A small, semi-preparative C(4) RP-HPLC column was used to set up the conclusive laboratory-scale purification of Chinese hamster ovary-derived human thyrotropin (hTSH), after a preliminary concentration-purification of an extremely dilute and poorly ( approximately 0.6 microg hTSH/mL; mass fraction=0.35%) conditioned medium on a cation exchanger. Several fractions of this eluate were repeatedly injected on the semi-preparative column, obtaining, in a single run (<1h chromatographic time), a concentrated pool ( approximately 1.2 mg/mL) of highly purified hTSH that could be further concentrated to >3 mg/mL and then efficiently lyophilized. The overall recovery in the rapid RP-HPLC purification step, including concentration and lyophilization, was of the order of 80%. The final product, when tested via a precise, single-dose in vivo bioassay, confirmed that it did not suffer any loss of bioactivity. This same methodology can be easily adapted to the small-scale purification of other recombinant products, even when obtained from genetically modified organisms at extremely low concentrations and mass fractions. 相似文献

Markowitz formulated the portfolio optimization problem through two criteria: the expected return and the risk, as a measure of the variability of the return. The classical Markowitz model uses the variance as the risk measure and is a quadratic programming problem. Many attempts have been made to linearize the portfolio optimization problem. Several different risk measures have been proposed which are computationally attractive as (for discrete random variables) they give rise to linear programming (LP) problems. About twenty years ago, the mean absolute deviation (MAD) model drew a lot of attention resulting in much research and speeding up development of other LP models. Further, the LP models based on the conditional value at risk (CVaR) have a great impact on new developments in portfolio optimization during the first decade of the 21st century. The LP solvability may become relevant for real-life decisions when portfolios have to meet side constraints and take into account transaction costs or when large size instances have to be solved. In this paper we review the variety of LP solvable portfolio optimization models presented in the literature, the real features that have been modeled and the solution approaches to the resulting models, in most of the cases mixed integer linear programming (MILP) models. The paper concerns the density points with respect to the sequences of intervals tending to zero in the family of Lebesgue measurable sets. It shows that for some sequences analogue of the Lebesgue density theorem holds. Simultaneously, this paper presents proof of theorem that for any sequence of intervals tending to zero a relevant operator ?J generates a topology. It is almost but not exactly the same result as in the category aspect presented in [WIERTELAK, R.: A generalization of density topology with respect to category, Real Anal. Crotalus durissus cascavella is a snake native of northeastern Brazil. The aim of the study was to investigate the effects of C. d. cascavella venom on rat mean arterial pressure and vascular reactivity in the mesenteric vascular bed. The venom evoked a dose-dependent decrease in mean arterial pressure, cardiac and respiratory frequency with increased plasma nitrite levels. L-NAME (10 mg/kg) blunted both the hypotension and increased nitrite production observed after the venom administration. To investigate the effects of C. d. cascavella in resistance vessels, the vascular mesenteric bed was studied, and the results suggested that the hypotensive effect of the venom is not dependent on a direct vasodilatory activity. In conclusion, C. d. cascavella venom presented indirect hypotensive effects with the involvement of nitric oxide. 相似文献
Block copolymer micelles with bactericidal properties were designed to deactivate pathogens such as E. coli bacteria. The micelles of PS‐b‐PAA and PS‐b‐P4VP block copolymers were loaded with biocides TCMTB or TCN up to 20 or 30 wt.‐%, depending on the type of antibacterial agent. Bacteria were exposed to loaded micelles and bacterial deactivation was evaluated. The micelles loaded with TCN are bactericidal; bacteria are killed in less than two minutes of exposure. The most likely interpretation of the data is that the biocide is transferred to the bacteria by repeated micelle/bacteria contacts, and not via the solution.
The objective of this research was to develop, optimize, and validate a modern, rapid method of preparation of human hair
samples, using microwave irradiation, for analysis of eight tricyclic antidepressants (TCADs): nordoxepin, nortriptyline,
imipramine, amitriptyline, doxepin, desipramine, clomipramine, and norclomipramine. It was based on simultaneous alkaline
hair microwave-assisted hydrolysis and microwave-assisted extraction (MAH–MAE). Extracts were analyzed by high-performance
liquid chromatography with diode-array detection (HPLC–DAD). A mixture of n-hexane and isoamyl alcohol (99:1, v/v) was used as extraction solvent and the process was performed at 60°C. Application of 1.0 mol L−1 NaOH and microwave irradiation for 40 min were found to be optimum for hair samples. Limits of detection ranged from 0.3
to 1.2 μg g−1 and LOQ from 0.9 to 4.0 μg g−1 for the different drugs. This enabled us to quantify them in hair samples within average therapeutic concentration ranges. 相似文献
This paper presents an optimization model for downsizing a multi-product supply chain facing bankruptcy risk, where multi-functional production facilities are shared for producing a group of substitutable products. In order to determine the potential demand after discontinuation of certain product types, the proposed mixed integer programming model incorporates a new general formulation of demand substitution, which allows arbitrary demand diversion and arbitrary replacement rates between products under investigation. The new substitution formulation enables considering uneven substitutions for supply chain network design and therefore it extends currently used substitution approaches. A series of systematically generated downsizing cases confirm the validity of the here proposed approach, and illustrate how downsized chains can successfully adopt to the new circumstances. 相似文献
Determinations of Cu, Fe, Mn and Zn were performed in an oxisol soil treated with effluent originated from a biodigester septic cesspool. The extracts were obtained from a DTPA/TEA (pH 7.3) solution and analysed by electroanalytical methodologies using square wave anodic stripping voltammetry (SWASV) for the analysis of Cu, Mn and Zn and square wave voltammetry (SWV) for Fe analysis, both of them with a thin mercury film microelectrode (ME-Hg). DTPA (diethylenetriaminepentaacetic acid) complexes with Cu, Mn and Fe show electroactivity in the potentials range close to the ones for the free ions in solution, but the Zn complex did not show any electroactivity at the potential range analysed. SWASV/ME-Hg and SWV/ME-Hg results were in good agreement (r2 = 0.996) with the flame atomic absorption spectroscopy (FAAS) analyses, demonstrating that electroanalytical methodologies can be used for micronutrient determinations in soil extracts without serious interferences of the matrix components. Some characteristics of the complex formed between DTPA-metals are explored by EPR experiments. 相似文献
