation. Certification of the candidate reference material was based on a global interlaboratory comparison in which 34 laboratories participated with various analytical methods (DLS, CLS, EM, SAXS, ELS). After scrutinising the interlaboratory comparison data, 4 different certified particle size values, specific for the corresponding analytical method, could be assigned. In recent days amino acid single crystals are gaining importance due to good optical behavior. The title compound consists of l-alanine; an amino acid and maleic acid; an aromatic organic compound. It has been successfully synthesized and the single crystal has been grown by slow evaporation solution growth. The grown single crystal has been characterized by powder X-ray diffraction (PXRD) analysis and FTIR measurements. The crystalline perfection was examined by high-resolution X-ray diffraction (HRXRD) technique and found that the specimen quality is quite good. Optical behavior has been assessed by UV–Vis analysis and found that there is no absorption in the entire visible region. The relative second harmonic generation (SHG) efficiency measurement reveals that the incorporation of maleic acid to l-alanine leads to increase its value of SHG. The mechanical behavior and the thermal analysis have been carried out using Vickers microhardness and TG/DTA measurements respectively. In the present paper, we report about synthesis of nanostructured organic–inorganic heterojunction of CdS/Polyaniline/CuInSe2 thin films by cost effective chemical route at room temperature, for solar cell application. As such obtained thin films are characterized for structural, compositional, morphological, optical and electrical properties by X-ray diffraction (XRD) pattern, energy dispersive X-ray (EDAX) analysis, scanning electron microscopy (SEM), optical absorbance spectra and I–V response respectively. The XRD reveals the polycrystalline nature of the thin films having tetragonal crystal structure and a crystallite size of 19 nm. The presence of observed and expected elements in the EDAX spectra confirms the elemental compositions in CdS/Polyaniline/CuInSe2 thin films. From SEM images it can be inferred that the surface morphology of the Polyaniline thin films exists like clothing fibers, while CdS/CuInSe2 shows granular shape particles distributed over the substrate and the SEM of CdS/Polyaniline/CuInSe2 represents mixing and attachment of circular particles to fiber like structure. The optical absorbance spectra have shown red shift in absorbance strength and energy band gap value of CdS/CuInSe2 from ~ 1.36 eV to ~ 1.62 eV upon formation of CdS/Polyaniline/CuInSe2 thin film. The I–V response of CdS/CuInSe2 and CdS/Polyaniline/CuInSe2 measured under dark and illumination to 100 mW/cm2 light, exhibited the solar characteristics from these graphs and the conversion efficiency calculated is observed to be 0.26 and 0.55% for CdS/CuInSe2 and CdS/Polyaniline/CuInSe2 thin films respectively. 相似文献
