The self-assembly in aqueous solution of a PEG-peptide conjugate is studied by spectroscopy, electron microscopy, rheology and small-angle X-ray and neutron scattering (SAXS and SANS). The peptide fragment, FFKLVFF is based on fragment KLVFF of the amyloid beta-peptide, Abeta(16-20), extended by two hydrophobic phenylalanine units. This is conjugated to PEG which confers water solubility and leads to distinct self-assembled structures. Small-angle scattering reveals the formation of cylindrical fibrils comprising a peptide core and PEG corona. This constrained structure leads to a model parallel beta-sheet self-assembled structure with a radial arrangement of beta sheets. On increasing concentration, successively nematic and hexagonal columnar phases are formed. The flow-induced alignment of both structures was studied in situ by SANS using a Couette cell. Shear-induced alignment is responsible for the shear thinning behaviour observed by dynamic shear rheometry. Incomplete recovery of moduli after cessation of shear is consistent with the observation from SANS of retained orientation in the sample.  相似文献

Directly obtaining kinetic and mechanistic data for the reactions of nitroxyl (HNO) with biomolecules (k≈103–107 m ?1 s?1) is not feasible for many systems because of slow HNO release from HNO donor molecules (t1/2 is typically minutes to hours). To address this limitation, we have developed a photoactivatable HNO donor incorporating the (3‐hydroxy‐2‐naphthalenyl)methyl phototrigger, which rapidly releases HNO on demand. A “proof of concept” study is reported, which demonstrates that, upon continuous xenon light excitation, rapid decomposition of the HNO donor occurs within seconds. The amount of HNO generated is strongly dependent on solvent and the rate of the reaction is dependent on the light intensity.  相似文献

Photodynamic therapy (PDT), the combined action of a photosensitizer and light to produce a cytotoxic effect, is an approved therapy for a number of diseases. At present, clinical PDT treatments involve one-photon excitation of the photosensitizer. A major limitation is that damage may be caused to healthy tissues that have absorbed the drug and lie in the beam path. Two-photon excitation may minimize this collateral damage, as the probability of absorption increases with the square of the light intensity, enabling spatial confinement of the photosensitizer activation. A potential application is the treatment of the wet-form of age-related macular degeneration, the foremost cause of central vision loss in the elderly. Herein, the commercial photosensitizers Visudyne and Photofrin are used to demonstrate quantitative in vitro two-photon PDT. A uniform layer of endothelial cells (YPEN-1) was irradiated with a Ti:sapphire laser (300 fs, 865 nm, 90 MHz) using a confocal scanning microscope. Quantification of the two-photon PDT effect was achieved using the permeability stain Hoechst 33258 and a SYTOX Orange viability stain. Visudyne was found to be around seven times more effective as a two-photon photosensitizer than Photofrin under the conditions used, consistent with its higher two-photon absorption cross-section. We also demonstrate for the first time the quadratic intensity dependence of cellular two-photon PDT. This simple in vitro method for quantifying the efficacy of photosensitizers for two-photon excited PDT will be valuable to test specifically designed two-photon photosensitizers before proceeding to in vivo studies in preclinical animal models.  相似文献

The aim of the paper is to show how the method of homogenization can be applied in modelling of soft tissue undergoing large deformation. Simplified microstructures are considered, which consist of hyperelastic porous matrix and periodic array of fluid‐filled cells. At the microscopic level diffusion processes are described by the Darcy law, permeability of the cellular membrane is introduced. Although at the macroscopic scale the tissue is incompressible, the flow inside microscopic volumes induces viscous relaxation effects. The homogenized problem is formulated.  相似文献

Synthetic dyes have become an integral part of many industries such as textiles, tannin and even food and pharmaceuticals. Industrial dye effluents from various dye utilizing industries are considered harmful to the environment and human health due to their intense color, toxicity and carcinogenic nature. To mitigate environmental and public health related issues, different techniques of dye remediation have been widely investigated. However, efficient and cost-effective methods of dye removal have not been fully established yet. This paper highlights and presents a review of recent literature on the utilization of the most widely available biopolymers, specifically, cellulose, chitin and chitosan-based products for dye removal. The focus has been limited to the three most widely explored technologies: adsorption, advanced oxidation processes and membrane filtration. Due to their high efficiency in dye removal coupled with environmental benignity, scalability, low cost and non-toxicity, biopolymer-based dye removal technologies have the potential to become sustainable alternatives for the remediation of industrial dye effluents as well as contaminated water bodies.  相似文献

Homogenization provides a reduced model of the acoustic wave propagation through a periodically perforated layer immersed in an inviscid fluid. The shape of the perforation influences some parameters of the non-local transmission condition. As discrete particles and/or as surface coatings on other minerals in natural systems, aluminum hydroxides are efficient sinks for Hg(II). The Hg(II) adsorption on gibbsite was determined as a function of temperature (T), pH, and the type of background electrolytes, i.e., NaNO(3), NaClO(4), and NaCl. When the equilibration time t(E) approximately 2 h, the Hg(II) retention on gibbsite was found to be a reversible process, which was ascribed to adsorption. The Hg(II) adsorption capacity, i.e., Gamma(Hg(II)), varied with the type of electrolyte used in accordance with the following order: Gamma(NO(3))(Hg(II)) > or = Gamma(ClO(4))(Hg(II)) > or = Gamma(Cl)(Hg(II)). In all cases, the estimated thermodynamic parameters showed that the Hg(II) adsorption on gibbsite was endothermic and spontaneous. The Hg(II) adsorption data were quantified with the Langmuir or Hill, and Dublin-Radushkevick (DR), isotherms at all temperatures and acidity levels examined. Always, the Hg(II) adsorption data were in compliance with the DR model. However, the Hg(II) adsorption in NaNO(3) or NaClO(4) was interpreted in terms of the Langmuir model. When NaCl was used as electrolyte, the Hg(II) adsorption was modeled well with the Hill equation. The mean free energy values calculated from DR plots concluded that Hg(II)-gibbsite interactions are a result of chemical bonding.  相似文献

Rapid maturational brain changes occur during adolescence--a time associated with risk-taking behaviors and improvements in cognition. The present study examined the relationship between white matter (WM) microstructure, impulsive behavior and response inhibition in female and male adolescents. Twenty-one healthy adolescents underwent diffusion tensor imaging using a 3.0-T magnetic resonance imaging system. Impulse control was assessed using the Bar-On Emotional Quotient Inventory, Youth Version. Response inhibition was assessed using the Stroop Color-Word Interference Test. Fractional anisotropy (FA), a measure of WM coherence, and trace, a measure of overall diffusivity, were determined from voxels manually placed in the midline and in the left and right forward-projecting arms of the genu and the splenium of the corpus callosum. Sex-specific differences were observed for the relationship between FA and impulsive behavior in the right anterior callosum for males and in the splenium for females. Males, compared to females, displayed significantly higher FA in the left WM region. Although trace was not associated with impulse control, trace in the genu (for females) and splenium (males and females) was associated with Stroop performance. Regional differences in trace also were evident, with lower values in the splenium observed than in all other regions. Although the latter significantly improved with age, no sex differences in impulse control or in Stroop performance were detected. The present findings provide supporting evidence for sex-related differences in the development of WM microstructure during adolescence. These data further suggest a neurobiological mechanism underlying some of the emotional and cognitive changes commonly observed in males versus females during the adolescent period.  相似文献

The aim of our work is to develop optimal dielectric composite structures with specific qualities. The task is to design interfaces of given material components such that the originated structure attains certain optical properties. Propagation of the electromagnetic waves in the composite is described by the Helmholtz equation. Success of the structure is enumerated by the objective function which is to be minimized. Interfaces of the given materials are parametrized by the cubic B–spline curves. The design variables are afterwards the positions of B–spline control points. For objective function evaluation one forward computation of the Helmholtz equation is needed. To get the sensitivity of the objective function we solve the backward (adjoint) equation.
