Targeted Photodynamic Therapy of Human Head and Neck Squamous Cell Carcinoma with Anti-epidermal Growth Factor Receptor Antibody Cetuximab and Photosensitizer IR700DX in the Mouse Skin-fold Window Chamber Model

Targeted photodynamic therapy (PDT) in head/neck cancer patients with a conjugate of the anti-epidermal growth factor receptor (EGFR) antibody, Cetuximab and a phthalocyanine photosensitizer IR700DX is under way, but the exact mechanisms of action are still not fully understood. In this study, the EGFR-overexpressing human head/neck OSC-19-luc2-cGFP tumor with transfected GFP gene was used in a skin-fold window chamber model in BALB/c nude mice. The uptake and localization of the conjugate in the tumor and its surrounding normal tissues were studied by an intravital confocal laser scanning microscopy with image analyses. The tumor was also irradiated with 690 nm laser light 24 h after conjugate administration. The vascular and tumor responses were examined by morphological evaluation and immunohistochemistry (IHC). The amount of conjugate in the tumor peaked at 24–48 h after injection. Image analyses of colocalization correlation parameters demonstrated a high fraction of the conjugate IR700DX colocalized in the GFP-expressing tumor cells. PDT-treated tumors showed extensive necrotic/apoptotic destruction with little vascular damage, while IHC showed no HIF-1α expression and decreased EGFR and Ki67 expression with activated caspase-3 overexpression, indicating a direct killing of tumor cells through both necrotic and apoptotic cell death.     

Late-Stage Functionalization by Chan–Lam Amination: Rapid Access to Potent and Selective Integrin Inhibitors

A late-stage functionalization of the aromatic ring in amino acid derivatives is described. The key step is a copper-catalysed diversification of a boronate ester by amination (Chan–Lam reaction) that can be carried out on a complex β-aryl-β-amino acid scaffold. This not only considerably extends the substrate scope of amination partners, but also delivers an array of potent and selective integrin inhibitors as potential treatment agents of idiopathic pulmonary fibrosis (IPF). This versatile chemical strategy, which is amenable to high-throughput-array protocols, allows the installation of pharmaceutically valuable heteroaromatic fragments at a late stage by direct coupling to NH heterocycles, leading to compounds with drug-like attributes. It thus constitutes a useful addition to the medicinal chemist's repertoire.     

Laser inhibited diffusion in rhodamine-ethanol solutions

The diffusion of rhodamine-6G dye in ethanol is observed to be inhibited by optical pumping by a cadmium laser. The diffusion process is observed as a function of the solution temperature. The relative difference in diffusion coefficients with and without optical pumping is calculated. The effect is interpreted as being due to a stronger solvent-dye interaction in the first excited singlet state of rhodamine-6G.

Characteristics of Add Drop Filter using single and dual PCRR in square lattice

A Photonic Crystal Ring Resonator (PCRR) based Add Drop Filter is designed using two dimensional (2D) square lattice PC silicon rods in air host. The normalized transmission spectra for a single-ring and dual-ring configurations have been investigated using 2D Finite Difference Time Domain (FDTD). The filter characteristics such as coupling efficiency, dropping efficiency, resonant wavelength and Q factor are numerically analyzed for single ring and dual ring PCRR. It is noted that the coupling efficiency, dropping efficiency and Q factor of single ring circular PCRR with scatterer rod are 100%, 100% and 186.75, respectively. The backward dropping is observed in single ring PCRR whereas forward dropping are noticed in dual ring PCRR based ADF. The Photonic Band Gap (PBG) with respect to structural parameters such as the radius of the rod, lattice constant and refractive index difference, and propagation modes in periodic and nonperiodic structure are calculated by Plane Wave Expansion (PWE) method.     

Counting bridgeless graphs

Methods are developed for finding the number of unlabeled bridgeless or 2-line-connected graphs of any order. These methods are based on cycle index sums, but it is shown how to avoid explicit compution with cycle index sums by using suitable inversion techniques. Similar results are obtained for unlabeled bridgeless graphs by numbers of points and lines, and connected graphs by numbers of points and bridges. Corresponding results for labeled graphs are found as corollaries. When lines or bridges are required as enumeration parameters in the labeled case it is also shown how to obtain improved recurrence relations. The latter appear to have no analog for unlabeled graphs.     

Modeling Ergodic, Measure Preserving Actionson Shifts of Finite Type

 In this paper we show that shifts of finite type satisfying a strong topological mixing property are universal models for ergodic measure preserving dynamical systems.
