Nonmammalian cytosine deaminases (CDs) have been investigated for last 30?years in the context of cancer therapy. The therapeutic effect of CD is based on its ability to catalyze the conversion of nontoxic prodrug 5-fluorocytosine (5FC) into the anticancer drug 5-fluorouracil (5FU) by deamination of the number 4 carbon of 5FC. This deamination property of CD has been explored to develop innovative therapeutic approach for treatment of cancer. A general overview is needed for the identification of efficient cytosine deaminases for potential use in cancer therapy. In this review, we have discussed about nonmammalian CDs for a variety of prodrug gene/enzyme therapy applications with several recent examples. Finally, we have provided a prospective on the future aspects of CDs and their applications in cancer therapy. 相似文献

An aromatic copolyester composed of 25 mol % phenyl hydroquinone, 10 mol % isophthalic acid, 40 mol % chloroterephthalic acid, and 25 mol % t-butyl hydroquinone (PICT) has been synthesized. This amorphous, glassy polymer is soluble in common organic solvents such as methylene chloride. Thin, solution-cast films may be prepared which are in a metastable, vitrified, optically isotropic state. On first heating of an isotropic film at 20°C/min in a calorimeter, one glass transition is observed at low temperature (approximately 49°C) and is ascribed to the glass/rubber transition of the metastable, isotropic polymer. This thermal event is followed by a small exotherm due to the development of order during the scan, which results in a second Tg at approximately 125°C. This Tg is associated with the glass/rubber transition of the ordered polymer. Nematic order can be developed by thermal annealing. The lower Tg increases toward the upper Tg as annealing time is increased. For an initially isotropic film annealed at 90°C, the increase of the lower Tg with annealing time and the increase in birefringence observed by optical microscopy are governed by similar kinetics. Isotropization occurs in the temperature range of 250–300°C. The nematic polymer is slightly more dense than its isotropic analog. No detectable differences between isotropic and nematic samples were observed in rotating frame proton spin lattice relaxation times. © 1996 John Wiley & Sons, Inc. 相似文献

A rare example of a mononuclear complex [(bpy)2Ru(L1?H)](ClO4), 1 (ClO4) and dinuclear complexes [(bpy)2Ru(μ‐L1?2H)Ru(bpy)2](ClO4)2, 2 (ClO4)2, [(bpy)2Ru(μ‐L2?2H)Ru(bpy)2](ClO4)2, 3 (ClO4)2, and [(bpy)2Ru(μ‐L3?2H)Ru(bpy)2](ClO4)2, 4 (ClO4)2 (bpy=2,2′‐bipyridine, L1=2,5‐di‐(isopropyl‐amino)‐1,4‐benzoquinone, L2=2,5‐di‐(benzyl‐amino)‐1,4‐benzoquinone, and L3=2,5‐di‐[2,4,6‐(trimethyl)‐anilino]‐1,4‐benzoquinone) with the symmetrically substituted p‐quinone ligands, L, are reported. Bond‐length analysis within the potentially bridging ligands in both the mono‐ and dinuclear complexes shows a localization of bonds, and binding to the metal centers through a phenolate‐type “O?” and an immine/imminium‐type neutral “N” donor. For the mononuclear complex 1 (ClO4), this facilitates strong intermolecular hydrogen bonding and leads to the imminium‐type character of the noncoordinated nitrogen atom. The dinuclear complexes display two oxidation and several reduction steps in acetonitrile solutions. In contrast, the mononuclear complex 1 + exhibits just one oxidation and several reduction steps. The redox processes of 1 1+ are strongly dependent on the solvent. The one‐electron oxidized forms 2 3+, 3 3+, and 4 3+ of the dinuclear complexes exhibit strong absorptions in the NIR region. Weak NIR absorption bands are observed for the one‐electron reduced forms of all complexes. A combination of structural data, electrochemistry, UV/Vis/NIR/EPR spectroelectrochemistry, and DFT calculations is used to elucidate the electronic structures of the complexes. Our DFT results indicate that the electronic natures of the various redox states of the complexes in vacuum differ greatly from those in a solvent continuum. We show here the tuning possibilities that arise upon substituting [O] for the isoelectronic [NR] groups in such quinone ligands. 相似文献

Study of electron drift velocity caused by Etimes B motion is done with the help of a Mach probe in a dc cylindrical magnetron sputtering system at different plasma discharge parameters like discharge voltage, gas pressure and applied magnetic field strength. The interplay of the electron drift with the different discharge parameters has been investigated. Strong radial variation of the electron drift velocity is observed and is found to be maximum near the cathode and it decreases slowly with the increase of radial distance from the cathode. The sheath electric field, E measured experimentally from potential profile curve using an emissive probe is contributed to the observed radial variation of the electron drift velocity. The measured values of the drift velocities are also compared with the values from the conventional theory using the experimental values of electric and magnetic fields. This study of the drift velocity variation is helpful in providing a useful insight for determining the discharge conditions and parameters for sputter deposition of thin film. 相似文献

Tensile properties of the polyimide and copolyimide films based on two dianhydrides, pyromellitic dianhydride (PMDA) and 3,3′,4,4′-benzophenonetetracarboxylic dianhydride (BTDA) and two diamines, 4,4′-oxydianiline (ODA), and a proprietary aromatic diamine (PD) have been described. The tensile strength of the films containing higher proportions of BTDA or PMDA and PD is much higher (except the fully rigid film based on PMDA-PD which is brittle in nature) than the films containing higher proportion of ODA moiety. The films containing PD as the diamine moiety exhibit high initial moduli than the films containing exclusively or mainly ODA as the diamine moiety. The films having higher concentration of the -O- linkage originated from diamine ODA are found to exhibit higher elongation values. There is found to be no direct correlation between ηinh of the precursor casting solutions and mechanical properties of structurally different polyimide/copolyimide films. For a particular polyimide or copolyimide film, the tensile strength value is found to be less sensitive than the elongation to the variation of ηinh value of the precursor poly(amic acid) or copoly(amic acid). Tensile strength and elongation of the film, basically rigid in nature, may be improved by post-curing at 360°C/370°C. While Kapton H film retains 78% and 63.5% of its tensile strength and % elongation at break (% Eb) respectively after hot-wet mechanical test, the film based on BTDA 80, PMDA 20 and PD shows an increase of about 27% and 22% in its tensile strength and % Eb respectively. 相似文献

Metallamacrocycles 1, 2, and 3 of the general formula [{Ir(ppy)(2)}(2)(μ-BL)(2)](OTf)(2) (ppyH = 2-phenyl pyridine; BL = 1,2-bis(4-pyridyl)ethane (bpa) (1), 1,3-bis(4-pyridyl)propane (bpp) (2), and trans-1,2-bis(4-pyridyl)ethylene (bpe) (3)) have been synthesized by the reaction of [{(ppy)(2)Ir}(2)(μ-Cl)(2)], first with AgOTf to effect dechlorination and later with various bridging ligands. Open-frame dimers [{Ir(ppy)(2)}(2)(μ-BL)](OTf)(2) were obtained in a similar manner by utilizing N,N'-bis(2-pyridyl)methylene-hydrazine (abp) and N,N'-(bis(2-pyridyl)formylidene)ethane-1,2-diamine (bpfd) (for compounds 4 and 5, respectively) as bridging ligands. Molecular structures of 1, 3, 4, and 5 were established by X-ray crystallography. Cyclic voltammetry experiments reveal weakly interacting "Ir(ppy)(2)" units bridged by ethylene-linked bpe ligand in 3; on the contrary the metal centers are electronically isolated in 1 and 2 where the bridging ligands are based on ethane and propane linkers. The dimer 4 exhibits two accessible reversible reduction couples separated by 570 mV indicating the stability of the one-electron reduced species located on the diimine-based bridge abp. The "Ir(ppy)(2)" units in compound 5 are noninteracting as the electronic conduit is truncated by the ethane spacer in the bpfd bridge. The dinuclear compounds 1-5 show ligand centered (LC) transitions involving ppy ligands and mixed metal to ligand/ligand to ligand charge transfer (MLCT/LLCT) transitions involving both the cyclometalating ppy and bridging ligands (BL) in the UV-vis spectra. For the conjugated bridge bpe in compound 3 and abp in compound 4, the lowest-energy charge-transfer absorptions are red-shifted with enhanced intensity. In accordance with their similar electronic structures, compounds 1 and 2 exhibit identical emissions. The presence of vibronic structures in these compounds indicates a predominantly (3)LC excited states. On the contrary, broad and unstructured phosphorescence bands in compounds 3-5 strongly suggest emissive states of mixed (3)MLCT/(3)LLCT character. Density functional theory (DFT) calculations have been carried out to gain insight on the frontier orbitals, and to rationalize the electrochemical and photophysical properties of the compounds based on their electronic structures. 相似文献
