We investigate the effect of specific conformations of double-bond segments in highly polyunsaturated acyl chains on the deuterium (2)H NMR order parameters of a fully hydrated 1-stearoyl-2-docosahexaenoyl-sn-glycero-3-phosphocholine (SDPC, 18:0/22:6 PC) lipid bilayer. The system is analyzed by performing a molecular dynamics simulation study at ambient conditions in the fluid lamellar phase. By separately calculating the different partial contributions to the total order parameter profiles measurable experimentally, we are able to get insights into the molecular origin of earlier experimental and theoretical observations. The effect of the position of the different conformations of double-bond segments along the polyunsaturated acyl chain is also examined. As in experiments performed in a series of lipid bilayers with an increasing number of cis double bonds per lipid molecule [Holte, L. L., et al. Biophys. J. 1995, 68, 2396], we find that unsaturations influence mainly the order of the bottom half of the saturated chain. Specific conformations of the polyunsaturated chain close to the lipid headgroups have a distinct effect on the order of the bottom half of the saturated chain and on the top half of the polyunsaturated chain. Our results indicate that for SDPC the conformation of the region of the polyunsaturated chain located between the first three cis double bonds is responsible for the major effects on the orientational order of both the saturated and the polyunsaturated chains. 相似文献

The aim of this study was to analyze the use of 12 single-nucleotide polymorphisms in genes ELAC2, RNASEL and MSR1 as biomarkers for prostate cancer (PCa) detection and progression, as well as perform a genetic classification of high-risk patients. A cohort of 451 men (235 patients and 216 controls) was studied. We calculated means of regression analysis using clinical values (stage, prostate-specific antigen, Gleason score and progression) in patients and controls at the basal stage and after a follow-up of 72 months. Significantly different allele frequencies between patients and controls were observed for rs1904577 and rs918 (MSR1 gene) and for rs17552022 and rs5030739 (ELAC2). We found evidence of increased risk for PCa in rs486907 and rs2127565 in variants AA and CC, respectively. In addition, rs627928 (TT–GT), rs486907 (AG) and rs3747531 (CG–CC) were associated with low tumor aggressiveness. Some had a weak linkage, such as rs1904577 and rs2127565, rs4792311 and rs17552022, and rs1904577 and rs918. Our study provides the proof-of-principle that some of the genetic variants (such as rs486907, rs627928 and rs2127565) in genes RNASEL, MSR1 and ELAC2 can be used as predictors of aggressiveness and progression of PCa. In the future, clinical use of these biomarkers, in combination with current ones, could potentially reduce the rate of unnecessary biopsies and specific treatments. 相似文献

The mechanical relaxation spectrum of poly(monocyclohexylmethylene itaconate) (PMCMI) exhibits two well-developed absorptions in the glassy state that in increasing temperature order are named γ and β absorptions. Owing to the restricted conformational versatility of the backbone, the polymer presents a weak glass-rubber relaxation whose intensity is significantly lower than that of the γ absorption. Comparison of the mechanical spectrum of this polymer with that of poly(dicyclohexylmethylene itaconate) (PDCMI) allows the conclusion that the β relaxation is produced by motions in which the ? COOCH2C6H11 side groups are involved. The location of the mechanical γ peak suggests that this absorption is produced by flipping conformational transitions in the cyclohexyl residue. Three dielectric absorptions are observed in the glassy state of PMCMI which in increasing temperature order are called δ, γ, and β relaxations. Both the location and the activation energy of the dielectric and mechanical β absorptions suggest that both relaxations are caused by the same molecular motions. The influence of the chain expansion produced by excluded volume on the mean-square optical anisotropy has been studied in six types of polymers. The mean-square optical anistropy for a specified configuration is calculated using the valence optical scheme. Realistic rotational isomeric state models are used for the configurational statistics of the unperturbed chains. Excluded volume is introduced by hard sphere interactions. Results obtained with chains of 100, 200, 300, and 400 bonds permit extrapolation to the behavior expected for much longer chains. The mean-square optical anisotropy of polyethylene is insensitive to excluded volume. A similar conclusion was obtained several years ago in a study of chains confined to a tetrahedral lattice and weighted in a manner appropriate for the short-range interactions in polyethylene.2 Different behavior is seen in poly(vinyl chloride), poly(vinyl bromide), polystyrene, poly(p-chlorostyrene), and poly(p-bromostyrene). The mean-square optical anisotropy of these five vinyl polymers is sensitive to the imposition of excluded volume if the stereochemical composition is exclusively racemic. The spreading of Si-Ca-Al-Ti-O glasses on molybdenum has been investigated. By controlling the oxygen activity in the furnace, spreading can take place under reactive or nonreactive conditions. As the nucleation of the reaction product under reactive conditions is slow in comparison to the spreading kinetics, in both cases the glass front moves on the metal surface with similar spreading velocities. Spreading can be described using a molecular dynamics model where the main contribution to the wetting activation energy comes from the viscous interactions in the liquid. Enhanced interfacial diffusions in low-oxygen activities (reactive cases) form triple-line ridges that can pin the wetting front and cause a stick-slip motion. 相似文献
