InhA, the enoyl reductase from Mycobacterium tuberculosis, catalyzes the NADH-dependent reduction of trans-2-enoyl-ACPs. In the present work, Raman spectroscopy has been used to identify catalytically relevant changes in the conformation of the nicotinamide ring that occur when NADH binds to InhA. For 4(S)-NADD, there is an 11 cm-1 decrease in the wavenumber of the C4-D stretching band (nuC-D) and a 50% decrease in the width of this band upon binding to InhA. While a similar reduction in line width is observed for the corresponding band arising from 4(R)-NADD, nuC-D for this isomer increases 34 cm-1 upon binding to InhA. These changes in nuC-D indicate that the nicotinamide ring adopts a bound conformation in which the 4(S)C-D bond is in a pseudoaxial orientation. Mutagenesis of F149, a conserved active site residue close to the cofactor, demonstrates that this enzyme-induced modulation in cofactor structure is directly linked to catalysis. In contrast to the wild-type enzyme, Raman spectra of NADD bound to F149A InhA resemble those of NADD in solution. Consequently, F149A is no longer able to optimally position the cofactor for hydride transfer, which correlates with the 30-fold decrease in kcat and 2-fold increase in D(V/KNADH) caused by this mutation. These studies thus substantiate the proposal that hydride transfer is promoted by pseudoaxial positioning of the NADH pro-4S bond, and indicate that catalysis of substrate reduction by InhA results, in part, from correct orientation of the cofactor in the ground state.  相似文献

The spontaneous micelle‐to‐vesicle transition in an aqueous mixture of two surface‐active ionic liquids (SAILs), namely, 1‐butyl‐3‐methylimidazolium n‐octylsulfate ([C4mim][C8SO4]) and 1‐dodecyl‐3‐methylimidazoium chloride ([C12mim]Cl) is described. In addition to detailed structural characterization obtained by using dynamic light scattering, transmission electron microscopy (TEM), and cryogenic TEM techniques, ultrafast fluorescence resonance energy transfer (FRET) from coumarin 153 (C153) as a donor (D) to rhodamine 6G (R6G) as an acceptor (A) is also used to study micelle–vesicle transitions in the present system. Structural transitions of SAIL micelles ([C4mim][C8SO4] or [C12mim]Cl micelles) to mixed SAIL vesicles resulted in significantly increased D –A distances, and therefore, increased timescale of FRET. In [C4mim][C8SO4] micelles, FRET between C153 and R6G occurs on an ultrafast timescale of 3.3 ps, which corresponds to a D –A distance of about 15 Å. As [C4mim][C8SO4] micelles are transformed into mixed micelles upon the addition of a 0.25 molar fraction of [C12mim]Cl, the timescale of FRET increases to 300 ps, which suggests an increase in the D –A distance to 31 Å. At a 0.5 molar fraction of [C12mim]Cl, unilamellar vesicles are formed in which FRET occurs on multiple timescales of about 250 and 2100 ps, which correspond to D –A distances of 33 and 47 Å. Although in micelles and mixed micelles the obtained D –A distances are well correlated with their radius, in vesicles the obtained D –A distance is within the range of the bilayer thickness.  相似文献

Monodisperse colloidal silver nanospheres were synthesized by the reaction of silver nitrate, hydroxylammonium hydrosulphate (NH2OH)2 · H2SO4 and sodium hydroxide in the presence of gelatin as stabilizer. Colloidal nanospheres were characterized by UV-vis absorption spectroscopy, transmission electron microscopy, X-ray diffraction and dynamic light scattering. X-ray diffraction data confirmed that the silver nanospheres were crystalline with face-centered-cubic structure. Transmission electron microscopy analysis revealed the formation of homogeneously distributed silver nanoparticles of spherical morphology and size of the nanoparticles was in the range of 0.7–5.2 nm. Silver nanospheres were stable for more than two months when stored at ambient temperature. Size and size distribution were studied by varying pH, reaction temperature, silver ion concentration in feed solution, concentration of reducing agent and concentration of the stabilizing agent. Catalytic activity of silver nanospheres was tested for the reduction reaction of nitro compounds in sodium borohydride solution. Monodisperse silver nanospheres showed excellent catalytic activity towards the reduction of aromatic nitro compounds. The reduction rate of aromatic nitro compounds had been observed to follow the sequence 4-nitrophenol > 2-nitrophenol > 3-nitrophenol.  相似文献

The development and progression of heart failure (HF) due to myocardial infarction (MI) is a major concern even with current optimal therapy. Resveratrol is a plant polyphenol with cardioprotective properties. Sacubitril/valsartan is known to be beneficial in chronic HF patients. In this study, we investigated the comparative and combinatorial benefits of resveratrol with sacubitril/valsartan alongside an active comparator valsartan in MI-induced male Sprague Dawley rats. MI-induced and sham-operated animals received vehicle, resveratrol, sacubitril/valsartan, valsartan alone or sacubitril/valsartan + resveratrol for 8 weeks. Echocardiography was performed at the endpoint to assess cardiac structure and function. Cardiac oxidative stress, inflammation, fibrosis, brain natriuretic peptide (BNP), creatinine and neutrophil gelatinase associated lipocalin were measured. Treatment with resveratrol, sacubitril/valsartan, valsartan and sacubitril/valsartan + resveratrol significantly prevented left ventricular (LV) dilatation and improved LV ejection fraction in MI-induced rats. All treatments also significantly reduced myocardial tissue oxidative stress, inflammation and fibrosis, as well as BNP. Treatment with the combination of sacubitril/valsartan and resveratrol did not show additive effects. In conclusion, resveratrol, sacubitril/valsartan, and valsartan significantly prevented cardiac remodeling and dysfunction in MI-induced rats. The reduction in cardiac remodeling and dysfunction in MI-induced rats was mediated by a reduction in cardiac oxidative stress, inflammation and fibrosis.  相似文献

Currently, gastrointestinal bleeding in the colon wall and the small bowel is diagnosed and treated with endoscopes. However, the locations of this condition are often problematic to treat using traditional flexible and tethered tools. New studies commonly consider untethered devices for solving this problem. However, there still exists a gap in the extant literature, and more research is needed to diagnose and deliver drugs in the lower gastrointestinal tract using soft robotic carriers. This paper discusses the development of an untethered, magnetically-responsive bio-inspired soft carrier. A molding process is utilized to produce prototypes from Diisopropylidene-1,6-diphenyl-1,6-hexanediol-based Polymer with Ethylene Glycol Dimethacrylate (DiAPLEX) MP-3510 - a shape memory polymer with a low transition temperature to enable the fabrication of these carriers. The soft carrier design is validated through simulation results of deformation caused by magnetic elements embedded in the carrier in response to an external field. The thermal responsiveness of the fabricated prototype carriers is assessed ex vivo and in a phantom. The results indicate a feasible design capable of administering drugs to a target inside a phantom of a large intestine. The soft carrier introduces a method for the controlled release of drugs by utilizing the rubbery modulus of the polymer and increasing the recovery force through magnetic actuation.  相似文献

In this paper we discuss the preparation and comparative evaluation of silver (I) [Ag(I)] nonwoven and woven antimicrobial barrier fabrics generated from commercial calcium‐sodium alginates and laboratory prepared sodium carboxymethyl (CM) cotton nonwovens and CM‐cotton printcloth for potential use as wound dressings. Degrees of CM substitution (DS) in cotton nonwoven and printcloth samples by titrimetry were 0.38 and 0.10, respectively. Coordination of Ag(I) with carboxylates on fabrics was effected by ion exchange and nitrates were removed by washing to mitigate nitrate ion toxicity issues. Durability of silver coordinated fabrics was tested by soaking them in deionized water with slight agitation at 50°C. Ag(I) alginates and nonwoven Ag(I)‐CM‐cottons lost structural integrity in water. Ag‐CM‐cotton printcloth samples retained structural integrity even after four soak‐and‐dry cycles, were smooth to the touch when dry, and were smoother when moistened. They could be easily peeled from wound surfaces without inducing trauma. Solid‐state carbon‐13 (13C) nuclear magnetic resonance (NMR) spectrometry was used to observe changes in carbonyl resonances in Ag(I) alginates and Ag(I)‐CM‐printcloth, and the chemical shift positions of carbonyl resonances of uncoordinated and Ag(I) coordinated fabrics did not change. Inductively coupled plasma‐mass spectrometry (ICP‐MS) was used following fabric digestion to determine the total Ag(I) ion content in fabrics. Ag(I) alginates were found to hold about 10–50 mg Ag(I) per gram fabric; and Ag(I) cotton woven and nonwoven fabrics held about 5–10 mg Ag(I) ions per gram fabric. Kinetic release of Ag(I) after soaking once in physiological saline was studied with ICP‐MS to estimate the availability of Ag(I) upon a single exchange with Na(I) ions on wound surfaces. Alginates released between ~13 and 28% of coordinated Ag(I), and CM‐cotton nonwovens and CM‐cotton printcloth released ~14 and 3% of coordinated Ag(I) ions, respectively. Finally, Ag(I) alginates and Ag(I)‐CM‐cotton printcloth samples were evaluated against Gram‐positive Staphylococcus aureus and Gram‐negative Klebsiella pneumoniae. Ag(I) alginates suppressed 99.95% of bacterial growth in vitro. Even after four soak‐and‐dry cycles in deionized water Ag(I)‐CM‐cotton printcloth suppressed 99.99% of bacterial growth in vitro. A gas chromatography-mass spectroscopic method in electron ionization (EI) mode with MS/MS ion preparation using helium at flow rate 1 ml min(-1) as carrier gas on DB-5 capillary column (30 m x 0.25 mm i.d. film thickness 0.25 microm) has been developed for the determination of benzene in indoor air. The detection limit for benzene was 0.002 microg ml(-1) with S/N: 4 (S: 66, N: 14). The benzene concentration for cooks during cooking time in indoor kitchen using dung fuel was 114.1 microg m(-3) while it was 6.6 microg m(-3) for open type kitchen. The benzene concentration was significantly higher (p < 0.01) in indoor kitchen with respect to open type kitchen using dung fuels. The wood fuel produces 36.5 microg m(-3) of benzene in indoor kitchen. The concentration of benzene in indoor kitchen using wood fuel was significantly (p < 0.01) lower in comparison to dung fuel. This method may be helpful for environmental analytical chemist dealing with GC-MS in confirmation and quantification of benzene in environmental samples with health risk exposure assessment.  相似文献

A new and efficient seven-step synthesis of 8,9-difluoro-2-methyl-6-oxo-1,2-dihydropyrrolo[3,2,1-ij]-quinoline-5-carboxylic acid ( 9 ), an important intermediate used in the synthesis of quinolone antibacterials, has been developed beginning with commercially available 2,3,4-trifluoronitrobenzene. Selective displacement of the 2-fluorine of the starting material with the anion of ethyl acetoacetate and subsequent hydrolysis and decarboxylation affords the arylacetone derivative 11 . Reduction of the ketone and nitro groups of 11 followed by condensation with diethyl ethoxymethylenemalonate gives 14 , which is cyclized to the indole derivative 15 by the Mitsunobu procedure. Friedel-Crafts cyclization of 15 and acid hydrolysis gives the title compound 9 .  相似文献

We have studied the spreading of phospholipid vesicles on photochemically patterned n-octadecylsiloxane monolayers using epifluorescence and imaging ellipsometry measurements. Self-assembled monolayers of n-octadecylsiloxanes were patterned using short-wavelength ultraviolet radiation and a photomask to produce periodic arrays of patterned hydrophilic domains separated from hydrophobic surroundings. Exposing these patterned surfaces to a solution of small unilamellar vesicles of phospholipids and their mixtures resulted in a complex lipid layer morphology epitaxially reflecting the underlying pattern of hydrophilicity. The hydrophilic square regions of the photopatterned OTS monolayer reflected lipid bilayer formation, and the hydrophobic OTS residues supported lipid monolayers. We further observed the existence of a boundary region composed of a nonfluid lipid phase and a lipid-free moat at the interface between the lipid monolayer and bilayer morphologies spontaneously corralling the fluid bilayers. The outer-edge of the boundary region was found to be accessible for subsequent adsorption by proteins (e.g., streptavidin and BSA), but the inner-edge closer to the bilayer remained resistant to adsorption by protein or vesicles. Mechanistic implications of our results in terms of the effects of substrate topochemical character are discussed. Furthermore, our results provide a basis for the construction of complex biomembrane models, which exhibit fluidity barriers and differentiate membrane properties based on correspondence between lipid leaflets. We also envisage the use of this construct where two-dimensionally fluid, low-defect lipid layers serve as sacrificial resists for the deposition of protein and other material patterns.  相似文献

This study assessed the acoustic and perceptual effect of noise on vowel and stop-consonant spectra. Multi-talker babble and speech-shaped noise were added to vowel and stop stimuli at -5 to +10 dB S/N, and the effect of noise was quantified in terms of (a) spectral envelope differences between the noisy and clean spectra in three frequency bands, (b) presence of reliable F1 and F2 information in noise, and (c) changes in burst frequency and slope. Acoustic analysis indicated that F1 was detected more reliably than F2 and the largest spectral envelope differences between the noisy and clean vowel spectra occurred in the mid-frequency band. This finding suggests that in extremely noisy conditions listeners must be relying on relatively accurate F1 frequency information along with partial F2 information to identify vowels. Stop consonant recognition remained high even at -5 dB despite the disruption of burst cues due to additive noise, suggesting that listeners must be relying on other cues, perhaps formant transitions, to identify stops.  相似文献
