The total synthesis of 6-amino-1,2,3-triazolo[4,5-c]pyridin-4(5H)one (8-aza-3-deazaguanine, 3 ) and 6-amino-1-(β-D-ribofuranosyl)-1,2,3-triazolo[4,5-c]pyridin-4(5H)one (8-aza-3-deazaguano-sine, 22 ) has been described for the first time by a novel base-catalyzed ring closure of 4(5)-cyanomethyl-1,2,3-triazole-5(4)carboxamide (14) and methyl 5-cyanomethyl-1-(2,3,5-tri-O-ben-zoyl-β-D-ribofuranosyl)-1,2,3-triazole-4-carboxylate (17) , respectively. Under the catalysis of DBU, 2,4-dinitrophenylhydrazone of dimethyl 1,3-acetonedicarboxylate (7) was converted to methyl 5-methoxycarbonylmethyl-1-(2,4-dinitroanilino)-1,2,3-triazole-4-carboxylate (12) via dimethyl 2-diazo-3-iminoglutarate (8) . Catalytic reduction of 12 gave methyl 4(5)methoxycar-bonylmethyl-1,2,3-triazole-5(4)carboxylate (11) from which methyl 4(5)carbamoylmethyl-1,2,3-triazole-5(4)carboxylate (10) was obtained by ammonolysis. Dehydration of 10 provided methyl 4(5)cyanomethyl-1,2,3-triazole-5(4)carboxylate (13) which on amination gave 14 . The 1,2,3-triazole nucleosides 17, 18 and 19 were obtained from the stannic chloride-catalyzed condensation of the trimethylsilyl 13 and a fully acylated β-D-ribofuranose. The yield and ratio of the ribofuranosyl derivatives of 13 markedly depends on the ratio of stannic chloride used. The structures of the nucleosides 22 and 23 were established by a combination of NOE, 1H-nmr and 13C-nmr spectroscopy.  相似文献

A new class of nonpeptidic inhibitors of the ZnII‐dependent metalloprotease neprilysin with IC50 values in the nanomolar activity range (0.034–0.30 μM ) were developed based on structure‐based de novo design (Figs. 1 and 2). The inhibitors feature benzimidazole and imidazo[4,5‐c]pyridine moieties as central scaffolds to undergo H‐bonding to Asn542 and Arg717 and to engage in favorable ππ stacking interactions with the imidazole ring of His711. The platform is decorated with a thiol vector to coordinate to the ZnII ion and an aryl residue to occupy the hydrophobic S1′ pocket, but lack a substituent for binding in the S2′ pocket, which remains closed by the side chains of Phe106 and Arg110 when not occupied. The enantioselective syntheses of the active compounds (+)‐ 1 , (+)‐ 2 , (+)‐ 25 , and (+)‐ 26 were accomplished using Evans auxiliaries (Schemes 2, 4, and 5). The inhibitors (+)‐ 2 and (+)‐ 26 with an imidazo[4,5‐c]pyridine core are ca. 8 times more active than those with a benzimidazole core ((+)‐ 1 and (+)‐ 25 ) (Table 1). The predicted binding mode was established by X‐ray analysis of the complex of neprilysin with (+)‐ 2 at 2.25‐Å resolution (Fig. 4 and Table 2). The ligand coordinates with its sulfanyl residue to the ZnII ion, and the benzyl residue occupies the S1′ pocket. The 1H‐imidazole moiety of the central scaffold forms the required H‐bonds to the side chains of Asn542 and Arg717. The heterobicyclic platform additionally undergoes π‐π stacking with the side chain of His711 as well as edge‐to‐face‐type interactions with the side chain of Trp693. According to the X‐ray analysis, the substantial advantage in biological activity of the imidazo‐pyridine inhibitors over the benzimidazole ligands arises from favorable interactions of the pyridine N‐atom in the former with the side chain of Arg102. Unexpectedly, replacement of the phenyl group pointing into the deep S1′ pocket by a biphenyl group does not enhance the binding affinity for this class of inhibitors.  相似文献

Thermal processing of as-made fluorozirconate glasses, which were additionally doped with rare-earth and chlorine ions, results in the formation of fluorescent nanocrystals therein. For medical applications, the glasses were doped with divalent europium ions as the fluorescent rare-earth ion, while trivalent neodymium was used to develop up-conversion systems. The samples were annealed up to 290 °C to initiate the growth of hexagonal or orthorhombic phase BaCl2 nanocrystals therein. Upon annealing some of the rare-earth ions were incorporated into the BaCl2 nanocrystals leading to enhanced fluorescence properties. The particle diameters were in the range of a few nanometers to several tens of nanometers.  相似文献

Herein, we report the synthesis and X-ray structural analysis of a collection of fluorinated metal N-heterocyclic carbenes (Ag, Au, Pd, Rh, Ir) and their precursor salts. The common structural feature of these species is a flanking fluoroethyl group, which is either freely rotating or embedded within a bicyclic framework. Solid state analysis confirmed a gauche conformational preference in all cases with the fluorine adopting a syn clinal arrangement (?[NCCF]~60°) with respect to the triazolium nitrogen at the vicinal position of the NHC. A density functional theory analysis was employed to quantify these effects and evaluate the influence of electronic modulation of the carbenic carbon [(CN+); neutral carbene (C:); metal-bound carbene (CM)], on the relative gauche/anti preference, thus highlighting the potential of this conformational phenomenon as a useful molecular design strategy for controlling the topology of organometallic complexes.  相似文献

In this paper, an approach for controlling the macro-step size in connection with co-simulation methods [1, 2, 4] is suggested. The investigated step-size controller is tailored for semi-implicit co-simulation techniques. Concretely, we consider predictor/corrector co-simulation approaches [3]. By comparing variables from the predictor and the corrector step, an error estimator for the local error can be constructed. Gas thrust bearings are often used in low-load applications, e.g. in air cycle machines, in micro gas turbines or in rotor systems for fuel cell applications, to support a shaft in axial direction. The pressure and temperature distribution in a gas thrust bearing pad are described by the generalized Reynolds equation according to Dowson and the 3D energy equation. In this paper, two different approaches are presented in order to reduce the dimension of the governing nonlinear integro-differential equation system and in order to stabilize the solution process. In the first reduction approach, the temperature in the fluid is averaged across the fluid film according to Lee and Kim. In the second approach, Legendre polynomials are used to approximate temperature, density and fluidity across the fluid film according to Elrod, Brewe and Moraru. A molecular level theory for the physical aging of polymer glasses is proposed. The nonequilibrium time evolution of the amplitude of long wavelength density fluctuations, and its influence on activated barrier hopping, plays an essential role. The theory predicts temperature-dependent apparent power-law aging of the segmental relaxation time and logarithmic aging of thermodynamiclike properties, in good accord with experiments. A physical origin for the quantitative nonuniversal aspects based on the amplitude of quenched density fluctuations is suggested.  相似文献
