The myotendon junction (MTJ) is a specialised area into the muscle fibers where the sarcoplasmic membranes connect to the collagen fibers bundles. There are few data about plasticity of the MTJ in aging processes. The aim of this study is to analyse the ultrastructure characteristics of MTJ of medial pterygoid muscle of adult and aged rats. Employing the transmission electron microscopy method, twenty male rats Wistar (Rattus norvegicus) were divided into two groups: A (n = 10) with 12 months of age; B (n = 10) 24 months of age. The animals were anaesthetised with overdose the urethane (3 g/kg, i.p.) and sacrified during the perfusion with modified Karnovsky solution. The specimens were post-fixed in a 1% osmium tetroxide solution, dehydrated in ascending concentration of ethanol and embedded in Spurr resin. The thin sections, of 90 nm thick, were counterstained with uranyl acetate and lead citrate solution, and examined in a Jeol 1010 transmission electron microscope. The fine structure of the MTJ of group A revealed the defined interdigitations and disposed in several levels of deep formations containing the collagen fibers. In the group B, such structures did not observed, detecting the projections irregular in shape, and large of extra matrix with in aspect of remodelling. In conclusion it was possible to identify the plasticity of MTJ in the group B which presented several morphological alterations comparing to the adult animals. These data of group B suggested the occurrence of aging processes in the MTJ in rats.  相似文献

Synthesis of a 6/6/6 tricyclic ether system (3) corresponding to the ABC ring fragment of yessotoxin (1) has been achieved via coupling of a triflate and a 2-lithiofuran followed by intramolecular hetero-Michael addition. The IJ ring fragment (4) of 1 was readily synthesized via successive Sharpless epoxidation and 6-endo cyclization of the resulting vinyl epoxide.  相似文献

We demonstrate a novel technique for molecular imprinting and immobilization on a surface of a polymer containing azo dyes (azopolymer). The azopolymer was found to be capable of immobilizing micrometer- and nanometer-scale macromolecules (e.g., lambda-DNA, immunoglobulin G (IgG), bacterial protease, and 1-mum polystyrene particles) through photoirradiation with blue-wavelength light. Fluorescence and atomic force microscopy studies revealed that the azopolymer surface deformed along with the shape of the macromolecules, holding them in place after photoirradiation. The desorption of the immobilized macromolecules from the azopolymer surface in an aqueous medium was observed to be very slow, on the time scale of 10 min to weeks, depending on the photoirradiation time. Immunological and enzymatic studies showed that IgG and bacterial protease immobilized on the azopolymer surface retained their original functionality. These results suggest that the azopolymer physically, not chemically, binds the macromolecules because of the increase in contact area between the macromolecules and the azopolymer surface after photoirradiation.  相似文献

The polarized total-reflection X-ray absorption fine structure method was applied to characterize zinc porphyrins at the air-water interface. The X-ray absorption near edge structure exhibited a significant difference depending on the polarization of the X-ray. A shoulder peak of the Zn K-edge corresponding to the 1s-4p(z) transition for a square planar metal complex without axial coordination(s) was observed at 9662 eV, which indicates that the axial coordination sites of zinc porphyrin molecules examined are not fully hydrated at the air-water interface. The molecular orientation of zinc porphyrins was determined by analyzing the polarization dependence of the transition peak intensity. The meso-substituted porphyrin derivative 5,10,15,20-tetraphenylporphyrinatozinc(II) (ZnTPP) orients rather parallel to the solution surface. In contrast to ZnTPP, the zinc(II) protoporphyrin IX (ZnPP) with hydrophilic carboxyl groups at one side of the molecule stands up with respect to the solution surface, and the average tilting angle of the porphyrin plane to the surface was evaluated to be between 57 degrees and 43 degrees. In addition, the axial coordination of ZnPP is modified depending on the surface concentration, in which the axial hydration to the zinc center is effectively inhibited in the compressed surface layer.  相似文献

A carbonyl ylide cycloaddition approach to the squalene synthase inhibitors zaragozic acids A and C is described. The carbonyl ylide precursor 8 was synthesized starting from di-tert-butyl D-tartrate (47) via an eleven-step sequence involving the regioselective reduction of the mono-MPM (MPM=4-methoxybenzyl) ether 48 with LiBH4 and the diastereoselective addition of sodium tert-butyl diazoacetate to alpha-keto ester 10. The reaction of alpha-diazo ester 8 with 3-butyn-2-one (40) in the presence of a catalytic amount of [Rh2(OAc)4] gave the desired cycloadduct 59 as a single diastereomer. The dihydroxylation of enone 59 followed by sequential transformations permitted the construction of the fully functionalized 2,8-dioxabicyclo[3.2.1]octane core 5. Alkene 79 derived from 5 serves as a common precursor to zaragozic acids A (1) and C (2), since the elongation of the C1 alkyl side chain can be attained by olefin cross-metathesis, especially under the influence of Blechert's catalyst (85).  相似文献

We have developed the NBS (2-nitrobenzenesulfenyl) method, a quantitative proteome analysis method utilizing stable isotope labeling followed by mass spectrometry. The potential of this method was reported previously, and the procedure has now been further optimized. Here, we describe a procedure utilizing urea or guanidine hydrochloride as a protein denaturant, in conjunction with an improved chromatographic enrichment method for the NBS-labeled peptides using a phenyl resin column. By using this new protocol, both sample loss throughout the protocol and the elution of unwanted unlabeled peptides can be minimized, improving the efficiency of the analysis significantly.  相似文献

The labeling of 5 major allergenic ingredients (egg, milk, wheat, buckwheat, and peanut) is mandatory in Japan, and 2 series of enzyme-linked immunosorbent assay (ELISA) kits have been established as official screening methods. However, these official methods have not provided the necessary sensitivity, due in part to poor extraction efficiency. To address this need, 2 novel ELISA kits have been developed: the FASTKIT ELISA Ver. II Series and the FASPEK Allergenic Substances Detection Kit. The new kit systems use an improved extraction buffer that can extract insoluble proteins produced by processing and feature new antibodies that bind to the denatured proteins extracted with the new extraction buffer. The analytical performances of the 2 new ELISA kit series were evaluated in an interlaboratory study. Ten laboratories participated in the study and determined the major allergenic ingredients contained in 5 types of model processed food. The 2 ELISAs displayed fairly good reproducibility and sufficient recovery.  相似文献

The natural product lovastatin and its semisynthetic, more effective derivative, simvastatin, are important drugs for the treatment of hypercholesterolemia. Here, we report the biochemical characterization of a dedicated acyltransferase, LovD, encoded in the lovastatin biosynthetic pathway. We demonstrate that LovD has broad substrate specificity towards the acyl carrier, the acyl substrate, and the decalin acyl acceptor. LovD can efficiently catalyze the acyl transfer from coenzyme A thioesters or N-acetylcysteamine (SNAC) thioesters to monacolin J. When alpha-dimethylbutyryl-SNAC was used as the acyl donor, LovD was able to convert monacolin J and 6-hydroxyl-6-desmethylmonacolin J into simvastatin and huvastatin, respectively. Using the Escherichia coli LovD overexpression strain as a whole-cell biocatalyst, preparative amounts of simvastatin were synthesized in a single fermentation step. Our results demonstrate LovD is an attractive enzyme for engineered biosynthesis of pharmaceutically important cholesterol-lowering drugs.  相似文献

The quenching rate constants of the excited triplet state of Rose Bengal (RB) by oxygen (k(obs)) were measured in ethylene glycol (EG) at different temperatures using nanosecond laser flash photolysis. Although a plot of the quenching rate constant k(obs) for RB triplet state vs oxygen concentration is linear at 20 degrees C, the oxygen dependence of k(obs) does not exhibit linearity but upward curvature at high temperatures from 130 to 140 degrees C. The upward curvature at high temperatures is not well-described by a kinetic scheme first postulated by Gijzeman et al., which is characterized by exciplex formation and a unimolecular dissociation of the exciplex to products, but instead by a more comprehensive mechanism involving a bimolecular dissociation in addition to a unimolecular one. The measurements of the oxygen dependence of k(obs) for RB triplet state at different temperatures yielded a reaction enthalpy for the exciplex formation of 150 kJ mol(-1). Due to the large exothermic reaction enthalpy, equilibrium was obtained for the exciplex at 20 degrees C even at low oxygen concentration and the bimolecular quenching by oxygen became the major dissociation process. The equilibrium attainment and bimolecular dissociation provide a linear oxygen dependence of k(obs) to all outward appearances. Therefore, linearity does not always mean that exciplex dissociation proceeds solely through a unimolecular mechanism.  相似文献
