Direct drug metabolism monitoring in a live single hepatic cell by video mass spectrometry

The metabolism of anti-breast cancer drug, tamoxifen, in a single human hepatocellular carcinoma cell, HepG2, was directly monitored by a video-mass spectroscope. The cytoplasm, a vacuole or nucleus of the cell was directly sucked by a nano-spray tip under a video-microscope, and then was introduced into a mass spectrometer. Unchanged drug molecules were found in cytoplasm and a vacuole, but the metabolites were only found in the cytoplasm. This direct detection of drug metabolites in a live single cell is useful for speedy drug metabolism monitoring.     

Fusion of the eight vertex SOS model

A higher spin analogue is presented of the eight vertex-SOS correspondence in the sense of Andrews-Baxter-Forrester. The resulting hierarchy of solvable models contain the hard hexagon model and its recent multi-state generalizations.     

The Landau electron problem on a cylinder

We consider the quantum mechanics of an electron confined to move on an infinite cylinder in the presence of a uniform radial magnetic field. This problem is in certain ways very similar to the corresponding problem on the infinite plane. Unlike the plane however the group of symmetries of the magnetic field, namely, rotations about the axis and the axial translations, is not realized by the quantum electron but only a subgroup comprising rotations and discrete translations along the axial direction, is. The basic step size of discrete translations is such that the flux through the ‘unit cylinder cell’ is quantized in units of the flux quantum. The result is derived in two different ways: using the condition of projective realization of symmetry groups and using the more familiar approach of determining the symmetries of a given Hamiltonian.     

HPTLC Determination of Cefpodoxime Proxetil in Formulations

An HPTLC method has been developed that coelutes both the isomers of cefpodoxime proxetil (CFP). CFP was chromatographed on a silica gel 60 F₂₅₄ TLC plate using toluene:acetonitrile (6:4) as a mobile phase and was quantified at 234 nm. The method was validated with respect to linearity, accuracy, precision and specificity. The limit of detection and limit of quantification for CFP were found to be 0.150 and 0.4 μg spot⁻¹, respectively. The proposed method was successfully used to determine the amount of CFP present in the marketed tablets and self-nanoemulsifying systems.

     

Coumarinolignans with Reactive Oxygen Species (ROS) and NF-κB Inhibitory Activities from the Roots of Waltheria indica

Seven new coumarinolignans, walthindicins A–F (1a, 1b, 2–5, 7), along with five known analogs (6, 8–11), were isolated from the roots of Waltheria indica. The structures of the new compounds are determined by detailed nuclear magnetic resonance (NMR), circular dichroism (CD) with extensive computational support, and mass spectroscopic data interpretation. Compounds were tested for their antioxidant activity in Human Cervical Cancer cells (HeLa cells). Compounds 1a and 6 showed higher reactive oxygen species (ROS) inhibitory activity at 20 μg/mL when compared with other natural compound-based antioxidants such as ascorbic acid. Considering the role of ROS in nuclear-factor kappa B (NF-κB) activation, compounds 1a and 6 were evaluated for NF-κB inhibitory activity and showed a concentration-dependent inhibition in Human Embryonic Kidney 293 cells (Luc-HEK-293).     

A facile conversion of ethoxydihydropyrans to 4-cyanoethylisoxazoles

The reaction of ethoxydihydropyrans 2 , which were prepared from 2-methylene-1,3-dicarbonyl compounds 1 with ethyl vinyl ether, and hydroxylamine hydrochloride gave 4-cyanoethylisoxazoles 3 whose substituents at the 3 and 5 positions were transformed from substituent at the 2 position and the acyl group of 2 regioselectively.     

Generation and Michael Reaction of 1, 3-Bis(trimethylsiloxy)-isoindoles by Reduction of N-Alkylphthalimides with a Zn-Me3SiCl-Lutidine System

Formation and concomitant Michael reaction of 1,3-bis(trimethylsiloxy)isoindoles have been achieved by the reduction of phthalimides with a Zn-Me₃SiCl-lutidine system in the presence of α, β-unsaturated carbonyl compounds under sonication.     

A new moment matching algorithm for sampling from partially specified symmetric distributions

A new algorithm is proposed for generating scenarios from a partially specified symmetric multivariate distribution. The algorithm generates samples which match the first two moments exactly, and match the marginal fourth moments approximately, using a semidefinite programming procedure. The performance of the algorithm is illustrated by a numerical example.
